OBJECTIVES/SPECIFIC AIMS: Deep brain stimulation is currently being evaluated as an experimental therapy for various psychiatric disorders, as well as being investigated as a method for mapping emotional brain functions. This growing area of research requires sensitive measures to quantify effects of stimulation on emotional processing. The current study examined the effects of acute stimulation to 2 limbic regions—the subcallosal cingulate (SCC) and the amygdala—on bias in the perception and evaluation of emotional facial expressions. We hypothesized that transient electrical stimulation to the limbic system would produce acute reductions in negative bias, consistent with its antidepressant effects in patients with severe depression. METHODS/STUDY POPULATION: The current study uses a novel affective bias task, developed to rapidly and covertly quantify emotional state. Over 4–6 minutes, patients rate the intensity and valence of static images of emotional facial expressions. We examined effects of electrical brain stimulation in 2 groups: patients with treatment-refractory depression undergoing SCC DBS therapy, and epilepsy patients undergoing amygdala stimulation via stereo-EEG electrodes during inpatient intracranial monitoring. DBS patients completed the task under stimulation and sham conditions during monthly visits over the first 6 months of therapy, as well as daily during a 1 week, blinded period of DBS discontinuation at the 6-month time point. Epilepsy patients completed the task under stimulation and sham conditions at a single visit. Mixed linear models and paired-samples t-test were used to investigate effects of stimulation as well as depression scale scores on affective bias ratings. RESULTS/ANTICIPATED RESULTS: Four SCC DBS patients showed significant effects of stimulation (p<0.0001) and depressive state (p<0.0001) on affective bias scores across 6 months of chronic DBS therapy, where emotional faces were perceived as less sad with stimulation ON, as well as during visits in which patients were nondepressed (typically later in the treatment course). Furthermore, 2 DBS patients showed rapid negative shifts in bias following acute blinded discontinuation of chronic stimulation, an effect which persisted over the 1-week period of discontinuation (t29=−2.58, p=0.015), in the absence of any self-reported change in mood. Likewise, 6 epilepsy patients showed significant positive shifts in affective bias with acute amygdala stimulation (t5=−4.75, p=0.005). Current analyses are investigating electrophysiological, autonomic and facial motor correlates to affective bias in these patients. DISCUSSION/SIGNIFICANCE OF IMPACT: Affective bias has revealed rapid, significant changes with stimulation at 2 limbic targets—one a white matter hub and one a nuclear subcortical structure—suggesting the task’s utility as an emotional outcome measure in brain stimulation studies. These stimulation-sensitive measures may provide a new metric to track treatment response to deep brain stimulation therapy for affective disorders. Future studies will determine whether affective bias can predict neuropsychiatric complications in patients undergoing stimulation mapping of brain circuitry ahead of resection surgery for epilepsy.

Tomography produces complex volumetric datasets containing the entire internal structure and density of an object in three dimensions (3D). Interpreting volumetric data requires 3D visualization but needs specialized software distinguishable from more familiar tools used in animation for 3D surface data. This tutorial reviews 3D visualization techniques for volumetric data using the open-source tomviz software package. A suite of tools including two-dimensional (2D) slices, surface contours, and full volume rendering provide quantitative and qualitative analysis of volumetric information. The principles outlined here are applicable to a wide range of 3D tomography techniques and can be applied to volumetric datasets beyond materials characterization.

Field and greenhouse studies were conducted to evaluate mesotrione alone and in combinations with low rates of atrazine and bentazon for control of yellow and purple nutsedge. Mesotrione alone at rates of 105 to 210 g ai/ha controlled yellow nutsedge 43 to 70%. Mixtures of mesotrione with atrazine at 280 g ai/ha did not always improve yellow nutsedge control over that by mesotrione alone, but increasing atrazine to 560 g ai/ha in these mixtures generally provided more consistent control of yellow nutsedge. Mesotrione at 105 g ai/ha mixed with bentazon at 280 or 560 g ai/ha controlled yellow nutsedge 88% or greater which was similar to control from the standard halosulfuron at 36 g ai/ha. Mesotrione, atrazine, and bentazon alone did not control purple nutsedge. Mixtures of mesotrione plus bentazon, however, did improve control of purple nutsedge over either herbicide applied alone, but this control was not considered commercially acceptable.

Studies were conducted in 2000 and 2001 to investigate responses of glyphosate-resistant cotton, glyphosate-resistant soybean, and selected weed species to postemergence applications of isopropylamine (Ipa) and diammonium (Dia) salts of glyphosate at selected rates ranging from 0.42 to 3.36 kg ae/ha. No differences were detected between either glyphosate salts or application timings for cotton injury, cotton lint yield, micronaire, fiber length, fiber strength, or fiber uniformity. In a weed-free soybean study, no differences in soybean injury occurred between early-postemergence treatments of the two glyphosate salts. Injury from late-postemergence treatments did not exceed 12% with glyphosate-Ipa or 9% with glyphosate-Dia at 3.36 kg/ha. Soybean treated with glyphosate-Ipa yielded 3,050 kg/ha, whereas soybean treated with glyphosate-Dia yielded 2,880 kg/ha, when averaged over glyphosate rate and application timing. In a soybean study that included weed control as a variable, weed control at 14 d after treatment (DAT), and soybean yield was independent of glyphosate salt. Control of common ragweed, ivyleaf morningglory, pitted morningglory, and large crabgrass at 28 DAT was similar at 0.84 kg/ha of either glyphosate salt.

Contributors to this Special Issue of the Cognitive Behaviour Therapist have considered the kind of infrastructure that should be in place to best support and guide CBT supervisors, providing practical advice and extensive procedural guidance. Here we briefly summarize and discuss in turn the 10 papers within this Special Issue, including suggestions for further enhancements. The first paper, by Milne and Reiser, conceptualized this infrastructure in terms of an ‘SOS’ (supporting our supervisors) framework, from identifying supervision competencies, to training, evaluation and feedback strategies. The next nine papers illustrate this framework with specific technical innovations, educational enhancements and procedural issues, or through comprehensive quality improvement systems, all designed to support supervisors. These papers suggest an assortment of workable infrastructure developments: two large-scale and comprehensive initiatives, some promising proposals and technologies, and a series of local, exploratory work. Collectively, they provide us with models for further developing evidence-based cognitive-behavioural supervision, and offer practical suggestions for giving supervisors the tools and support to maximize their supervisees’ learning, and to improve the associated client outcomes. Much research and development work remains to be done, and successful implementation will require institutional and political support, as well as cross-cultural adaptations. We conclude with an optimistic assessment of progress toward addressing some of the infrastructure improvements required to adequately support supervisors.

For exploring the prospect of higher-k dielectric phase engineering on a high mobility substrate, films of Hf1-xZrxO2 with varying x-values (0 ≤ x ≤ 1) were deposited on Al2O3 passivated Ge substrates using atomic layer deposition (ALD) with a cyclic deposit-anneal-deposit-anneal (DADA) scheme. The evolution of monoclinic to higher-k tetragonal structure with increasing ZrO2 concentration was probed by grazing incident x-ray diffraction and partial reciprocal space maps using the highly brilliant synchrotron x-ray source at the Cornell High Energy Synchrotron Source (CHESS). A primarily amorphous/nano-crystalline matrix of the asdeposited films changed to randomly aligned grains of nanocrystalline MO2 (M=Hf, Zr) after post deposition annealing at 800 °C for 200 seconds. In contrast, the DADA films annealed for same thermal budget showed high degree of preferred orientation along certain crystallographic directions. With increasing ZrO2 content, the structure of the films changed from a monoclinic to a tetragonal phase. A lower amount of ZrO2 (x = 0.33) was required for stabilizing the tetragonal phase in films grown on Al2O3 passivated Ge substrate as compared to similar films grown on a Si substrate via the same DADA process (x ≥ 0.50).
