Sulla eredopatologia delle mesenchimopatie reattive (cosidette malattie del collageno). II - Ricerche sui fattori ereditari del reumatismo cronico primario

This study is based on pedigrees of 192 patients with rheumatoid arthritis which are compared with pedigrees of 300 controls.

6,3 per cent of all relatives in the first group had rheumatoid arthritis compared to 0,38 per cent of all relatives in the control group. An hereditary predisposition to rheumatoid arthritis is definitevely to be admitted.

This predisposition is due to the presence of a principal autosomal dominant gene with reduced penetrance and variable expressivity. It seems to have no genetic relationship to the diathesis of allergic diseases and the diathesis of degenerative connective tissue disorders (mesenchymotic diathesis).

RIASSUNTO

Dall'esame di 192 famiglie con probando affetto da reumatismo cronico primario e di 300 famiglie di controllo, gli AA. concludono per l'esistenza di una predisposizione genetica al reumatismo cronico primario. Su di un totale di 1744 membri delle 192 famiglie reumatiche, esclusi i probandi, si osserva una incidenza di RCP pari al 6,3%, mentre nelle famiglie di controllo tale incidenza scende allo 0,38%.

La predisposizione al reumatismo cronico primario è condizionata alla presenza di un gene principale autosomico a carattere dominante, con penetranza ridotta ed espressività variabile.

La sua penetranza e la sua espressività sotto forma di reumatismo cronico primario sono in funzione della presenza di altri fattori genotipici e paratipici.

La predisposizione al RCP non ha alcun rapporto genetico con la diatesi allergica e con la diatesi mesenchimosica, dimostrando così un comportamento analogo alla predisposizione al reumatismo acuto primario.

RÉSUMÉ

Après l'examen de 192 familles avec un sujet affecté de polyarthrite chronique évolutive (rhumatisme chronique primaire) et de 300 familles de contrôle, les AA. concluent pour l'existen, ce d'une prédisposition héréditaire à la polyarthrite chronique évolutive.

Cette prédisposition est liée à la présence d'un géne principal autosomique, dominant avec defaut de pénétrance et avec une expressivité variable.

La prédisposition à la p. c. e. ne semble avoir aucune relation génétique avec la diathèse allergique et avec la diathèse mesenchymotique.

ZUSAMMENFASSUNG

Die V. auf Grund von 192 Familien der Probanden mit chronischen primären Gelenkrheumatismus, und von 300 Kontroll familien, beweisen das Bestehen einer erbbedingten « Prädisposition zum chronischen primären Gelenkrheumatismus. Diese Prädisposition ist abhänging von der Anwesenheit eines autosomischen dominanten Hauptgen, mit verkleinert Penetranz und veränderlicher Expressivtät. Die Prädisposition zum akuten Gelenkrheumatismus scheint keine genetische Beziehung mit allergischer und mesenchymotischer Diathese zu haben.
