The timing and duration of the coldest period in the last glacial stage, often referred to as the last glacial maximum (LGM), has been observed to vary spatially and temporally. In Australia, this period is characterised by colder, and in some places more arid, climates than today. We applied Monte-Carlo change point analysis to all available continuous proxy records covering this period, primarily pollen records, from across Australia (n = 37) to assess this change. We find a significant change point occurred (within uncertainty) at 28.6 ± 2.8 ka in 25 records. We interpret this change as a shift to cooler climates, associated with a widespread decline in biological productivity. An additional change point occurred at 17.7 ± 2.2 ka in 24 records. We interpret this change as a shift towards warmer climates, associated with increased biological productivity. We broadly characterise the period between 28.6 (± 2.8) – 17.7 (± 2.2) ka as an extended period of maximum cooling, with low productivity vegetation that may have occurred as a combined response to reduced temperatures, lower moisture availability and atmospheric CO2. These results have implications for how the spatial and temporal coherence of climate change, in this case during the LGM, can be best interrogated and interpreted.

OBJECTIVES/GOALS: Our goal is to develop a non-invasive stimulation technique using magneto-electric nanoparticles (MENs) for inducing and enhancing neuronal activity with high spatial and temporal resolutions and minimal toxicity, which can potentially be used as a more effective approach to brain stimulation. METHODS/STUDY POPULATION: MENs compose of core-shell structures that are attracted to strong external magnetic field (~5000 Gauss) but produces electric currents with weaker magnetic field (~450 Gauss). MENs were IV treated into mice and drawn to the brain cortex with a strong magnetic field. We then stimulate MENs with a weaker magnetic field via electro magnet. With two photon calcium imaging, we investigated both the temporal and spatial effects of MENs on neuronal activity both in vivo and in vitro. We performed mesoscopic whole brain calcium imaging on awake animal to assess the MENs effects. Furthermore, we investigated the temporal profile of MENs in the vasculatures post-treatment and its toxicities to CNS. RESULTS/ANTICIPATED RESULTS: MENs were successfully localized to target cortical regions within 30 minutes of magnetic application. After wirelessly applying ~450 G magnetic field between 10-20 Hz, we observed a dramatic increase of calcium signals (i.e. neuronal excitability) both in vitro cultured neurons and in vivo treated animals. Whole brain imaging of awake mice showed a focal increase in calcium signals at the area where MENs localized and the signals spread to regions further away. We also found MENs stimulatory effects lasted up to 24 hours post treatment. MEN stimulation increases c-Fos expression but resulted in no inflammatory changes, up to one week, by assessing microglial or astrocytes activations. DISCUSSION/SIGNIFICANCE OF IMPACT: Our study shows, through controlling the applied magnetic field, MENs can be focally delivered to specific cortical regions with high efficacy and wirelessly activated neurons with high spatial and temporal resolution. This method shows promising potential to be a new non-invasive brain modulation approach disease studies and treatments.

A thorough understanding of how communities respond to extreme changes, such as biotic invasions, is essential to manage ecosystems today. Here we constructed fossil food webs to identify changes in Late Ordovician (Katian) shallow-marine paleocommunity structure and functioning before and after the Richmondian invasion, a well-documented ancient invasion. Food webs were compared using descriptive metrics and cascading extinction on graphs models. Richness at intermediate trophic levels was underrepresented when using only data from the Paleobiology Database relative to museum collections, resulting in a spurious decrease in modeled paleocommunity stability. Therefore, museum collections and field sampling may provide more reliable sources of data for the reconstruction of trophic organization in comparison to online data repositories. The invasion resulted in several changes in ecosystem dynamics. Despite topological similarities between pre- and postinvasion food webs, species loss occurred corresponding to a minor decrease in functional groups. Invaders occupied all of the preinvasion functional guilds, with the exception of four incumbent guilds that were lost and one new guild, corroborating the notion that invaders replace incumbents and fill preexisting niche space. Overall, models exhibited strong resistance to secondary extinction, although the postinvasion community had a lower threshold of collapse and more variable response to perturbation. We interpret these changes in dynamics as a decrease in stability, despite similarities in overall structure. Changes in food web structure and functioning resulting from the invasion suggest that conservation efforts may need to focus on preserving functional diversity if more diverse ecosystems are not inherently more stable.

The mid-to-late Pleistocene Devils Hole δ18O record has been extended from 60,000 to 4500 yr ago. The new δ18O time series, in conjunction with the one previously published, is shown to be a proxy of Pacific Ocean sea surface temperature (SST) off the coast of California. During marine oxygen isotope stages (MIS) 2 and 6, the Devil Hole and SST time series exhibit a steady warming that began 5000 to > 10,000 yr prior to the last and penultimate deglaciations. Several possible proximate causes for this early warming are evaluated. The magnitude of the peak δ18O or SST during the last interglacial (LIG) is significantly greater (1 per mill and 2 to 3°C, respectively) than the peak value of these parameters for the Holocene; in contrast, benthic δ18O records of ice volume show only a few tenths per mill difference in the peak value for these interglacials. Statistical analysis provides an estimate of the large shared information (variation) between the Devils Hole and Eastern Pacific SST time series from ∼ 41 to ∼ 2°N and enforces the concept of a common forcing among all of these records. The extended Devils Hole record adds to evidence of the importance of uplands bordering the eastern Pacific as a source of archives for reconstructing Pacific climate variability.

Many advances in disciplines such as chemistry, biochemistry, plant breeding, genetics, engineering, and others have been applied in a positive manner to improve knowledge in weed science. The emerging field of genomics is likely to have a similar positive effect on our understanding of weeds and their management in various plant agriculture systems. Genomics involves the large-scale use of molecular techniques for identification and functional analysis of complete or nearly complete genomic complements of genes. Commercial application of genomics has already occurred for improvement in certain crop input and output traits, including improved quality characteristics and herbicide and insect resistance. Additional commercial applications of genomics in weed science will be identification of genes involved in a crops' competitive ability. Genes controlling early crop shoot emergence, rapid early-season leaf and root development for fast canopy closure, production of allelochemicals for natural weed control, identification of novel herbicide target sites, resistance mechanisms, and genes for safening crops against specific herbicides can and will be identified. Successful crop improvement in these areas using the tools of genomics will dramatically affect weed–crop interactions and improve crop yields while reducing weed problems. In relation to improved basic knowledge of weeds and the resulting ability to improve our weed management techniques, genomics will offer the weed science community many new and exciting research opportunities. Scientists will be able to determine the genetic composition of weed populations and how it changes over time in relation to agricultural practices. Identification of genes contributing to weediness, perennial growth habit, herbicide resistance, seed and vegetative structure dormancy, plant architecture and morphology, plant reproductive characters (outcrossing and hybridization, introgression), and allelopathy will be identified and utilized with high-throughput DNA sequencing and other genomics-based technologies. Using genomics to improve our understanding of weed biology by determining which genes function to affect the fitness, competitiveness, and adaptation of weeds in agricultural environments will allow the development of improved management strategies. This review provides a summary of the various plant genomic research methods being used. Information is provided concerning the current state of molecular research in various areas of weed science and specific genomic research currently being conducted at Purdue University using transfer DNA (T-DNA) activation tagging to generate large populations of mutated plants that can be screened for genes of importance to weed science.

With the loss of methyl bromide (MeBr) and high prices of alternatives, tomato growers are applying lower fumigant rates or adopting a reduced system. Without the broad-spectrum control provided by the complete fumigant system, a fallow weed program might be needed to avoid an increase in pest pressure with consecutive years of application of the reduced-fumigant system. Nutsedges are among the pests of interest due to their fast reproduction by underground structures and ability to spread and quickly infest a field. Field trials were conducted between February and December of 2011 in Balm, FL, to evaluate the impacts of fallow treatments, fumigants, and halosulfuron on nutsedge control. The trial design was a split–split plot with main, sub-, and subsubplots being fallow, fumigant, and herbicide treatment, respectively. Fallow treatments were spaced evenly throughout the fallow season and consisted of sequential combinations of cultivation (C) and/or glyphosate (G) applied at 2.24 kg ae ha−1; including: C, CC, G, GG, CG, GC, GCG, and NO (nontreated control). Fumigant treatments included a reduced-fumigant system of 1,3-dichloropropene plus chloropicrin 40:60 (1,3-D + pic) at 281 kg ha−1, a complete fumigant system of dimethyl disulfide plus chloropicrin 79:21 (DMDS + pic) at 545 kg ha−1, and no fumigant (NoFum). Herbicide treatments were either two POST applications of halosulfuron at 39 g ai ha−1 (Hal) or no halosulfuron (NoHal). In general, the fallow weed treatment GCG was the most effective in reducing nutsedge shoot emergence through the plastic mulch. When the reduced-fumigant system 1,3-D + pic was combined with GCG fallow treatment and halosulfuron (GCG:1,3-D + pic:Hal), no differences were found between this combination and the complete fumigant system DMDS + pic with halosulfuron and combined with CC, G, GG, CG, GC or GCG. This study shows the importance of adding a fallow weed program and halosulfuron to either a reduced or complete fumigation system to minimize the reproduction and growth of nutsedges.

As the amount of methyl bromide approved for use in Florida strawberry diminishes, growers are faced with a forced transition to alternative fumigants. Many of these methyl bromide alternatives have been associated with reductions in weed control, requiring additional but complementary measures. POST herbicide options for annual strawberry are limited, resulting in significant portions of the strawberry acreage in Florida being hand-weeded when troublesome weeds escape conventional control methods. Strawberry has shown acceptable tolerance to clopyralid in other areas and production systems; however, its integration into the Florida production system and ramifications of applications during fruiting warrants further research. Eight trials were conducted, with three common strawberry cultivars grown in West Central Florida subjected to POST spray and drip-tape-injected applications of clopyralid. Formation of new strawberry leaves was not affected by clopyralid application, except for a reduction in new leaves of the cultivar ‘Strawberry Festival' at the highest rate of application of 261 g ae ha−1 in comparison with the nontreated control. Strawberry leaf malformation was best explained by an exponential growth equation, whereas marketable yield followed the trend of a Weibull peak. At the maximum labeled rate (66 g ha−1), leaf malformation was less than 5% for all cultivars tested, and marketable yield was estimated at 104% of the nontreated control.

For $T$ a compact torus and $E_{T}^{*}$ a generalized $T$-equivariant cohomology theory, we provide a systematic framework for computing $E_{T}^{*}$ in the context of equivariantly stratified smooth complex projective varieties. This allows us to explicitly compute $H_{T}^{*}\left( X \right)$ as an $H_{T}^{*}\left( pt \right)$-module when $X$ is a direct limit of smooth complex projective ${{T}_{\mathbb{C}}}$-varieties. We perform this computation on the affine Grassmannian of a complex semisimple group.

Farms producing similar products have become increasingly concentrated geographically over the past century in the United States (US). Due to the concentration of food production, a disruption in key production areas may reduce the availability of certain foods nationwide. For example, climate change poses such a threat, with projections of altered precipitation patterns, increased temperature and pest outbreaks, which may result in reduced crop yields and geographic shifts in crop adaptation. Analyses of the degree to which US regions can satisfy the food needs of their resident populations—a concept we refer to as regional self-reliance (RSR)—are therefore warranted. We focus on the Northeast region because of its high population density and declining agricultural landbase. Our objectives are to: (1) determine how agricultural land is used in the Northeast region; (2) determine the variety and amount of foods produced; and (3) analyze the relationship between food consumption and agricultural output. Annual (2001–2010) data on land area, yield and output of all crops and major livestock categories, as well as seafood landings, were catalogued. National annual (2001–2009) data on food availability were used as a proxy for estimates of food consumption, and these data were downscaled to a regional level and compared with regional production data in order to estimate RSR. In the Northeast region, approximately 65% of land in farms contributed directly to the food supply from 2001 to 2010, although this varied significantly across states. Just over one-half of all land in farms in the region was devoted to the production of livestock feed. The region produced >100 food crops annually from 2001 to 2009, and vegetables represented the majority of food crop production by weight. Chicken accounted for the largest weight of meat products produced. Compared to the Northeast region's share (~6%) of total land in farms in the nation, it accounted for disproportionately higher amounts of the national production of dairy (16%), eggs (13%), chicken (9%), lamb (7%) and vegetables (7%). However, the region accounted for ~22% of the national population and therefore produced a disproportionately low share of food on a per capita basis. RSR for plant-based foods was lowest for pulses (7%) and highest for vegetables (26%). There are four specific factors in the RSR in our analysis, each of which could result in substantial shifts (upward or downward) of the RSR in the future: land used for agriculture, crop (or animal) productivity, population and dietary preferences.
