Carotenoids are important bioactive substances in breast milk, the profile of which is seldom studied. This study aimed to explore the profile of carotenoids in breast milk and maternal/cord plasma of healthy mother–neonate pairs in Shanghai, China, and their correlation with dietary intake. Maternal blood, umbilical cord blood and breast milk samples from five lactation stages (colostrum, transitional milk and early-, mid- and late-term mature milk) were collected. Carotenoid levels were analysed by HPLC. Carotenoid levels in breast milk changed as lactation progressed (P < 0·001). β-Carotene was the primary carotenoid in colostrum. Lutein accounted for approximately 50 % of total carotenoids in transitional milk, mature milk and cord blood. Positive correlations were observed between five carotenoids in umbilical cord blood and maternal blood (P all < 0·001). β-Carotene levels were also correlated between maternal plasma and three stages of breast milk (r = 0·605, P < 0·001; r = 0·456, P = 0·011, r = 0·446; P = 0·013, respectively). Dietary carotenoid intakes of lactating mothers also differed across lactation stages, although no correlation with breast milk concentrations was found. These findings suggest the importance of exploring the transport mechanism of carotenoids between mothers and infants and help guide the development of formulas for Chinese infants as well as the nutritional diets of lactating mothers.

Developmental signals and environmental stresses regulate carbon distribution in the vegetative and reproductive organs of plants and affect seed yield. Cleistogenes songorica is a xerophytic grass with great potential application value in ecological restoration. However, how carbohydrate transport and distribution during grain filling affect the seed yield of C. songorica under water stress is not clear. The present study showed that the soluble sugar and starch contents of cleistogamous (CL) spikes and chasmogamous (CH) spikes were significantly higher at the milk stage, which was attributed to a significantly higher seed number and seed yield per spike under water stress conditions than under well-watered conditions (P < 0.01). RNA-seq data revealed a total of 54,525 differentially expressed genes (DEGs) under water stress conditions, but only 3744 DEGs were shared among all comparison groups. Weighted gene co-expression network analysis showed that the transport and distribution of carbohydrates were regulated by ABA-responsive genes (CsABA8OX1_1, CsABA8OX1_2, CsABA8OX2_1, CsABA8OX2_2, CsNCED3, CsNCED1_1, CsNCED1_2 and CsNCED4_1) and sugar transport and starch synthesis genes (CsSUS1, CsSUS2, CsSUS3, CsAGP1, CsAGP4, CsAGP5, CsSSS1 and CsSBE5) under water stress conditions. These genes jointly regulated carbohydrate remobilization in sources (stems, leaves and sheaths) to promote grain filling and improve seed yield. The present study helped to clarify the phenotypic, metabolic and transcriptional response mechanisms of vegetative organs, such as stems and leaves, and reproductive organs, such as CL spikes and CH spikes, to promote carbohydrate redistribution under water stress, and it provides theoretical guidance for improving seed yields.

Common vetch (Vicia sativa) is an important forage and green manure crop that is widely cultivated around the world. However, the large number of subspecies and accessions of common vetch has made taxonomic research on this species difficult. Pollen morphology data can provide important evidence in the study of plant phylogeny. Therefore, in this research, light microscopy and scanning electron microscopy were used to observe seven morphological traits of pollens from 22 common vetch accessions, and residual maximum likelihood and pattern analysis was conducted. The results showed that the pollen grains of the 22 accessions were all monad pollen and the polar view revealed three-lobed circular and triangular shapes, while the equatorial view mainly revealed an oblate shape; only one accession showed an oblate spherical shape. All accessions were 3-zonocolporate and the colpus length extended close to the poles. The polar axis length was (19.39 ± 0.97)–(42.12 ± 0.76) μm and the equatorial axis length was (35.97 ± 1.27)–(45.25 ± 0.81) μm. We found that qualitative traits of pollen shape, aperture polar view and ornamentation were highly stable. Among them, polar axis length, equatorial axis length and colpus length and width had significant differences among the accessions. The ratios of the equatorial and polar axes had significant differences among the subspecies. This trait information could be used for the classification and identification of common vetch accessions and subspecies.

Bile acids (BA) have emerged as signalling molecules regulating intestinal physiology. The importance of intestinal microbiota in production of secondary BA, for example, lithocholic acid (LCA) which impairs enterocyte proliferation and permeability, triggered us to determine the effects of oral probiotics on intestinal BA metabolism. Piglets were weaned at 28 d of age and allocated into control (CON, n 14) or probiotic (PRO, n 14) group fed 50 mg of Lactobacillus plantarum daily, and gut microbiota and BA profile were determined. To test the potential interaction of LCA with bacteria endotoxins in inducing damage of enterocytes, IPEC-J2 cells were treated with LCA, lipopolysaccharide (LPS) and LCA + LPS and expressions of genes related to inflammation, antioxidant capacity and nutrient transport were determined. Compared with the CON group, the PRO group showed lower total LCA level in the ileum and higher relative abundance of the Lactobacillus genus in faeces. In contrast, the relative abundances of Bacteroides, Clostridium_sensu_stricto_1, Parabacteroides and Ruminococcus_1, important bacteria genera in BA biotransformation, were all lower in the PRO than in the CON group. Moreover, PRO piglets had lower postprandial glucagon-like peptide-1 level, while higher glucose level than CON piglets. Co-administration of LPS and LCA led to down-regulated expression of glucose and peptide transporter genes in IPEC-J2 cells. Altogether, oral L. plantarum altered BA profile probably by modulating relative abundances of gut microbial genera that play key roles in BA metabolism and might consequently impact glucose homoeostasis. The detrimental effect of LCA on nutrient transport in enterocytes might be aggravated under LPS challenge.

Human milk fat is specially tailored to supply the developing infant with adequate and balanced nutrients. The present study aimed to quantify the composition of fatty acids, tocopherols and carotenoids in human milk, with special emphasis on the lactational changes. Colostrum, transitional and mature milk samples were collected longitudinally from the same forty-two healthy, well-nourished Chinese mothers. Fatty acids were quantified by GC with carotenoids (carotenes and xanthophylls) and tocopherols (α-, γ-tocopherol) determined by HPLC. Total fatty acid (TFA) content increased from 15·09 g/l in colostrum to 32·57 g/l in mature milk with the percentages of DHA and arachidonic acid (ARA) decreased. The ratio of n-6:n-3 PUFA and ARA:DHA remained constant during lactation at about 11:1 and 1·3:1, respectively. Both α-tocopherol and γ-tocopherol decreased over lactation with the ratio of α-:γ-tocopherol declined significantly from 7·21:1 to 4·21:1 (P < 0·001). Carotenoids all dropped from colostrum to mature milk as the less polar carotenes dropped by 88·67 %, while xanthophylls only dropped by 35·92 %. Lutein was predominated in both transitional and mature milk carotenoids (51·64–52·49 %), while colostrum carotenoids were mainly composed of lycopene (32·83 %) and β-carotene (30·78 %). The concentrations of tocopherols and xanthophylls but not carotenes were positively associated with TFA content in milk. These results suggested that colostrum and mature milk contained divergent lipid profiles and selective transfer mechanisms related to polarity might be involved. The present outcomes provide new insights for future breast-feeding studies, which also add in scientific evidences for the design of both initial and follow-on infant formulas.

Altered gut microbial ecology contributes to the development of metabolic diseases including obesity. However, studies based on different populations have generated conflicting results due to diet, environment, methodologies, etc. The aim of our study was to explore the association between gut microbiota and BMI in Chinese college students. The 16S next-generation sequencing (NGS) was used to test the gut microbiota of nine lean, nine overweight/obesity and ten normal-weight male college students. The differences in gut microbiota distribution among three groups were compared, and the relationship between the richness, diversity, composition of gut microbiota and BMI were analysed. The predominant phyla Bacteroidetes and Firmicutes were further confirmed by real-time PCR. Metagenomic biomarker discovery was conducted by linear discriminant analysis (LDA) effect size (LEfSe). NGS revealed that gut microbiota composition was different among three groups, but there was no difference in the abundance ratio of Firmicutes:Bacteroidetes. Several bacterial taxa were in linear relationship with BMI (positive relationship: uncultured bacterium (Bacteroides genus); negative relationship: Porphyromonadaceae, Acidaminococcaceae, Rikenellaceae, Desulfovibrionaceae, Blautia, Anaerotruncus, Parabacteroides, Alistipes). Moreover, gut microbiota diversity decreased with the increase in BMI. And LEfSe analysis indicated that Blautia, Anaerotruncus and its uncultured species were significantly enriched in the lean group (LDA score ≥ 3), Parasuterella and its uncultured species were significantly enriched in the overweight/obese groups (LDA score ≥ 3). In general, gut microbiota composition and microbial diversity were associated with BMI in Chinese male college students. Our results might enrich the understanding between gut microbiota and obesity.

In central Asia, Agriophyllum squarrosum is the first species to become established during natural succession on sand dunes. However, low germination percentages and thus poor stand establishment greatly inhibit the use of this key species in the stabilization of dunes. The aim of this review is to critically analyse published information on the seed biology of A. squarrosum with particular reference to identifying the factors limiting germination of seeds sown in the field. A conceptual model is used to illustrate the complexities of factors as well as the unknowns we found about the seed/seedling stage of the life cycle of this sand dune annual. A major result of this review is that we now know that high germination percentages can be obtained by storing freshly collected seeds dry at room temperatures for 2 to 3 months to allow dormancy break to occur via afterripening, and then storing them dry at low (e.g. 4–5°C) temperature to prevent them from entering secondary dormancy. Non-dormant seeds should be sown in the field in late spring when wind-blown sand will cover them, thus ensuring that they are in darkness, which promotes germination, at the time summer rains occur.

Medicago truncatula, soybean (Glycine max), Arabidopsis thaliana and rice (Oryza sativa) all belong to the core angiosperm group of plants. Seed-specific genes are important for seed formation and development in these angiosperms. The identification of genes specifically expressed in angiosperm seeds and the comparison of the expression patterns of homologous genes among different angiosperm species can provide novel insights into the functions of genes that control seed development and the evolution of angiosperms. We downloaded the sequences and expression data from the relevant databases, and the seed-specific expression of genes was identified with cut-offs of a gene expression level ratio ≥ 5 and a Z-score ≥ 6. The genes were analysed using local BLAST software with an E-value ≤ 1.0E − 505. A total of 605, 581, 778 and 722 genes showed specific expression in the seeds of Medicago, soybean, Arabidopsis and rice, respectively. Additionally, we compared the expression patterns of seed-specific genes from each species with their homologues in the other three species, and found that the degree of variation in the expression patterns of homologous genes was low among closely related species but higher among more distantly related ones. The discrepancy between the homologous gene expression patterns may be caused by the different characteristics of the cis-elements in the promoter regions of the homologous genes.

Although seed dormancy of temperate legumes is well understood, less is known about it in species that grow in subalpine/alpine areas. This study investigated dormancy and germination of four Vicia species from the Tibetan Plateau. Fresh seeds of V. sativa were permeable to water, whereas those of V. angustifolia, V. amoena and V. unijuga had physical dormancy (PY). One year of dry storage increased the proportion of impermeable seeds in V. angustifolia, but showed no effect on seed coat permeability in V. amoena or V. unijuga. Seeds of all four species also had non-deep physiological dormancy (PD), which was especially apparent in the two annuals at a high germination temperature (20°C). After 1 year of storage, PD had been lost. The hydrotime model showed that fresh seeds obtained a significantly higher median water potential [Ψb(50)] than stored seeds, implying that PD prevents germination in winter for seeds dispersed without PY when water availability is limited. After 6 months on the soil surface in the field, a high proportion of permeable seeds remained ungerminated, further suggesting that PD plays a key role in preventing germination after dispersal. Addition of fluridone, an inhibitor of abscisic acid (ABA) biosynthesis, evened-out the differences in germination between fresh and stored seeds, which points to the key role of ABA biosynthesis in maintaining dormancy. Further, fresh seeds were more sensitive to exogenous ABA than stored seeds, indicating that storage decreased embryo sensitivity to ABA. On the other hand, the gibberellic acid GA3 increased germination rate, which implies that embryo sensitivity to GA is also involved in seed dormancy regulation. This study showed that PY, PD or their combination (PY+PD) plays a key role in timing germination after dispersal, and that different intensities of dormancy occur among these four Vicia species from the Tibetan Plateau.

Lanthanum-modified lead titanate (PLT) ferroelectric thin films were fabricated by the RF magnetron sputtering system on Pt/Ti/SiO2/Si(100) substrates. The x-ray diffraction (XRD) patterns of the PLT films showed that the pure perovskite structure was formed in the PLT thin films. The Piezoresponse Force Microscopy (PFM) was used for determining the domain structure of these films. It was found that the 90 degree domain was the main domain structure of PLT thin films. It was found that the PLT films prepared by RF sputtering have relatively large pyroelectric coefficient γ=2.20×10-8C·(cm2·K)-1 and relatively high figures of merit for current responsivity, voltage responsivity and specific detectivity.
