The predominantly Afrotropical fruit fly genus Ceratitis contains many species of agricultural importance. Consequently, quarantine of Ceratitis species is a major concern for governmental regulatory agencies. Although diagnostic keys exist for identification of all described Ceratitis species, these tools are based on adult characters. Flies intercepted at ports of entry are usually immatures, and Ceratitis species cannot be diagnosed based on larval morphology. To facilitate identification of Ceratitis pests at ports of entry, this study explores the utility of DNA-based diagnostic tools for a select group of Ceratitis species and related tephritids, some of which infest agriculturally important crops in Africa. The application of the polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP) method to analyse three mitochondrial genes (12S ribosomal RNA, 16S ribosomal RNA, and NADH-dehydrogenase subunit 6) is sufficient to diagnose 25 species and two species clusters. PCR analysis of the internal transcribed spacer region 1 (ITS-1) is able to distinguish three of the five species left unresolved by mitochondrial DNA analysis.

Botanical preparations, usually from non-host plants, can be used to manipulate the behaviour of insect pests and their natural enemies. In this study, the effects of extracts of Chrysanthemum morifolium, a non-host plant of the diamondback moth, Plutella xylostella (Linnaeus), on the olfactory and oviposition responses of this phytophagous insect and on levels of parasitism by its specialist parasitoid Cotesia plutellae (Kurdjumov) were examined, using Chinese cabbage Brassica campestris L. ssp. pekinensis as the test host plant. Olfactometer tests showed that volatiles of chrysanthemum extract-treated host plants were less attractive to P. xylostella females than those from untreated host plants; and in contrast, volatiles of the chrysanthemum extract-treated host plants were more attractive to females of its parasitoid C. plutellae than those from untreated host plants. Oviposition preference tests showed that P. xylostella females laid only a small proportion of their eggs on chrysanthemum extract-treated host plants, while ovipositing parasitoid females parasitized a much higher proportion of host larvae feeding on the treated host plants than on untreated host plants. These results suggest that certain non-host plant compounds, when applied onto a host plant, may render the plant less attractive to a phytophagous insect but more attractive to its parasitoids. Application of such non-host plant compounds can be explored to develop push-pull systems to reduce oviposition by a pest insect and at the same time enhance parasitism by its parasitoids in crops.

Field infestation rates of an invasive fruit fly species, Bactrocera invadens Drew Tsuruta & White on mango was determined at different localities in Kenya. At most of the locations and especially at low elevations, B. invadens frequently shared the same fruit with the indigenous fruit fly species Ceratitis cosyra (Walker) but often occurred at higher numbers than C. cosyra. The level of infestation varied with location ranging from 3.0 to 97.2 flies per kg of fruit. There was a significant inverse relationship between numbers of flies per kg of fruit and elevation at which fruit was collected, suggesting that B. invadens is a predominantly lowland pest. On an artificial diet, development of B. invadens immatures lasted 25 days; egg incubation required 1.2 days, larval development 11.1 days and puparia-adult development 12.4 days. About 55% of eggs developed to the adult stage. Life expectancy at pupal eclosion was 75.1 days in females and 86.4 days in males. Average net fecundity and net fertility were 794.6 and 608.1 eggs, respectively, while average daily oviposition was 18.2 eggs. Daily population increase was 11% and mean generation time was 31 days. Results are discussed in relation to the biology and ecology of the insect and in the development of mass rearing and control measures for B. invadens.

In Zimbabwe, studies were made of the numbers of tsetse (Glossina spp.) and stable flies (Stomoxys spp.) attracted to cattle of different nutritional status, age and sex. Host odours were analysed to determine the physiological basis of these differences and improved methods are described for measuring rates of production of kairomones. Seasonal fluctuations in host weight, related to changes in pasture quality, had no significant effect on attraction of tsetse or Stomoxys. However, both attraction to different individuals and carbon dioxide production by these individuals were strongly correlated with weight, suggesting a possible link. Attraction to the odour from different types of cattle decreased in the order ox > cow > heifer > calf, and oxen were twice as attractive as calves of less than 12 months old. Lactation did not alter the relative attractiveness of cows. Calves less than six months old produced lower levels of carbon dioxide, acetone, octenol and phenols than oxen, but for older calves and cows, levels of production of known kairomones and repellents were similar to those of an ox. Carbon dioxide produced by cattle varied according to time of day and the animal's weight; cattle weighing 500 kg produced carbon dioxide at a mean rate of 2.0 l min−1 in the morning and 2.8 l min−1 in the afternoon compared to respective rates of 1.1 and 1.9 l min−1 for cattle weighing 250 kg. Artificially adjusting the doses of carbon dioxide produced by individual cattle to make them equivalent did not remove significant differences in attractiveness for tsetse but did for Stomoxys. Increasing the dose of carbon dioxide from 1 to 4 l min−1 in a synthetic blend of identified kairomones simulating those produced by a single ox, increased attractiveness to tsetse but not to the level of an ox. The results suggest that the main sources of differences in the attractiveness of individual cattle are likely to be variation in the production of carbon dioxide and, for tsetse, other unidentified kairomone(s). The biological and practical implications of these findings are discussed.

Aphis gossypii Glover is a polyphagous aphid pest with a worldwide distribution. However, there is evidence that on a global scale the name A. gossypii is being applied to a number of forms with different life cycles and/or host-plant associations. Morphometric variation of A. gossypii samples from crops and non-cultivated plants in many parts of the world was examined, to determine whether this variation is correlated with the hosts from which the aphids originated. Samples of A. gossypii were collected from Cucurbitaceae and Malvaceae in Europe, and from Compositae in various parts of the world. Morphometric data for 13 parameters measured from 97 clonal lineages (728 specimens) and 27 field-collected samples (313 specimens) were analysed by a series of canonical variates analyses, using the field sample/clonal lineage as grouping factor. Clonal lineages were reared on a common host in controlled conditions to standardize the effect of host and environment on morphology. The analyses provided a clear morphometric separation of the aphids originating from Compositae and those collected on Cucurbitaceae and Malvaceae, regardless of the geographical origin of the aphids and the host plant on which they were reared. This indicates that within A. gossypii there are two widely distributed host races or subspecies with different plant family associations. The taxonomic implications are discussed.

The randomly amplified polymorphic DNA technique was used to trace the geographic origin of Liposcelis bostrychophila Badonnel populations in Australia from unknown geographic sources internationally. Haplotype (or clonal) diversity was high, with 474 unique haplotypes found from 616 individuals genotyped. Gene diversity estimates (0.10–0.28) and percent polymorphic loci (38.1–88.1%) were moderate to high for most populations. This resulted in genetic distance estimates that ranged from 0.04 to 0.26 and were significantly different for most pairwise population combinations. GST values for all populations were also moderate (0.04–0.54) and again were significantly different for most pairwise population comparisons. Analysis of molecular variance revealed that the majority of variation was apportioned among individuals within populations regardless of the level at which they were grouped. Gene flow (Nm) was mostly low for all pairwise populations comparisons with an average Nm = 1.8. A non-significant negative correlation between genetic distance and geographic distance was found for worldwide populations. In contrast, within Australian populations a significant positive correlation between genetic distance and geographic distance was detected. Genetic relationships explored using unweighted pair group method analysis and non-metric multidimensional scaling indicated a mixed pattern of genetic similarities among all populations. Multiple introductions, from a wide range of international source populations, have obscured the ability to accurately determine the geographic origin of L. bostrychophila in Australia.

Two forms of Myzus persicae (Sulzer), F-15 and R306N′, resembling M. antirrhinii (Macchiati) in esterase profile have been found in Japan. To determine the genetic relationship of F-15 and R306N′ with M. antirrhinii, clonal lineages of F-15, R306N′ and those of 14 other forms of M. persicae collected in Japan were analysed for 17 polymorphic microsatellite DNA loci. Two microsatellite multi-locus (MS) genotypes of M. persicae and two MS genotypes, Ma3 and Ma4, of M. antirrhinii from Australia were used as references. The 16 Japanese forms were represented by different MS genotypes. F-15 and R306N′ had, respectively, 44.1% and 35.3% of their alleles at the 17 loci in common with Ma3 and Ma4, and some of the common alleles were peculiar in size. F-15 and R306N′ shared 22.6% and 29.6%, respectively, of their alleles with the other 16 MS genotypes of M. persicae. The corresponding values for Ma3 and Ma4 were both 27.4%. F-15 and R306N′ were grouped reliably with the two Australian MS genotypes of M. antirrhinii both by neighbour-joining method based on shared allele distance and by a Bayesian method with Markov Chain Monte Carlo algorithm. These results suggest that F-15 and R306N′ are genetically closely related to M. antirrhinii and isolated from the gene pool of M. persicae despite their ability to produce sexual morphs. It is therefore proposed that F-15 and R306N′ should be classified as M. antirrhinii.

Fruit flies (Diptera: Tephritidae) are a species-rich and economically important group. The phylogenetic relationships among the many taxa are still to be fully resolved and the monophyly of several groups is still to be confirmed. This paper reports a study of the phylogenetic relationships among 23 economically important tephritid species (representing several major lineages of the family) which examines the sequence of a region of mitochondrial DNA encompassing the cytb, tRNASer and ND1 genes. Substitutions characteristic of particular taxa were found that could help classify members of the family at any developmental stage. The trees obtained by the maximum parsimony, neighbour joining and maximum likelihood methods were generally compatible with present morphological classification patterns. However, the data reveal some characteristics of the phylogenetic relationships of this family that do not agree with present classifications. The results support the probable non-monophyletic nature of the subfamily Trypetinae and suggest that Bactrocera cucurbitae (Coquillet) is more closely related to the genus Dacus than to other species of Bactrocera.

The pine sawyer Monochamus galloprovincialis (Olivier) is the vector of the introduced pine wood nematode Bursaphelenchus xylophilus (Steiner & Bührer) Nickle in Portugal, and until recently was considered a secondary forest insect. Under laboratory conditions, a study of biological and reproductive traits of 37 insect pairs was conducted. The longevity of both sexes was similar, being 61.2±6.5 days for males and 64.0±6.3 days for females (mean±SE). Sixteen small-sized insects (22% of the population) died within 20 days and before starting to reproduce. The sexual maturation period (without egg laying) was 20.4±0.7 days (mean±SE), while the oviposition period lasted 54.0±4.2 days (mean±SE). The oviposition rate increased very quickly during the first weeks of life, peaking to almost two eggs per day during days 30–44, and gradually dropping in the following weeks. The females laid an average of 67.0±5.96 (mean±SE) eggs through their lives. The hatch rate was 92.6±1.0%; (mean±SE). There were large individual variations in longevity and fecundity parameters, and principal component analysis based on 16 morphological and biological parameters separated the breeding insects into four distinct groups. Almost half of the reproducing beetles were large-sized insects, with high longevity and fecundity traits. Overall, the reproductive potential of the Portuguese population of M. galloprovincialis seems to be smaller than that described for other Monochamus vectors of the pine wood nematode both in North America and Japan.

The Chinese citrus fruit fly, Bactrocera (Tetradacus) minax (Enderlein), is one of the major citrus pests in Bhutan and can cause >50% mandarin (Citrus reticulata Blanco) fruit drop. As part of the development of a management strategy for the fly in mandarin orchards, population monitoring and experimental manipulations were carried out to determine: (i) adult emergence period; (ii) adult phenology patterns; (iii) period of crop susceptibility; and (iv) period from fruit drop to pupation. In western Bhutan, adult flies emerge from the overwintering pupal stage in late April/early May. Most flies are mature by the end of May and it is inferred that mating occurs at this time: from the beginning of June males rapidly disappear from the population and by mid- to late June are rare or absent from traps. Mature females are present in the mandarin crop at the beginning of June, but very little oviposition occurs until mid-June, while most damage has occurred by mid-July. Initiation of oviposition into mandarins is almost certainly linked to crop phenology. Adult flies disappear from the orchard system during August. After fruit drop, larvae were recorded leaving the fruit to pupate within 13 days. The use of early to mid-season protein bait sprays and/or targeted use of systemic insecticides during the one month oviposition period, plus the removal of fallen fruit once every 10 days, are recommended as control strategies.

The performance of Nzi traps for tabanids (Tabanus similis Macquart, T. quinquevittatus Wiedemann, Chrysops aberrans Philip, C. univittatus Macquart, C. cincticornis Walker, Hybomitra lasiophthalma (Macquart)), stable flies (Stomoxys calcitrans Linnaeus) (Diptera: Muscidae) and mosquitoes (Aedes) (Diptera: Culicidae) was investigated at various sites in Canada (Ontario, Alberta) and USA (Iowa, Florida, Louisiana). Traps made from selected fabrics, insect nettings and hand-dyed blue cotton were compared to the African design to provide practical recommendations for temperate environments. Comparisons of substituted materials showed that trap performance was optimal only when traps were made from appropriate fabrics in the colours produced by either copper phthalocyanine (phthalogen blue), or its sulphonated forms (turquoise). Fabrics dyed with other blue chromophores were not as effective (anthraquinone, disazo, formazan, indanthrone, triphenodioxazine). An appropriate texture as well as an appropriate colour was critical for optimal performance. Smooth, shiny synthetic fabrics (polyester, nylon) and polyester blends reduced catches. Low catches occurred even for nominal phthalogen blue, but slightly-shiny, polyester fabrics in widespread use for tsetse. The most suitable retail fabric in place of phthalogen blue cotton was Sunbrella Pacific Blue acrylic awning/marine fabric. It was both attractive and durable, and had a matching colour-fast black. Nzi traps caught grossly similar numbers of biting flies as canopy, Vavoua, and Alsynite cylinder traps, but with differences in relative performance among species or locations.

Digestion in the larger black flour beetle, Cynaeus angustus (LeConte), was studied to identify new control methods for this pest of stored grains and grain products. The physiological pH of the larval gut, as measured with extracts in water, was approximately 6.1, and the pH for optimal hydrolysis of casein by gut extracts was 6.2 when buffers were reducing. However, under non-reducing conditions, hydrolysis of casein and synthetic serine proteinase substrates was optimal in alkaline buffer. Three major proteinase activities were observed in zymograms using casein or gelatin. Caseinolytic activity of C. angustus gut extracts was inhibited by inhibitors that target aspartic and serine proteinase classes, with minor inhibition by a cysteine proteinase inhibitor. In particular, soybean trypsin and trypsin/chymotrypsin inhibitors were most effective in reducing the in vitro caseinolytic activity of gut extracts. Based on these data, further studies are suggested on the effects of dietary soybean inhibitors of serine proteinases, singly and in combination with aspartic and cysteine proteinase inhibitors, on C. angustus larvae. Results from these studies can be used to develop new control strategies to prevent damage to grains and stored products by C. angustus and similar coleopteran pests.

The incubation period of Spodoptera exigua (Hübner) was not influenced by the host plant, whereas larval development time and pupal period were affected. Larval development time was longest on shallot and lady's finger, followed by cabbage and long bean. Larvae did not develop beyond the first instar when fed on chilli. The pupal period was longer on lady's finger than on cabbage, shallot and long bean. Overall, adult longevity was not influenced by the host plant but there was a difference between female and male longevity among the host plants. Survival of S. exigua was affected by the host plant at the larval stage. The number of larval instars varied between 5 and 8 within and between the studied host plants. Long bean was found to be the most suitable host plant and provide the best food quality for S. exigua compared to the other host plants, as it allowed faster development, fewer larval instars and a higher survival rate.

The efficacy of baiting as a pest control method relies on the bait appealing to the pest species. In the case of wood-eating termites, bait stations should be designed to encourage termite presence and to maximize their consumption of bait matrix in order to expedite control in minimal time. A field experiment examined the effect of bait size (one large bait or four small baits of equivalent total size, with commensurate inspection and replacement schedules), compaction (tightly rolled or loosely folded) and composition (paper only or paper plus wood) on termite presence and on untreated bait paper removal rates over four months. All three factors were significant, with bait size the most important factor, followed by compaction and then composition. The least effective baits were small, compacted (rolled) paper-only baits with monthly inspections; these had the highest abandonment rate (70%) and had the least paper removed (mean of 24 g). The most effective baits were large, folded paper-plus-wood baits with inspections at two months; these had the lowest abandonment rate (20%) and had the highest paper removal (mean of 112 g). The more than four-fold difference between these baits types demonstrates that bait efficacy can be altered considerably merely by changing bait design without adding new ingredients to the bait matrix.

The debate about mechanisms underlying the evolution of host specialization by herbivorous insects remains open. Natural selection may act locally and lead to different patterns of geographic variation in life history traits of polyphagous herbivores. The hypothesis of genetically-based trade-offs in offspring performance on different hosts has been proposed but this has rarely been demonstrated. Under laboratory conditions, the biological performance of two populations of the hemlock looper Lambdina fiscellaria (Guenée), a highly polyphagous lepidopteran, was compared when reared on three different tree host species: balsam fir, eastern hemlock and sugar maple. One population originated from Anticosti Island, Québec, Canada, where the insect has evolved without having access to two of the three tree species tested, the other being from the mainland where all tree species are present. When reared on balsam fir foliage, which was naturally available to each population, larvae from Anticosti Island underwent four instars compared with five for the mainland population, indicating the existence of geographic biotypes in L. fiscellaria. When reared on the foliage of non-naturally available host trees, larvae from Anticosti Island had a higher incidence of supernumerary instars. This is a unique example where local adaptation to environmental conditions of an insect herbivore is expressed through a differential number of larval instars. Moreover, the Anticosti Island population showed a higher growth related index on the host available to both populations indicating that a fitness trade-off was the evolutionary process underlying the local adaptation of this population on balsam fir.

Various physical and chemical properties of host plants influence insect larval performance and subsequent adult fitness. Tomato plants are relatively new hosts to the potato tuber moth, Phthorimaea operculella (Zeller), with the fruit being its preferred feeding site. However, it is unclear how the biochemical and physical properties of tomato fruits relate to potato tuber moth performance. Significant amounts of α-tomatine were detected in maturing green and ripening fruits of cherry (cv. Ceres) and processing (cv. Serio) types of tomatoes whereas none was detected in a fresh market variety (cv. Marglobe), at comparable stages. α-Tomatine is negatively and significantly correlated with development rate (head capsule size) of larvae reared in the fruits of the cherry and processing type tomatoes. Generally, survival, growth and development were significantly superior for larvae reared in the ripening fruits of the fresh market cultivar. At this stage, the fruits of this cultivar are also the largest. Based on these results it is concluded that fruit α-tomatine content, as wellas fruit size and maturity, all affect performance of P. operculella larvae in the fruits of cultivated tomatoes.

The utility of temperature gradient gel electrophoresis (TGGE) as a means of analysing the gut contents of predators was evaluated. Generalist predators consume multiple prey species and a species-specific primer approach may not always be a practical means of analysing predator responses to prey diversity in complex and biodiverse ecosystems. General invertebrate primers were used to amplify the gut contents of predators, generating banding patterns that identified component prey remains. There was no evidence of dominance of the polymerase chain reaction (PCR) by predator DNA. When applied to field samples of the carabid predator Pterostichus melanarius (Illiger) nine banding patterns were detected, including one for aphids. To further distinguish between species, group-specific primers were designed to separate species of earthworm and aphid. TGGE of the earthworm PCR products generated banding patterns that varied with haplotype in some species. Aphid and earthworm DNA could be detected in the guts of carabids for up to 24 h using TGGE. In P. melanarius, with low numbers of prey per insect gut (mean < 3), interpretation of banding patterns proved to be tractable. Potential problems of interpretation of TGGE gels caused by multiple prey bands, cryptic bands, haplotype variation, taxonomic uncertainties (especially with regard to earthworms), secondary predation, scavenging and presence of parasites and parasitoids in the prey or the predators, are discussed. The results suggest that PCR, using combinations of general invertebrate and group-specific primers followed by TGGE, provides a potentially useful approach to the analysis of multiple uncharacterized prey in predators.

Protease inhibitors cause mortality in a range of insects, and transgenic plants expressing protease inhibitors have been protected against pest attack, particularly internal feeders that are not amenable to control by conventional means. A study of luminal proteases in Conogethes punctiferalis Guenée was performed to identify potential targets for proteinaceous biopesticides, such as protease inhibitors. The midgut protease profile of the gut lumen from C. punctiferalis was studied to determine the conditions for optimal protein hydrolysis. Optimum conditions for peptidase activity were found to be in 50 mm Tris-HCl, pH 10 containing 20 mm CaCl2; incubation for 30 min at 40°C. Four synthetic substrates, i.e. benzoyl-arg-p-nitroanilide, benzoyl-tyr-p-nitroanilide, succinyl-ala-ala-pro-leu-p-nitroanilide (SAAPLpNA) and leu-p-nitroanilide were hydrolysed by C. punctiferalis gut proteases in Tris-HCl buffer pH 10. Trypsin and elastase-like chymotrypsin were the prominent digestive proteases, and age-related modulation of midgut proteases existed for trypsin, chymotrypsin, elastase-like chymotrypsin and leucine aminopeptidase. Serine protease inhibitors such as aprotinin, soybean trypsin inhibitor and phenylmethanesulfonyl fluoride inhibited peptidase activity. Some metal ions such as Ca2+, Mg2+, Pb2+ and Co2+ enhanced BApNA-ase activity whereas others like Mn2+, Zn2+, Cu2+, Fe2+ and Hg2+ were inhibitory at 6 mm concentration. Trypsin and elastase-like chymotrypsin were significantly inhibited by 94% and 29%, respectively, by aprotinin (150 nm) under in vitro conditions. A possible incorporation of protease inhibitors into transgenic plants is discussed.

Establishment of non-native species and human-driven alteration of habitats are major causes of the decline of native faunas. Interference between native and non-native cockroach species was evaluated here by comparing their distribution areas in two oceanic islands: Mayotte and La Réunion. A factorial correspondence analysis showed that their presence is related to environmental parameters among which human activity, humidity and altitude are the three most important. Human activity and humidity favour establishment and development of non-native species, whereas altitude limits their presence. Non-native species form guilds (up to seven species). Their distribution areas overlap and illustrate their colonizing capacities. On the contrary, distribution areas of native species never overlap, although some co-occur with non-native species. Native species are more endangered when they are established in cultivated areas than when they are in protected natural areas.

The life table statistics of six native Kenyan species/strains of Trichogramma and Trichogrammatoidea were established using a factitious host Corcyra cephalonica, Stainton (Lepidoptera: Pyralidae), at eight different temperatures (10, 15, 20, 25, 28, 30, 32 and 35°C) and two humidity levels (40–50 and 70–80%). The objective was to select insects with superior attributes for augmentative release against lepidopteran pests in horticultural crops. Both temperature and humidity affected developmental time and life table parameters of the parasitoids but temperature played a more critical role. Developmental time was inversely related to temperature. The intrinsic and finite rates of increase increased with temperature up to 30°C. Both net reproduction rate and intrinsic rate of increase were higher at the lower humidity. Temperature inversely affected generation time of parasitoid strains regardless of the relative humidity. Two strains of Trichogramma sp. nr. mwanzai collected from both low and medium altitudes and Trichogrammatoidea sp. nr. lutea from the mid-altitudes, were better adapted to both low and high temperatures than the other strains, as indicated by the high intrinsic and net reproductive rates, at both humidity levels. These three strains appear to be promising candidates for augmentation biocontrol against the African bollworm Helicoverpa armigera in Kenya.
