The objective of this study was to test the hypothesis that stearic acid (SA) supplementation increases milk fat content and overcomes the antilipogenic effects of trans-10, cis-12 conjugated linoleic acid (CLA) in lactating ewes. Twenty-eight Lacaune ewes (36 (sd 2) days in lactation; 70·5 (sd) 9·6 kg of body weight), producing 1·8 (sd 0·4) kg of milk/d, were used in a completely randomised design (seven ewes/treatment) for 21 d. The treatments were: (1) Control; (2) CLA (6·4 g/d of trans-10, cis-12 CLA); (3) SA (28 g/d of SA) and (4) SA in association with trans-10, cis-12 CLA (CLASA; 6·4 g/d of trans-10, cis-12 CLA plus 28 g/d of SA). All data were analysed using a mixed model that included the fixed effect of treatment and the random effect of ewe. SA did not alter milk fat content and yield relative to Control (91·9 v. 91·2 (sd 4·1) g/d). CLASA was not able to overcome the reduction in fat content and fat yield induced by CLA (75 v. 82 (sd 0·14) g/d). SA increased the relative abundance of CD36, fatty acid-binding protein 4 (FABP4) and PPAR-γ mRNA by 140, 112 and 68 % compared with CLASA. SA also reduced the relative abundance of acetyl-CoA carboxylase α promoter II and stearoyl-CoA desaturase (SCD) when compared with Control (45 and 39 %). Compared with CLA, CLASA treatment had no effect on the mRNA abundance of fatty acid synthase, lipoprotein lipase, CD36, SCD, FABP4, acylglycerolphosphate acyltransferase 6, sterol regulatory element-binding protein 1 and PPAR-γ. In conclusion, SA supplementation did not increase milk fat synthesis and did not overcome the CLA-induced milk fat depression when associated with trans-10, cis-12 CLA.

The diagnosis of visceral leishmaniasis (VL) has improved with the search of novel antigens; however, their performance is limited when samples from VL/human immunodeficiency virus (HIV)-coinfected patients are tested. In this context, studies conducted to identify more suitable antigens to detect both VL and VL/HIC coinfection cases should be performed. In the current study, phage display was performed using serum samples from healthy subjects and VL, HIV-infected and VL/HIV-coinfected patients; aiming to identify novel phage-exposed epitopes to be evaluated with this diagnostic purpose. Nine non-repetitive and valid sequences were identified, synthetized and tested as peptides in enzyme-linked immunosorbent assay experiments. Results showed that three (Pep2, Pep3 and Pep4) peptides showed excellent performance to diagnose VL and VL/HIV coinfection, with 100% sensitivity and specificity values. The other peptides showed sensitivity varying from 50.9 to 80.0%, as well as specificity ranging from 60.0 to 95.6%. Pep2, Pep3 and Pep4 also showed a potential prognostic effect, since specific serological reactivity was significantly decreased after patient treatment. Bioinformatics assays indicated that Leishmania trypanothione reductase protein was predicted to contain these three conformational epitopes. In conclusion, data suggest that Pep2, Pep3 and Pep4 could be tested for the diagnosis of VL and VL/HIV coinfection.

This study aimed to determine the requirements of standardized ileal digestible (SID) methionine (Met) + cysteine (Cys) of 15–30 kg barrows, maintaining or increasing the 1:1 ratio between SID Met and SID Cys. Seventy crossbred barrows averaging 15.2 ± 0.54 kg of live weight were allotted in a randomized block design 2 × 3 + 1 factorial scheme, with five replicates and two animals per pen. Treatments consisted of maintaining or increasing the 1:1 ratio between SID Met and SID Cys, three levels of SID Met + Cys (5.8, 6.4 and 7.0 g/kg) and a basal diet containing the lowest SID Met + Cys level (5.2 g/kg), formulated to provide a 1:1 ratio of SID Met (2.6 g/kg) and SID Cys (2.6 g/kg). Performance, blood parameters, longissimus dorsi muscle depth, backfat thickness and S–S linkages in the bristles were evaluated. The best average daily gain was estimated at 6.61 g/kg of SID Met + Cys, without maintaining the 1:1 ratio between sulphur amino acids (SAA). The daily intake of SID Met + Cys increased due to dietary SID Met + Cys levels, maintaining or increasing the same ratio between SAA. Plasma glucose increased and total cholesterol decreased according to SID Met + Cys levels, without maintaining the same ratio between the SAA. The requirement of SID Met + Cys for 15–30 kg barrows was 6.61 g/kg (7.88 g/day) for an optimum average daily gain, with no need to maintain the same ratio between the SAA.

The nanoencapsulation of biocomposites with anthelmintic action has been proposed as an alternative for improving their efficiency. Thus, the current study aimed to evaluate the efficacy of carvacryl acetate nanoencapsulated with biopolymers (nCVA) in the control of sheep gastrointestinal nematodes. CVA was nanoencapsulated with chitosan/chichá gum and characterized in terms of its efficacy of encapsulation (EE), yield and zeta potential. The acute toxicity of nCVA was evaluated in mice. For the fecal egg count reduction test, 40 animals were divided into four groups (n = 10) and orally administered the following treatments: G1, 250 mg kg−1 CVA; G2, 250 mg kg−1 nCVA; G3, chitosan/chichá gum (negative control) and G4, 2.5 mg kg−1 monepantel (positive control). Feces were collected on days 0 and 16 posttreatment to determine the eggs per gram of feces (epg). The EE and yield of nCVA were 72.8 and 57.5%, respectively. The nanoparticles showed a size of 764.5 ± 302.5 nm, and the zeta potential at pH 3.2 was +22.0 mV. nCVA presented a 50% lethal dose (LD50) of 2609 mg kg−1. By 16 days posttreatment, CVA, nCVA and monepantel reduced the epg by 52.9.7, 71.5 and 98.7%, respectively, and the epg of sheep treated with nCVA differed from that of the negative control (P > 0.05) but did not differ from that of sheep treated with CVA. In conclusion, the nanoencapsulation of CVA reduced its toxicity, and nCVA showed anthelmintic activity.

Identifying dietary patterns in different environments attended by children is relevant to guide public politics. The aim of this study was to analyse the factors associated with dietary patterns of children under 2 years of age in childcare centres and at homes. This transversal study was enrolled in municipal childcare centres of Guaratuba, Paraná, Brazil. Food consumption data from 256 children were obtained by the food record method. From the consumption data, four dietary patterns were identified by factor analysis using the principal component method in each environment. ‘Traditional’ patterns were observed in both environments; the ‘less healthy’ pattern was found only at homes. Other patterns identified in childcare centres include ‘snacks’, ‘nutritive’ and ‘pasta and meats’; at homes, it was possible to identify patterns such as ‘milk and cereals’ and ‘mixed’. Children over 12 months presented higher scores for all the patterns in both environments. Obese children had lower scores for all the patterns in childcare centres. There was an association between maternal age below 21 years and higher adherence to a ‘less healthy’ pattern, maternal level of education less than 8 years and lower adherence to the ‘mixed’ pattern and lower familiar income per capita and higher adherence to the ‘snacks’ and ‘traditional’ patterns in childcare centres. In conclusion, adherence to dietary patterns was associated with socio-economic, demographic variables and nutritional status and further studies are needed, especially those with a longitudinal design, enabling the monitoring of dietary patterns.

We investigated the effects of pathogens associated with subclinical intramammary infections on yield, composition and quality indicators of goat milk. By means of a longitudinal study, individual half udder milk samples (n = 132) were collected at different lactation periods and assessed for milk yield and physicochemical composition, somatic cell count (SCC), total bacteria count (TBC) and microbiological culture. Staphylococci species accounted for the great majority of the isolates (96.1%). Intramammary infections significantly reduced fat and total solids in goat milk and increased both SCC and TBC. However, these indicators were significantly higher in udder halves affected by S. aureus compared with other staphylococci species.

Amid widespread occurrence of herbicide-resistant weeds in the United States, the use of PRE herbicides and cover crops have resurged once again as important strategies for weed management in cropping systems. The objective of this experiment was to evaluate the length of soil residual weed control from PRE soybean herbicides and the detrimental impact of these herbicides on cover crop species using field treated soil in greenhouse bioassays. Greenhouse bioassays were conducted using soil from field experiments conducted in 2018 and 2019 in Arlington and Lancaster, WI. PRE herbicides consisted of imazethapyr, chlorimuron-ethyl, and cloransulam-methyl (acetolactate synthase [ALS] inhibitors); metribuzin (photosystem II [PS II] inhibitor); sulfentrazone, flumioxazin, and saflufenacil (protoporphyrinogen oxidase [PPO] inhibitors); acetochlor, S-metolachlor, dimethenamid-P, and pyroxasulfone (very long-chain fatty acid [VLCFA] inhibitors); and a nontreated control. Greenhouse bioassays were conducted using soil (depth, 0 to 10 cm) sampled at 0, 10, 20, 30, 40, and 50 d after treatment (DAT). Palmer amaranth and giant foxtail (weeds), and radish and cereal rye (cover crops) were used as bioindicators of herbicide levels in the soil. Bioassay results showed extended soil residual control of Palmer amaranth with sulfentrazone and pyroxasulfone; extended residual control of giant foxtail was observed with pyroxasulfone and S-metolachlor. Chlorimuron-ethyl and metribuzin were the most injurious herbicides to radish and cereal rye shortly after application, respectively, but minimal injury was observed from soil samples collected 50 DAT, indicating the use of PRE and fall-seeded cover crops in southern Wisconsin can be compatible. These results can support growers and practitioners with selection of effective PRE herbicides for Palmer amaranth and giant foxtail control and reduced impact on fall-seeded radish and cereal rye cover crops, altogether leading to more effective, diverse, and sustainable weed management programs.

It was aimed to simultaneously study standardized ileal digestible (SID) tryptophan (Trp) and lysine (Lys) for gilts. A digestibility assay was previously conducted to determine the SID amino acid in the basal diet (low levels of SID Trp and Lys). Sixty-four gilts (15.04 ± 1.44 kg) were allotted to 16 diets in a 4 × 4 factorial arrangement (1.55, 1.85, 2.15 and 2.45 g/kg SID Trp and 9.72, 11.12, 12.52 and 13.92 g/kg SID Lys) with four replicates per treatment. Performance, longissimus muscle (LM), backfat thickness (BF) and blood variables were evaluated. An interaction was observed for G:F, and by response surface model, the optimum Trp level was achieved at 2.15 g/kg (0.159 g/MJ of ME). A quadratic effect of Trp was observed on body weight (BW) and average daily gain (ADG); the daily feed intake increased linearly as Trp increased. The optimum Trp levels of 2.25 and 2.24 g/kg were estimated for BW and ADG, respectively. The BF increased with increasing levels of Trp. There was a quadratic and linear effect of Trp and Lys, respectively, on the LM, in which the optimum Trp level was determined as 2.05 g/kg in the diet. Plasma urea nitrogen decreased as Trp and Lys levels increased. Using estimates provided by response surface, maximized G:F ratio was obtained at 2.15 g SID Trp/kg of diet and at least 13.92 g SID Lys/kg of diet is necessary to optimize the G:F for 15–30 kg gilts, providing a Trp:Lys ratio of 15.4:100.

We evaluated the differences between the supplementation of urea in rumen and/or abomasum on forage digestion, N metabolism and urea kinetics in cattle fed a low-quality tropical forage. Five Nellore heifers were fitted with rumen and abomasum fistulas and assigned to a Latin square design. The treatments were control, continuous infusion of urea in the abomasum (AC), continuous infusion of urea in the rumen, a pulse dose of urea in the rumen every 12 h (PR) and a combination of PR and AC. The control exhibited the lowest (P < 0·10) faecal and urinary N losses, which were, overall, increased by supplementation. The highest urinary N losses (P < 0·10) were observed when urea was either totally or partially supplied as a ruminal pulse dose. The rumen N balance was negative for the control and when urea was totally supplied in the abomasum. The greatest microbial N production (P < 0·10) was obtained when urea was partially or totally supplied in the abomasum. Urea supplementation increased (P < 0·10) the amount of urea recycled to the gastrointestinal tract and the amount of urea-N returned to the ornithine cycle. The greatest (P < 0·10) amounts of urea-N used for anabolism were observed when urea was totally and continuously infused in the abomasum. The continuous abomasal infusion also resulted in the highest (P < 0·10) assimilation of microbial N from recycling. The continuous releasing of urea throughout day either in the rumen or abomasum is able to improve N accretion in the animal body, despite mechanism responsible for that being different.

The farrowing process is one of the most energy-demanding activities for the modern hyperprolific sow. This study evaluated the effects of supply of energy on the expected date of farrowing on the farrowing kinetics and piglets’ performance during the first 24 h after birth. A total of 80 sows were used. The sows and their respective litters were considered as the experimental unit. On the expected day of farrowing, the sows were allocated to one of the following groups: sows that did not have access to feed from farrowing induction until the end of the farrowing process (CON, n = 40); sows fed 500 g of energetic supplement, which consisted of 250 g of the basal lactation diet plus 250 g of cane sugar, 18 h after farrowing induction (SUP, n = 40). The farrowing duration, farrowing assistance, birth interval, number of total born, stillborn and mummified piglets were recorded for each sow. Piglets were weighed individually at birth and 24 h later. The interval from birth to first suckle was evaluated individually for each piglet in 16 randomly selected litters (eight litters per treatment group). Blood glucose concentrations of six sows were measured shortly after expulsion of the first piglet. Farrowing duration, farrowing assistance and stillborn rate tended to be greater (P = 0.06, P = 0.09 and P = 0.07, respectively) in sows from the CON group compared to sows from the SUP group. However, there was no difference (P > 0.05) between the groups for birth interval. Colostrum intake was greater (P < 0.05) for piglets from the SUP group compared to piglets from the CON group. Additionally, BW gain of the piglets suckling the SUP group was greater (P < 0.05) than those suckling the CON group at 24 h after birth. The blood glucose concentrations during the expulsive stage of farrowing were greater (P < 0.05) in the SUP group than for sows from the CON group. In conclusion, supplying modern hyperprolific sows energy on the expected day of farrowing is a valuable nutritional intervention to improve the farrowing kinetics and piglets’ performance in early life.

Spontaneous abortion is considered a public health problem having several causes, including infections. Among the infectious agents, bacteria of the vaginal microbiota and Ureaplasma parvum have been associated with abortion, but their participation needs to be further elucidated. This study aims to evaluate the influence of Mollicutes on the development of spontaneous abortion. Women who underwent spontaneous abortion and those with normal birth (control) were studied. Samples of cervical mucus (CM) and placental tissue were collected to identify Mollicutes using the quantitative polymerase chain reaction methodology. Eighty-nine women who had a miscarriage and 20 women with normal pregnancies were studied. The presence of Mollicutes in placental tissue increased the chance of developing miscarriage sevenfold. The prevalence of U. parvum in women who experienced spontaneous abortion was 66.3% in placental tissue. A positive association was observed between the detection of U. parvum in samples of placental tissue and abortion. There was a significant increase in microbial load in placental tissue for M. hominis, U. urealyticum and U. parvum compared to the control group. Detection of U. parvum in CM in pregnant women can ascend to the region of the placental tissue and trigger a spontaneous abortion.

The co-infection between visceral leishmaniasis (VL) and human immunodeficiency virus (HIV) has increased in several countries in the world. The current serological tests are not suitable since they present low sensitivity to detect the most of VL/HIV cases, and a more precise diagnosis should be performed. In this context, in the present study, an immunoproteomics approach was performed using Leishmania infantum antigenic extracts and VL, HIV and VL/HIV patients sera, besides healthy subjects samples; aiming to identify antigenic markers for these clinical conditions. Results showed that 43 spots were recognized by antibodies in VL and VL/HIV sera, and 26 proteins were identified by mass spectrometry. Between them, β-tubulin was expressed, purified and tested in ELISA experiments as a proof of concept for validation of our immunoproteomics findings and results showed high sensitivity and specificity values to detect VL and VL/HIV patients. In conclusion, the identified proteins in the present work could be considered as candidates for future studies aiming to improvement of the diagnosis of VL and VL/HIV co-infection.

Chronically stressed adult male Balb C mice were submitted to the tail suspension test. Chronic immobilization stress (6 h/d for 14 consecutive days) induced a significant reduction in immobility time when compared to non-stressed controls. Pretreatment with LY 53857, a serotonin 5HT2 antagonist, and IPS 339, a selective beta-2 adrenoceptor blocker, reversed immobility time to the levels of non-stressed controls. Chronic administration of corticosterone (100 mg/kg for 7 d) did not modify immobility time as compared to saline treated controls. It is suggested that both serotonergic and adrenergic pathways in the brain may participate in the stress-induced changes occurring in the tail suspension test response and that corticosterone does not appear to play a role in this process.

Chitosan (CHI) is a natural biopolymer with antimicrobial, anti-inflammatory, antioxidant and digestive modulatory effects, which can be used in the ruminant diet to replace antibiotics. The aim of this study was to evaluate the effects of CHI on lamb growth traits, nutrients digestibility, muscle and fatty deposition, meat fatty acid (FA) profile, meat quality traits and serum metabolome. Thirty 30-month-old male lambs, half Suffolk and half Dorper, with an average BW of 21.65 ± 0.86 kg, were fed in a feedlot system for a total of 70 days. The lambs were separated into two groups according to the diet: the control (CON) group which received the basal diet and the CHI group which received the basal diet with the addition of CHI as 2 g/kg of DM in the diet. Lambs supplemented with CHI had a greater (P < 0.05) final BW, DM intake, final body metabolic weight (P < 0.05) and lower residual feed intake than the CON group. Animals fed CHI had a greater (P < 0.05) starch digestibility at 14 and 28 days, average daily gain at 14, 42 and 56 days, greater feed efficiency at 28 days and feed conversation at 14 and 42 days in feedlot. Most of the carcass traits were not affected (P > 0.05) by the treatment; however, the CHI supplementation improved (P < 0.05) dressing and longissimus muscle area. The treatments had no effect (P > 0.05) on the meat colour and other quality measurements. Meat from the CHI-fed lambs had a greater concentration (P < 0.05) of oleic-cis-9 acid, linoleic acid, linolenic-trans-6 acid, arachidonic acid and eicosapentaenoic acid. According to the variable importance in projection score, the most important metabolites to differentiate between the CON and the CHI group were hippurate, acetate, hypoxanthine, arginine, malonate, creatine, choline, myo-inositol, 2-oxoglutarate, alanine, glycerol, carnosine, histidine, glutamate and 3-hydroxyisobutyrate. Similarly, fold change (FC) analysis highlighted succinate (FC = 1.53), arginine (FC = 1.51), hippurate (FC = 0.68), myo-inositol (FC = 1.48), hypoxanthine (FC = 1.45), acetate (FC = 0.73) and malonate (FC = 1.35) as metabolites significantly different between groups. In conclusion, the present data showed that CHI changes the muscle metabolism improving muscle mass deposition, the lamb’s performance and carcass dressing. In addition, CHI led to an alteration in the FA metabolism, changes in the meat FA profile and improvements in meat quality.

Schizophrenia (SCZ) is a complex and severe chronic mental disease presenting deficits in an operational measure of sensorimotor gating and cognitive impairments. The history of delayed developmental milestones and the presence of minor physical abnormalities may represent indirect evidences of abnormal prenatal development. Premorbid cognitive, personality and social functioning deficits in patients severely affected are also reported. Among the several candidate risk genes potentially associated with SCZ via pleiotropic mechanisms and/or other genes specific to susceptibility for SCZ, the Disrupted-in-Schizophrenia 1 gene (DISC1) is the most studied. Interestingly, the main binding partner and effector of DISC1 protein is the Nuclear-distribution gene E homolog like-1 (Ndel1), which is an oligopeptidase capable to degrade small peptides such as bradykinin (BK) and neurotensin (NT). Both neuropeptides were implicated in SCZ, and NT was also suggested to be an endogenous antipsychotic. DISC1 binding inhibits the Ndel1 enzyme activity competing with the peptide substrates, and the described translocation of DISC1 gene cosegregated with SCZ suggest a DISC1 and Ndel1 complex formation impairment with potential deregulation of the Ndel1 total activity. Other oligopeptidase, as Angiotensin I-Converting Enzyme (ACE), is also able to cleave these peptides. Therefore, our group is focused on measuring the oligopeptidase activity not only in clinical samples to allow the comparison between patients and health controls (HCs), but we have also performed the same measurements in plasma and different brain regions of animal models aiming to have insights into what may be occurring in the human brain based on peripheral tissues measurements. A significant lower Ndel1 oligopeptidase activity (Gadelha et al., J Psych Res 2013) and significant higher ACE activity (Gadelha and Vendramine et al., submitted 2014) in the plasma of SCZ patients compared to HCs were observed. Moreover, a potential association of the ACE oligopeptidase activity with the cognitive/disorganization symptoms was observed in both SCZ patients and animal models. The evaluation of the activity of these oligopeptidases in patients and animal models treated with the same antipsychotics are currently ongoing in our laboratory. The results presented here support the potential involvement of Ndel1 and ACE in SCZ, and they may contribute to the discovery of molecular biomarkers for diagnosis and/or treatment follow-up of a severe chronic mental disease as SCZ, aiming to contribute to foster the translation of basic neurobiological and behavioral research to an improved integrative understanding of psychopathology for the development of a new and/or optimized treatments.
