Prior data on long-term association between habitual legume consumption and hypertension risk remained sparse. We aimed to evaluate whether total legume and subtype intakes were prospectively related to lower hypertension incidence among 8,758 participants (aged ≥30 years) from the China Health and Nutrition Survey 2004-2011. Dietary intakes were assessed by interviews combining 3-day 24-h food recalls and household food inventory weighing method at each survey round (median dietary assessment times during follow-up was three). Incident hypertension was identified by self-reports or blood pressure measurements. Multivariable Cox regression model was applied to estimate hazard ratio (HR) for hypertension across increasing categories of cumulatively averaged legume intake. For 35,990 person-years (median 6.0 years per person), we documented 944 hypertension cases. After adjustment for covariates, higher consumption of total legumes was significantly associated with a lower hypertension risk, with HR comparing extreme categories being 0.56 (95% CI 0.43-0.71; P for trend <0.001). Then we found that intakes of dried legumes (HR 0.53; 95% CI 0.43-0.65; P for trend <0.001) and fresh legumes (HR 0.67; 95% CI 0.55-0.81; P for trend <0.001) were both related to reduced hypertension risks. However, further classification of dried legumes revealed that the inverse association with hypertension substantially held for higher soybean (HR 0.51; 95% CI 0.41-0.62; P for trend <0.001) but not non-soybean intakes. In stratified analyses, the association of interest remained similar within strata defined by gender, body mass index, physical activity, smoking and drinking status; however, significant heterogeneity of results was detected across age strata (P for interaction = 0.02). Total legume intake related to a more pronounced decrease in hypertension risk for the elderly (≥65 years [HR 0.47; 95% CI 0.30-0.73; P for trend <0.001]), in contrast to that for the non-elderly. Our findings suggest inverse associations of all kinds of legume but may not non-soybean intakes with risk of developing hypertension.
This study was conducted to evaluate the effects of glucose, soy oil, or glutamine on jejunal morphology, protein metabolism, and protein expression of the mammalian target of rapamycin complex 1 (mTORC1) signaling pathway in jejunal villus or crypt compartment of piglets. Forty-two 21 d-weaned piglets were randomly allotted to one of the three isocaloric diets formulated with glucose, soy oil, or glutamine for 28 days. On day 14 or 28, the proteins in crypt enterocytes were analyzed with isobaric tags for relative and absolute quantification, and proteins involved in mTORC1 signaling pathway in villus or crypt compartment cells were determined by western blotting. Our results showed no significant differences (P > 0.05) in jejunal morphology among the three treatments on day 14 or 28. The differentially expressed proteins mainly took part in a few network pathways, including antimicrobial or inflammatory response, cell death and survival, digestive system development and function, and carbohydrate metabolism. On day 14 or 28, there were higher protein expression of 4EBP1 in jejunal crypt compartment of piglets supplemented with glucose or glutamine compared with soy oil. On day 28, higher protein expression of p-mTOR in crypt compartment was observed in piglets supplemented with glucose compared with the soy oil. In conclusion, the isocaloric glucose, soy oil, or glutamine did not affect the jejunal morphology of piglets; however, they had different effects on the protein metabolism in crypt compartment. Compared to soy oil, glucose or glutamine may be better energy supplies for enterocytes in jejunal crypt compartment.
The present study was performed on antigen-presenting cells (APCs) of Theileria annulata transformed dendritic cells (TaDCs) and monocyte-derived dendritic cells (MoDCs) to compare differences in antigen presentation and stimulation of T lymphocyte proliferation. Antigen presentation for T lymphocyte proliferation was analysed by flow cytometry. Additionally, the level of mRNA transcription of small GTPases of the Rab family expressed in the TaDC cell line was analysed by quantitative real-time polymerase chain reaction (Q-RT-PCR). The endocytosis rate of TaDCs was significantly (P < 0.01) lower than in MoDCs. In contrast, when T lymphocytes were co-cultured with TaDC-APCs T cell proliferation was similar, while co-culture with MoDC-APC stimulated proliferation of CD4+ cells to a greater degree than CD8+ cells. However, the efficacy of TaDC-APCs to stimulate T lymphocytes dropped as the number of passages of TaDC-APC increased. Likewise, the transcription level of Rab family genes also significantly (P > 0.001) declined with progressive passages (>50) of the TaDC cell line. We conclude that initially the TaDC cell line efficiently presents antigen to stimulate T lymphocyte proliferation to produce a cellular immune response against the presented antigen.
Item 9 of the Patient Health Questionnaire-9 (PHQ-9) queries about thoughts of death and self-harm, but not suicidality. Although it is sometimes used to assess suicide risk, most positive responses are not associated with suicidality. The PHQ-8, which omits Item 9, is thus increasingly used in research. We assessed equivalency of total score correlations and the diagnostic accuracy to detect major depression of the PHQ-8 and PHQ-9.
We conducted an individual patient data meta-analysis. We fit bivariate random-effects models to assess diagnostic accuracy.
16 742 participants (2097 major depression cases) from 54 studies were included. The correlation between PHQ-8 and PHQ-9 scores was 0.996 (95% confidence interval 0.996 to 0.996). The standard cutoff score of 10 for the PHQ-9 maximized sensitivity + specificity for the PHQ-8 among studies that used a semi-structured diagnostic interview reference standard (N = 27). At cutoff 10, the PHQ-8 was less sensitive by 0.02 (−0.06 to 0.00) and more specific by 0.01 (0.00 to 0.01) among those studies (N = 27), with similar results for studies that used other types of interviews (N = 27). For all 54 primary studies combined, across all cutoffs, the PHQ-8 was less sensitive than the PHQ-9 by 0.00 to 0.05 (0.03 at cutoff 10), and specificity was within 0.01 for all cutoffs (0.00 to 0.01).
PHQ-8 and PHQ-9 total scores were similar. Sensitivity may be minimally reduced with the PHQ-8, but specificity is similar.
Giardia duodenalis is a common intestinal parasite in humans and other mammals, and it causes major public and veterinary health problems worldwide. China is a major pig-raising country, and studies on Giardia in pigs have important public health significance. The present study was conducted to investigate the prevalence of Giardia and assess its genetic characterization. A total of 93 samples were collected from two farms in Shanghai. The presence of Giardia was determined using PCR and sequence analysis of glutamate dehydrogenase, beta-giardin and triose phosphate isomerase genes. The average prevalence of G. duodenalis infection was 26.88% (25/93) in the pigs, with 28.13% (18/64) in farm 1 vs 24.14% (7/29) in farm 2. All the PCR-positive products were successfully sequenced, and assemblage E was more prevalent. Zoonotic assemblages A and B and canine-specific assemblage C were identified in farm 1, whereas, only assemblage E was detected in farm 2. Interestingly, two pig isolates showed 100% homology with human-derived isolates from Australia and China at the bg and tpi loci respectively. Pigs infected with Giardia infect humans by polluting the environment; whether pigs are a potential environmental source of the human pathogen in China requires more epidemiological data.
Paediatric Mycoplasma pneumoniae pneumonia (MPP) is a major cause of community-acquired pneumonia in China. Data on epidemiology of paediatric MPP from China are little known. This study retrospectively collected data from June 2006 to June 2016 in Beijing Children's Hospital, Capital Medical University of North China and aims to explore the epidemiological features of paediatric MPP and severe MPP (SMPP) in North China during the past 10 years. A total of 27 498 paediatric patients with pneumonia were enrolled. Among them, 37.5% of paediatric patients had MPP. In this area, an epidemic took place every 2–3 years at the peak, and the positive rate of MPP increased during these peak years over time. The peak age of MPP was between the ages of 6 and 10 years, accounting for 75.2%, significantly more compared with other age groups (χ2 = 1384.1, P < 0.0001). The epidemics peaked in September, October and November (χ2 = 904.9, P < 0.0001). Additionally, 13.0% of MPP paediatric patients were SMPP, but over time, the rate of SMPP increased, reaching 42.6% in 2016. The mean age of paediatric patients with SMPP (6.7 ± 3.0 years old) was younger than that of patients with non-SMPP (7.4 ± 3.2 years old) (t = 3.60, P = 0.0001). The prevalence of MPP and SMPP is common in China, especially in children from 6 to 10 years old. Paediatric patients with SMPP tend to be younger than those with non-SMPP. MPP outbreaks occur every 2–3 years in North China. September, October and November are the peak months, unlike in South China. Understanding the epidemiological characteristics of paediatric MPP can contribute to timely treatment and diagnosis, and may improve the prognosis of children with SMPP.
The antibacterial hydrogels can be widely used in the biomedical area owing to their excellent properties. The main limitation of antibacterial hydrogels is their poor mechanical strength. In this study, the novel hydrogels were fabricated with a mixture of silk fibroin (SF), chitosan (CH), agarose (AG), and silver nanoparticles (SNPs) via facile reaction condition without inorganic substances. The mechanical property of these fabricated hydrogels can be modulated by the concentration of SF or AG. The rheological studies demonstrated enhanced elasticity of CH-doped hydrogels. Because of the presence of CH and Ag in hydrogels, the antimicrobial property against gram-positive and gram-negative bacteria was exhibited. Cytocompatibility test proved the very low toxic nature of the hydrogels. In addition, these composite hydrogels have a smaller porosity, higher swelling ratio, and good compatibility, indicating their great potential for biomedical application.
The aim of this meta-analysis was to provide a comprehensive overview of human immunodeficiency virus (HIV)-1 subtypes and to investigate temporal and geographical trends of the HIV-1 epidemic among men who have sex with men (MSM) in China. Chinese and English articles published between January 2007 and December 2017 were systematically searched. Pooled HIV-1 prevalence was calculated, and its stability was analysed using sensitivity analysis. Subgroups were based on study time period, sampling area and prevalence. Publication bias was measured using Funnel plot and Egger's test. A total of 68 independent studies that included HIV-1 molecular investigations were eligible for meta-analysis. Circulating recombinant form (CRF) 01_AE (57.36%, 95% confidence interval (CI) 53.76–60.92) was confirmed as the most prevalent HIV-1 subtype among MSM in China. Subgroup analysis for time period found that CRF01_AE steadily increased prior to 2012 but decreased during 2012–2016. Further whereas CRF07_BC increased over time, B/B′ decreased over time. CRF55_01B has increased in recent years, with higher pooled estimated rate in Guangdong (12.22%, 95% CI 10.34–13.17) and Fujian (8.65%, 95% CI 4.98–13.17) provinces. The distribution of HIV-1 subtypes among MSM in China has changed across different regions and periods. HIV-1 strains in MSM are becoming more complex. Long-term molecular monitoring in this population remains necessary for HIV-1 epidemic control and prevention.
This paper presents new LA-ICP-MS zircon U–Pb chronology, whole-rock geochemical and zircon Hf isotopic data for the felsic lavas of the Huili Group from the southwestern Yangtze Block. LA-ICP-MS zircon U–Pb dating shows that these rocks were emplaced in Late Mesoproterozoic time (∼1028 to 1019 Ma). Relative to typical I-type and S-type granitoids, all the samples are characterized by low Sr and Eu, and high high-field-strength element contents, high TFeO/MgO, enriched rare earth element compositions and negative Eu anomalies, indicating that they share the geochemical signatures of A-type granitoid. They can be further divided into two groups: Group I and Group II. Group I are A1-type felsic rocks and were produced by fractional crystallization of alkaline basaltic magmas. The Group II felsic lavas belong to the A2-type and were derived by partial melting of a crustal source with mixing of mantle-derived magmas. Both Group I and Group II felsic lavas may erupt in a continental back-arc setting. The coexistence of A1- and A2-type rocks in the southwestern Yangtze Block suggests that they can occur in the same tectonic setting.
Feeding ruminants a high-grain (HG) diet is a widely used strategy to improve milk yield and cost efficiency. However, it may cause certain metabolic disorders. At present, information about the effects of HG diets on the systemic metabolic profile of goats and the correlation of such diets with rumen bacteria is limited. In the present study, goats were randomly divided into two groups: one was fed the hay diet (hay; n = 5), while the other was fed HG diets (HG; n = 5). On day 50, samples of rumen contents, peripheral blood serum and liver tissues were collected to determine the metabolic profiles in the rumen fluid, liver and serum and the microbial composition in rumen. The results revealed that HG diets reduced (P < 0.05) the community richness and diversity of rumen microbiota, with an increase in the Chao 1 and Shannon index and a decrease in the Simpson index. HG diets also altered the composition of rumen microbiota, with 30 genera affected (P < 0.05). Data on the metabolome showed that the metabolites in the rumen fluid, liver and serum were affected (variable importance projection > 1, P <0.05) by dietary treatment, with 47, 10 and 27 metabolites identified as differentially metabolites. Pathway analysis showed that the common metabolites in the shared key pathway (aminoacyl-transfer RNA biosynthesis) in the rumen fluid, liver and serum were glycine, lysine and valine. These findings suggested that HG diets changed the composition of the rumen microbiota and metabolites in the rumen fluid, liver and serum, mainly involved in amino acid metabolism. Our findings provide new insights into the understanding of diet-related systemic metabolism and the effects of HG diets on the overall health of goats.
Ca2+-sensing receptor (CaSR) represents a potential therapeutic target for inflammatory bowel diseases and strongly prefers aromatic amino acid ligands. We investigated the regulatory effects of dietary supplementation with aromatic amino acids – tryptophan, phenylalanine and tyrosine (TPT) – on the CaSR signalling pathway and intestinal inflammatory response. The in vivo study was conducted with weanling piglets using a 2 × 2 factorial arrangement in a randomised complete block design. Piglets were fed a basal diet or a basal diet supplemented with TPT and with or without inflammatory challenge. The in vitro study was performed in porcine intestinal epithelial cell line to investigate the effects of TPT on inflammatory response using NPS-2143 to inhibit CaSR. Dietary supplementation of TPT alleviated histopathological injury and decreased myeloperoxidase activity in intestine challenged with lipopolysaccharide. Dietary supplementation of TPT decreased serum concentration of pro-inflammatory cytokines (IL-1β, IL-6, IL-8, IL-12, granulocyte-macrophage colony-stimulating factor, TNF-α), as well as the mRNA abundances of pro-inflammatory cytokines in intestine but enhanced anti-inflammatory cytokines IL-4 and transforming growth factor-β mRNA levels compared with pigs fed control diet and infected by lipopolysaccharide. Supplementation of TPT increased CaSR and phospholipase Cβ2 protein levels, but decreased inhibitor of NF-κB kinase α/β and inhibitor of NF-κB (IκB) protein levels in the lipopolysaccharide-challenged piglets. When the CaSR signalling pathway was blocked by NPS-2143, supplementation of TPT decreased the CaSR protein level, but enhanced phosphorylated NF-κB and IκB levels in IPEC-J2 cells. To conclude, supplementation of aromatic amino acids alleviated intestinal inflammation as mediated through the CaSR signalling pathway.
A 24-year-old woman with a history of idiopathic dilated cardiomyopathy status post heart transplant gave birth to a healthy term female infant. At 5 months of age, the infant was diagnosed with severe left ventricular dysfunction with an ejection fraction of 18% and moderate non-compaction of the left ventricle. She received a heart transplant at 7 months of age. Familial dilated cardiomyopathy was diagnosed. Genetic testing revealed a likely pathogenic variant in the TPM1 gene. Fetal cardiac screening is critical for offspring of heart transplant recipients, especially when the reason for transplant was cardiomyopathy. Early genetic consultation and counselling is necessary for all heart transplant recipients, preferably prenatally. Postnatal screening of offspring is essential at birth, at 3-month intervals until 1 year of age, and then annually until the risk for familial cardiomyopathy is assessed.
Jurassic sandstones in the Xiongcun porphyry copper–gold district, southern Lhasa subterrane, Tibet, China were analysed for petrography, major oxides and trace elements, as well as detrital zircon U–Pb and Hf isotopes, to infer their depositional age, provenance, intensity of source-rock palaeo-weathering and depositional tectonic setting. This new information provides important evidence to constrain the tectonic evolution of the southern Lhasa subterrane during the Late Triassic – Jurassic period. The sandstones are exposed in the lower and upper sections of the Xiongcun Formation. Their average modal abundance (Q21F11L68) classifies them as lithic arenite, which is also supported by geochemical studies. The high chemical index of alteration values (77.19–85.36, mean 79.96) and chemical index of weathering values (86.19–95.59, mean 89.98) of the sandstones imply moderate to intensive weathering of the source rock. Discrimination diagrams based on modal abundance, geochemistry and certain elemental ratios indicate that felsic and intermediate igneous rocks constitute the source rocks, probably with a magmatic arc provenance. The detrital zircon ages (161–243 Ma) and εHf(t) values (+10.5 to +16.2) further constrain the sandstone provenance as subduction-related Triassic–Jurassic felsic and intermediate igneous rocks from the southern Lhasa subterrane. A tectonic discrimination method based on geochemical data of the sandstones, as well as detrital zircon ages from sandstones, reveals that the sandstones were most likely deposited in an oceanic island-arc setting. These results support the hypothesis that the tectonic background of the southern Lhasa subterrane was an oceanic island-arc setting, rather than a continental island-arc setting, during the Late Triassic – Jurassic period.
Iron-deficiency anemia is a public health concern that frequently occurs in pregnant mammals and neonatal offspring. Ferrous N-carbamylglycinate chelate (Fe-CGly) is a newly designed iron fortifier with proven effects in iron-deficient rats and weanling piglets. However, the effects of this new compound on pregnant mammals are unknown. Therefore, this experiment was conducted to evaluate the effects of Fe-CGly on sow reproductive performance and iron status of both sows and neonatal piglets. A total of 40 large-white sows after second parity were randomly assigned to two groups (n=20). They were receiving a diet including 80 mg Fe/kg as FeSO4 or Fe-CGly, respectively, from day 85 of gestation to parturition. The serum (day 110 of pregnancy) and placentas of sows were sampled. Litter size, mean weight of live born piglets, birth (live) litter weight, number of live born piglets, and the number of still-born piglets, mummies, and weak-born piglets were recorded. Once delivered, eight litters were randomly selected from the 20 litters per treatment, and one new-born male piglet (1.503±0.142 kg) from each selected litter was slaughtered within 3 h after birth from the selected litters, without colostrum ingestion. The serum, longissimus muscle, liver and kidneys of the piglets were collected. The iron status of the serum samples and the messenger RNA level of iron-related genes in the placenta, liver and kidney were analyzed. The results showed that litter weight of live born piglets was higher (P=0.030) in the Fe-CGly group (19.86 kg) than in the FeSO4 group (17.34 kg). Fe-CGly significantly increased placental iron concentration (P<0.05) of sows. It also significantly increased iron saturation and reduced the total iron-binding capacity of piglets (P<0.05) at birth. However, the results revealed that supplementation of Fe-CGly in sows reduced liver and kidney iron concentration of neonatal piglets (P<0.05), indicating decreased iron storage. In addition, the concentration of iron in the colostrum was not significantly changed. Therefore, the present results suggested that replacement of maternal FeSO4 supplement with Fe-CGly in the late-gestating period for sows could improve litter birth weight, probably via enhanced iron transportation in the placenta.
In recent years, hand, foot, and mouth disease (HFMD) has been increasingly recognized as a critical challenge to disease control and prevention in China. Previous studies have found that meteorological factors such as mean temperature and relative humidity were associated with HFMD. However, little is known about whether the diurnal temperature range (DTR) has any impact on HFMD. This study aimed to quantify the impact of DTR on childhood HFMD in 18 cities in Sichuan Province. A distributed lag non-linear model was adopted to explore the temporal lagged association of daily temperature with age-, gender- and pathogen-specific HFMD. A total of 290 123 HFMD cases aged 0–14 years were reported in the 18 cities in Sichuan Province. The DTR–HFMD relationships were non-linear in all subgroups. Children aged 6–14 years and male children were more vulnerable to the temperature changes. Large DTR had the higher risk estimates of HFMD incidence in cases of EV71 infection, while small DTR had the higher risk estimates of HFMD incidence in cases of CV-A16 infection. Our study suggested that DTR played an important role in the transmission of HFMD with non-linear and delayed effects.
Effects of Zn content on microstructures and mechanical properties of as-cast Mg–4Y–xZn alloys (x = 1, 2, 3, 4) have been studied in the article. The results indicate that the X phase is formed firstly, and the X phase, X + W phases, and W phase precipitated in order when the Zn contents increased from 1 to 4 wt%. The secondary dendritic arm spacing of the tested alloys first decrease as the Zn content increases within the range of 1–2% and then it increases. The ultimate tensile strength (UTS) and elongation of the four experimental alloys increase until the addition of Zn reaches to 3%, with the maximum values of 213.6 MPa and 11.82%, respectively. Besides, the fracture behaviors of Mg–4Y–1Zn, Mg–4Y–2Zn, and Mg–4Y–3Zn were a quasi-cleavage fracture, while Mg–4Y–4Zn belonged to cleavage.
In this work the quantitative and qualitative ability of a kinetic-theory-based two-fluid model (KT-TFM) is assessed in a state of fully periodic sedimentation (fluidization), with a focus on statistically steady, unstable (clustered) states. The accuracy of KT-TFM predictions is evaluated via direct comparison to direct numerical simulation (DNS) data. The KT-TFM and DNS results span a rather wide parameter space: mean-flow Reynolds numbers on the order of 1 and 10, mean solid volume fractions from 0.1 to 0.4, solid-to-fluid density ratios from 10 to 1000 and elastic and moderately inelastic (restitution coefficient of 0.9) conditions. Data from both KT-TFM and DNS display a rich variety of statistically steady yet unstable structures (clusters). Instantaneous snapshots of KT-TFM and DNS demonstrate remarkable qualitative agreement. This qualitative agreement is quantified by calculating the critical density ratio at which the structure transitions from a chaotic, dynamic state to a regular, plug-flow state, with good overall comparisons. Further quantitative assessments of mean and fluctuating velocities show good agreement at high density ratios but weaker agreement at intermediate to low density ratios depending on the mean-flow Reynolds numbers and solid fractions. Deviations of the KT-TFM results from the DNS data were traced to a breakdown in one of the underlying assumptions of the kinetic theory derivation: high thermal Stokes number. Surprisingly, however, even though the low Knudsen number assumption, also associated with the kinetic theory derivation, is violated throughout most of the parameter space, it does not seem to affect the good quantitative accuracy of KT-TFM simulations.
Antimony doped tin oxide (ATO) is an ideal material for thermal insulation. To obtain the stable performance of ATO nanodispersions and measure the transparent and thermal insulation properties of the ATO coatings, we investigated how a dispersant and sand milling affect the stability of ATO dispersion, which has come to the results that adding an appropriate dispersant and sand milling for 2.0 h were beneficial to the ATO dispersion. We characterized the morphology, nanostructure, particle size distribution, zeta potential, and optical properties of the ATO dispersions by a transmission electron microscope (TEM), a laser particle size analyzer, and a spectrometer. The results show that the average particle size of the dispersions is about 50 nm and their absolute values of all zeta potentials are more than 40 mV. We coated the thermal insulation water-based coatings on quartz glasses by spin coating method, the effect of thermal insulation is evident with the increase of the ATO content, and there exists approximately 10 °C difference between the ATO sample and blank sample with the condition for maintaining high transmittance of visual light.
Apical membrane antigen-1 (AMA-1) has been described as a potential vaccine candidate in apicomplexan parasites. Here we characterize the ama-1 gene. The full-length ama-1 gene of Babesia sp. BQ1 (Lintan) (BLTAMA-1) is 1785 bp, which contains an open reading frame (ORF) encoding a 65-kDa protein of 594 amino acid residues; by definition, the 5′ UTR precedes the first methionine of the ORF. Phylogenetic analysis based on AMA-1 amino acid sequences clearly separated Piroplasmida from other Apicomplexa parasites. The Babesia sp. BQ1 (Lintan) AMA-1 sequence is most closely associated with that of B. ovata and B. bigemina, with high bootstrap value. A recombinant protein encoding a conserved region and containing ectodomains I and II of BLTAMA-1 was constructed. BLTrAMA-1-DI/DII proteins were tested for reactivity with sera from sheep infected by Babesia sp. BQ1 (Lintan). In Western-blot analysis, native Babesia sp. BQ1 (Lintan) AMA-1 proteins were recognized by antibodies raised in rabbits against BLTrAMA-1 in vitro. The results of this study are discussed in terms of gene characterization, taxonomy and antigenicity.
