Direct numerical simulations (DNSs) are performed to investigate the roughness effects on the statistical properties and the large-scale coherent structures in the turbulent channel flow over three-dimensional sinusoidal rough walls. The outer-layer similarities of mean streamwise velocity and Reynolds stresses are examined by systematically varying the roughness Reynolds number $k^{+}$ and the ratio of the roughness height to the half-channel height $k / \delta$. The energy transfer mechanism of turbulent motions in the presence of roughness elements with different sizes is explored through spectral analysis of the transport equation of the two-point velocity correlation and the scale-energy path display of the generalized Kolmogorov equation. The results show that, with increasing $k^+$, the downward shift of the mean streamwise velocity profile in the logarithmic region increases and the peak intensities of turbulent Reynolds stresses decrease. At an intermediate Reynolds number ($Re_{\tau }= 1080$), the length scale and intensity of the large-scale coherent structures increase for a small roughness ($k^{+}=10$), which leads to failure of the outer-layer similarity in rough-wall turbulence, and decrease for a large roughness ($k^{+}=60$), as compared with the smooth-wall case. The existence of the small roughness ($k^{+}=10$) enhances the mechanism of inverse energy cascade from the inner-layer small-scale structures to the outer-layer large-scale structures. Correspondingly, the self-sustaining processes of the outer-layer large-scale coherent structures, including turbulent production, interscale transport, pressure transport and spatial turbulent transport, are all enhanced, whereas the large roughness ($k^{+}=60$) weakens the energy transfer between the inner and outer regions.

Elastocapillarity has attracted increasing interest in recent years due to its important roles in many industrial applications. In this work, we derive a thermodynamically consistent continuum model for the dynamics of two immiscible fluids on a thin and inextensible elastic sheet in two dimensions. With the sheet being modelled by a deformable curve with the Wilmore energy and local inextensibility constraint, we derive a two-phase hydrodynamics model with the interfacial and boundary conditions consistent with the second law of thermodynamics. In particular, the boundary conditions on the sheet and at the moving contact line take the form of force balances involving the fluid stress, surface tensions, the sheet bending force and sheet tension, as well as friction forces arising from the slip of fluids on the sheet. The resulting model obeys an energy dissipation law. To demonstrate its capability of modelling complex elastocapillary interactions, we consider two applications: (1) the relaxation dynamics of a droplet on an elastic sheet and (2) the transport of a droplet driven by bendotaxis in a channel bounded by elastic sheets. Numerical solutions for the coupled fluid–sheet dynamics are obtained using the finite element method. The detailed information provided by the full hydrodynamics model allows us to better understand the dynamical processes as compared to other simplified models that were used in previous work.

The low maturation rate of oocytes is an important reason for female infertility and failure of assisted pregnancy. The germinal vesicle breakdown (GVBD) is a landmark event of oocyte maturation. In our previous studies, we found that zona pellucida 3 (ZP3) was strongly concentrated in the nuclear region of germinal vesicle (GV) oocytes and interacted with aryl hydrocarbon receptor-interacting protein-like 1 (AIPL1) and lamin A to promote GVBD. In the current study, we found that lamin A is mainly concentrated in the nuclear membrane. When ZP3 is knocked down, lamin A will be partially transferred to the nucleus of oocytes. The prelamin A is increased in both the nuclear membrane and nucleus, while phosphorylated lamin A (p-lamin A) is significantly reduced. AIPL1 was also proved to accumulate in the GV region of oocytes, and ZP3 deletion can significantly inhibit the aggregation of AIPL1 in the nuclear region. Similar to ZP3 knockdown, the absence of AIPL1 resulted in a decrease in the occurrence of GVBD, an increase in the amount of prelamin A, and a significant decrease in p-lamin A in oocytes developed in vitro. Finally, we propose the hypothesis that ZP3 can stabilize farnesylated prelamin A on the nuclear membrane of AIPL1, and promote its further processing into mature lamin A, therefore promoting the occurrence of GVBD. This study may be an important supplement for the mechanism of oocyte meiotic resumption and provide new diagnostic targets and treatment clues for infertility patients with oocyte maturation disorder.

Hormone-sensitive lipase (HSL) is one of the rate-determining enzymes in the hydrolysis of TAG, playing a crucial role in lipid metabolism. However, the role of HSL-mediated lipolysis in systemic nutrient homoeostasis has not been intensively understood. Therefore, we used CRISPR/Cas9 technique and Hsl inhibitor (HSL-IN-1) to establish hsla-deficient (hsla-/-) and Hsl-inhibited zebrafish models, respectively. As a result, the hsla-/- zebrafish showed retarded growth and reduced oxygen consumption rate, accompanied with higher mRNA expression of the genes related to inflammation and apoptosis in liver and muscle. Furthermore, hsla-/- and HSL-IN-1-treated zebrafish both exhibited severe fat deposition, whereas their expressions of the genes related to lipolysis and fatty acid oxidation were markedly reduced. The TLC results also showed that the dysfunction of Hsl changed the whole-body lipid profile, including increasing the content of TG and decreasing the proportion of phospholipids. In addition, the systemic metabolic pattern was remodelled in hsla-/- and HSL-IN-1-treated zebrafish. The dysfunction of Hsl lowered the glycogen content in liver and muscle and enhanced the utilisation of glucose plus the expressions of glucose transporter and glycolysis genes. Besides, the whole-body protein content had significantly decreased in the hsla-/- and HSL-IN-1-treated zebrafish, accompanied with the lower activation of the mTOR pathway and enhanced protein and amino acid catabolism. Taken together, Hsl plays an essential role in energy homoeostasis, and its dysfunction would cause the disturbance of lipid catabolism but enhanced breakdown of glycogen and protein for energy compensation.

Microtubule-severing proteins (MTSPs) play important roles in mitosis and interphase. However, to the best of our knowledge, no previous studies have evaluated the role of MTSPs in female meiosis in mammals. It was found that FIGNL1, a member of MTSPs, was predominantly expressed in mouse oocytes and distributed at the spindle poles during meiosis in the present study. FIGNL1 was co-localized and interacted with γ-tubulin, an important component of the microtubule tissue centre (MTOC). Fignl1 knockdown by specific small interfering RNA caused spindle defects characterized by an abnormal length:width ratio and decreased microtubule density, which consequently led to aberrant chromosome arrangement, oocyte maturation and fertilization obstacles. In conclusion, the present results suggested that FIGNL1 may be an essential factor in oocyte maturation by influencing the meiosis process via the formation of spindles.

Microtubule-severing protein (MTSP) is critical for the survival of both mitotic and postmitotic cells. However, the study of MTSP during meiosis of mammalian oocytes has not been reported. We found that spastin, a member of the MTSP family, was highly expressed in oocytes and aggregated in spindle microtubules. After knocking down spastin by specific siRNA, the spindle microtubule density of meiotic oocytes decreased significantly. When the oocytes were cultured in vitro, the oocytes lacking spastin showed an obvious maturation disorder. Considering the microtubule-severing activity of spastin, we speculate that spastin on spindles may increase the number of microtubule broken ends by severing the microtubules, therefore playing a nucleating role, promoting spindle assembly and ensuring normal meiosis. In addition, we found the colocalization and interaction of collapsin response mediator protein 5 (CRMP5) and spastin in oocytes. CRMP5 can provide structural support and promote microtubule aggregation, creating transportation routes, and can interact with spastin in the microtubule activity of nerve cells (30). Knocking down CRMP5 may lead to spindle abnormalities and developmental disorders in oocytes. Overexpression of spastin may reverse the abnormal phenotype caused by the deletion of CRMP5. In summary, our data support a model in which the interaction between spastin and CRMP5 promotes the assembly of spindle microtubules in oocytes by controlling microtubule dynamics, therefore ensuring normal meiosis.

Invasive species face new selective pressures and low genetic variation caused by genetic bottlenecks and founder effects when they are introduced into novel environments. Epigenetic variation may help them to cope with these problems. Mile-a-minute (Mikania micrantha Kunth) is a highly invasive exotic weed that has seriously damaged biodiversity and agricultural ecosystems. We first adopted methylation-sensitive amplified polymorphism (MSAP) markers to investigate epigenetic variation of 21 M. micrantha populations in southern China, and further explored the effects of environmental factors on population epigenetic differentiation by correlating epigenetic and climate and soil data. Adaptive epiloci positively correlated with climate/soil variables were identified. Minimum temperature of the coldest month and mean temperature of the coldest quarter were considered as decisive factors for its distribution. Climate is presumed to play a relatively more important role than soil in shaping the adaptive epigenetic differentiation in M. micrantha. Under ongoing global warming, populations of M. micrantha are predicted to expand northward. In addition, the weed also presented higher epigenetic variation compared with genetic variation. Leaf shape variation was detected related to methylation-state change at the population level.

The flow topology and leading-edge vortex (LEV) formation and detachment mechanism of a pitching and plunging flat plate are experimentally investigated in this study. Focus is placed on three novel aspects. First, to examine the differences between start-up and cyclic motions, the flow fields of one-shot experiments are compared to cyclic cases. The results show that the start cycle has very limited effect on the cyclic LEV development and flow topology evolution. Next, the influence of the maximum effective angle of attack on the LEV development in cyclic motion is introduced. Different secondary structures that lead to the detachment of LEV are found with variation of maximum effective angle of attack. Depending on the leading-edge shear-layer angle, three different flow topologies develop on the plate: quasi-steady development, boundary-layer eruption and secondary vortex formation. Which of these three topological scenarios occurs depends entirely on the maximum effective angle of attack. A vortex Reynolds number based on the transition time of the leading-edge shear-layer angle is defined to quantitatively assess which of the flow topologies will appear. Finally, a simplified model to describe the observed LEV growth is proposed, based on the assumptions that the velocity is constant at the outer vortex boundary and that the vortex boundary is a circular arc starting from the leading edge. The LEV circulation growth is found to increase linearly with the combination of the effective inflow velocity and the effective angle of attack.

A roughness scaling behaviour is tested by performing the direct numerical simulation (DNS) of a turbulent channel flow over three-dimensional sinusoidal rough walls. By systematically varying the roughness height ${{k}^{+}}$ and the roughness steepness S, the results for three groups of cases are considered and compared with those for flat-wall turbulence. The results show that the mean velocity and Reynolds stresses are highly dependent on both ${{k}^{+}}$ and S. To describe these specific relationships, we define a coupling scale ${{k}^{+}} S$. With this coupling scale, all the simulated data for the roughness function (${\rm \Delta} {{U}^{+}}$), the ratio of the pressure drag to the total wall resistance (${{\gamma }_{p}}$), the normalized bulk mean velocity ($U_{b}^{+}$) and the peak of the streamwise turbulent velocity fluctuations ($\overline {u_{p}^{\prime +}}$) collapse onto single curves, which shows that there is a strong direct correlation between them, i.e. ${\rm \Delta} U^{+}, \gamma _{p}, U_{b}^{+}, \overline {u_{p}^{\prime +}} \propto f(k^{+} S)$. Furthermore, a model for the prediction of wall resistance based on the roughness function can be established by defining a drag increasing ratio (DI). Accordingly, the wall resistance coefficient ${{C}_{f}}$ can be estimated directly from ${{k}^{+}}S$ of a given rough surface. These results suggest that this coupling scale provides a useful alternative to the equivalent sand grain roughness ${{k}_{s}}$.

Dolostones are widely developed in the middle Permian rocks of East Yunnan, China, mainly in the shoal-facies Maokou Formation. The previously reported dolostone formation mechanisms cannot explain the distribution and geochemical characteristics of these dolostones, in particular their strontium, magnesium and oxygen isotope signatures. To help predict the distribution of dolostone reservoirs and reduce the exploration risk and cost, this study proposes a new model of dolomitization: open thermal convection dolomitization. In this new dolomitization model, Mg2+ in dolomitizing fluids originates mostly from seawater, with a minor component coming from deep hydrothermal fluids. Elevated heat flux (in this case due to the nearby Emei mantle plume) causes spatial temperature variations in the fluid along the circulation flow pathways, resulting in fast and pervasive dolomitization of limestone. The proposed model not only explains the characteristics and distribution of dolostones in the study area but also serves as a reference for predicting the distribution of dolostones in other areas subjected to thermal convection.

To investigate the protein-sparing effect of α-lipoic acid (LA), experimental fish (initial body weight: 18·99 (sd 1·82) g) were fed on a 0, 600 or 1200 mg/kg α-LA diet for 56 d, and hepatocytes were treated with 20 μm compound C, the inhibitor of AMP kinase α (AMPKα), treated for 30 min before α-LA treatment for 24 h. LA significantly decreased lipid content of the whole body and other tissues (P<0·05), and it also promoted protein deposition in vivo (P<0·05). Further, dietary LA significantly decreased the TAG content of serum and increased the NEFA content of serum (P<0·05); however, there were no significant differences among all groups in the hepatopancreas and muscle (P>0·05). Consistent with results from the experiment in vitro, LA activated phosphorylation of AMPKα and notably increased the protein content of adipose TAG lipase in intraperitoneal fat, hepatopancreas and muscle in vivo (P<0·05). Meanwhile, LA significantly up-regulated the mRNA expression of genes involved in fatty acid β-oxidation in the same three areas, and LA also obviously down-regulated the mRNA expression of genes involved in amino acid catabolism in muscle (P<0·05). Besides, it was observed that LA significantly activated the mammalian target of rapamycin (mTOR) pathway in muscle of experimental fish (P<0·05). LA could promote lipolysis and fatty acid β-oxidation via increasing energy supply from lipid catabolism, and then, it could economise on the protein from energy production to increase protein deposition in grass carp. Besides, LA might directly promote protein synthesis through activating the mTOR pathway.

The hydrogen concentration and composition of garnets in the ultrahigh pressure eclogites at Shuanghe, eastern Dabieshan, were investigated using Fourier transform infrared spectroscopy and electron microprobe analysis. The OH absorption bands can be divided into four groups: (1) 3635–3655 cm–1; (2) 3600–3630 cm–1; (3) 3540–3580 cm–1; and (4) 3400–3450 cm–1 and the water content ranges from 45 to 2529 ppm. Based on the behaviour of the OH absorption band and the relationship between water content and the composition of garnets, the samples can be divided into two classes: samples with >400 ppm H2O and samples with ≤400 ppm H2O. The water content of the former shows an obvious positive correlation with Ca atoms and a negative correlation with the Si, Mg and Fe2+ atoms per 12 anions, whereas the water content of the latter shows no obvious linear correlation with cations. It is concluded that the major mechanism of hydroxyl incorporation in garnets with >400 ppm H2O is by the coupled substitution 4H +Z□ → □+ZSi in the tetrahedral site, and that several mechanisms are responsible for OH incorporation in garnets with ≤400 ppm H2O.

The present study examines the characteristics of clay minerals in shale gas reservoirs and their influence on reservoir properties based on X-ray diffraction and scanning electron microscopy. These analyses were combined with optical microscopy observations and core and well-log data to investigate the genesis, distribution characteristics, main controlling factors and pore features of clay minerals of the Lower Silurian Longmaxi Formation in the East Sichuan area, China. The clay mineral assemblage consists of illite + mixed-layer illite-smectite (I-S) + chlorite. This assemblage includes three sources of clay minerals: detrital, authigenic and diagenetic minerals. The lower section of the Longmaxi Formation in the Jiaoshiba area has sealing ability which resulted in abnormal high pressures during hydrocarbon generation which inhibited illitization. Therefore, an anomalous transformation sequence is present in which the mixed-layer I-S content increases with depth. This anomalous transformation sequence can be used to infer the existence of abnormal high pressures. The detrital components of the formation also affect the clay-minerals content indirectly, especially the abundance of K-feldspar. The transformation of mixed-layer I-S to illite is limited due to the limited availability of K+, which determines the extent of transformation. Three types of pores were observed in the shale reservoir rocks of the Longmaxi Formation: interparticle (interP) pores, intraparticle (intraP) pores and organic-matter pores. The clay-mineral content controls the development of intraP pores, which are dominated by pores within clay particles. For a given clay mineral content, smectite and mixed-layer I-S were more conducive to the development of shale-gas reservoirs than other clay minerals.

An effective multiplex real-time polymerase chain reaction (PCR) assay for the simultaneous detection of three major pathogens, Nosema bombycis Nägeli (Microsporidia: Nosematidae), Bombyx mori nucleopolyhedrovirus (Baculoviridae: genus Alphabaculovirus) (NPV), and Bombyx mori densovirus (Parvoviridae: genus Iteravirus) (DNV), in silkworms (Bombyx mori (Linnaeus); Lepidoptera: Bombycidae) was developed in this study. Polymerase chain reaction and real-time PCR tests and basic local alignment search tool searches revealed that the primers and probes used in this study had high specificities for their target species. The ability of each primer/probe set to detect pure pathogen DNA was determined using a plasmid dilution panel, in which under optimal conditions the multiplex real-time PCR assay showed high efficiency in the detection of three mixed target plasmids with a detection limit of 8.5×103 copies for N. bombycis and Bombyx mori NPV (BmNPV) and 8.5×104 copies for Bombyx mori DNV (BmDNV). When the ability to detect these three pathogens was examined in artificially inoculated silkworms, our method presented a number of advantages over traditional microscopy, including specificity, sensitivity, and high-throughput capabilities. Under the optimal volume ratio for the three primer/probe sets (3:2:2=N. bombycis:BmNPV:BmDNV), the multiplex real-time PCR assay showed early detection of BmNPV and BmDNV by day 1 post inoculation using DNA templates of the three pathogens in various combinations from individually infected silkworms; the early detection of N. bombycis was possible by day 3 post inoculation using the DNA isolated from the midgut of N. bombycis-infected silkworms.

Maritime anomaly detection is a key technique in intelligent vessel traffic surveillance systems and implementation of maritime situational awareness. In this paper, we propose a method which combines vessel trajectory clustering and Naïve Bayes classifier to detect anomalous vessel behaviour in the maritime surveillance system. A similarity measurement between vessel trajectories is designed based on the spatial and directional characteristics of Automatic Identification System (AIS) data, then the method of hierarchical and k-medoids clustering are applied to model and learn the typical vessel sailing pattern within harbour waters. The Naïve Bayes classifier of vessel behaviour is built to classify and detect anomalous vessel behaviour. The proposed method has been tested and validated on the vessel trajectories from AIS data within the waters of Xiamen Bay and Chengsanjiao, China. The results indicate that the proposed method is effective and helpful, thus enhancing maritime situational awareness in coastal waters.

In this paper, we proposed a simple approach to analyse the efficiency and propulsive characteristics of the high-altitude propeller in accordance to the Vortex Lattice Lifting line Method (VLM) theory, which is commonly used in preliminary design and parametric studies of propeller propulsion. The Computational Fluid Dynamics (CFD) method was used to obtain aerofoil aerodynamic data. The optimal pitch angle and propeller blade chord length (along the radial direction) can be calculated using the information from the database. The propeller wake model sees helical slipstreams applied to both lightly and moderately loaded propellers. The proposed method is capable of identifying the optimal efficiency through varying the number of propeller blades, radius and the rotational speed. The relationship between the optimal efficiency and design parameters is then established. This method was verified using CFD calculations.

The high-energy oscillating electric current pulse (ECP) technology was introduced to relieve the residual stresses in the small AISI 1045 steel specimens treated by the pulsed-laser surface irradiation. The high-energy oscillating ECP stress relief experiments were conducted to study the effectiveness of the high-energy oscillating ECP technology. In addition, the electroplasticity framework was developed based on the thermal activation theory to reveal the mechanism of the high-energy oscillating ECP stress relief. The results show that the high-energy oscillating ECP stress relief has good effects on eliminating the residual stress. Furthermore, the residual stress relieving mechanism of the high-energy oscillating ECP stress relief can be attributed to the electric softening effect and the dynamic stress effect. The findings confirm that the significant effects of high-energy oscillating ECP on metal plasticity and provide a basis to understand the underlying mechanism of the high-energy oscillating ECP stress relief.

The high-frequency vibration technology was introduced to relieve the quenched residual stress in the Cr12MoV steel based on the high-frequency vibration system that mainly consisted of an electromagnetic vibrator and an amplitude boost unit. The high-frequency vibratory stress relief (VSR) experiments were conducted to study the effectiveness of the high-frequency vibration technology. In addition, the high-frequency vibration plasticity model was developed based on the thermal activation theory to reveal the mechanism of the high-frequency VSR. The results show that the high-frequency VSR has good effects on eliminating residual stress, while the surface hardness for the Cr12MoV steel remains almost the same. Moreover, there are no changes in the grain size of the Cr12MoV steel during the high-frequency VSR, while the dislocation density for the Cr12MoV steel during the high-frequency VSR decreases by 27.21%. The decrease of dislocation density in the Cr12MoV steel is the essence of residual stress relaxation. The findings confirm the significant effects of high-frequency vibration on metal plasticity and provide a basis to understand the underlying mechanism of the high-frequency VSR.
