Instability evolutions of shock-accelerated thin cylindrical SF$_6$ layers surrounded by air with initial perturbations imposed only at the outer interface (i.e. the ‘Outer’ case) or at the inner interface (i.e. the ‘Inner’ case) are numerically and theoretically investigated. It is found that the instability evolution of a thin cylindrical heavy fluid layer not only involves the effects of Richtmyer–Meshkov instability, Rayleigh–Taylor stability/instability and compressibility coupled with the Bell–Plesset effect, which determine the instability evolution of the single cylindrical interface, but also strongly depends on the waves reverberated inside the layer, thin-shell correction and interface coupling effect. Specifically, the rarefaction wave inside the thin fluid layer accelerates the outer interface inward and induces the decompression effect for both the Outer and Inner cases, and the compression wave inside the fluid layer accelerates the inner interface inward and causes the decompression effect for the Outer case and compression effect for the Inner case. It is noted that the compressible Bell model excluding the compression/decompression effect of waves, thin-shell correction and interface coupling effect deviates significantly from the perturbation growth. To this end, an improved compressible Bell model is proposed, including three new terms to quantify the compression/decompression effect of waves, thin-shell correction and interface coupling effect, respectively. This improved model is verified by numerical results and successfully characterizes various effects that contribute to the perturbation growth of a shock-accelerated thin heavy fluid layer.

A novel data-driven modal analysis method, reduced-order variational mode decomposition (RVMD), is proposed, inspired by the Hilbert–Huang transform and variational mode decomposition (VMD), to resolve transient or statistically non-stationary flow dynamics. First, the form of RVMD modes (referred to as an ‘elementary low-order dynamic process’, ELD) is constructed by combining low-order representation and the idea of intrinsic mode function, which enables the computed modes to characterize the non-stationary properties of space–time fluid flows. Then, the RVMD algorithm is designed based on VMD to achieve a low-redundant adaptive extraction of ELDs in flow data, with the modes computed by solving an elaborate optimization problem. Further, a combination of RVMD and Hilbert spectral analysis leads to a modal-based time-frequency analysis framework in the Hilbert view, providing a potentially powerful tool to discover, quantify and analyse the transient and non-stationary dynamics in complex flow problems. To provide a comprehensive evaluation, the computational cost and parameter dependence of RVMD are discussed, as well as the relations between RVMD and some classic modal decomposition methods. Finally, the virtues and utility of RVMD and the modal-based time-frequency analysis framework are well demonstrated via two canonical problems: the transient cylinder wake and the planar supersonic screeching jet.

Although ethanol treatment is widely used to activate oocytes, the underlying mechanisms are largely unclear. Roles of intracellular calcium stores and extracellular calcium in ethanol-induced activation (EIA) of oocytes remain to be verified, and whether calcium-sensing receptor (CaSR) is involved in EIA is unknown. This study showed that calcium-free ageing (CFA) in vitro significantly decreased intracellular stored calcium (sCa) and CaSR expression, and impaired EIA, spindle/chromosome morphology and developmental potential of mouse oocytes. Although EIA in oocytes with full sCa after ageing with calcium does not require calcium influx, calcium influx is essential for EIA of oocytes with reduced sCa after CFA. Furthermore, the extremely low EIA rate in oocytes with CFA-downregulated CaSR expression and the fact that inhibiting CaSR significantly decreased the EIA of oocytes with a full complement of CaSR suggest that CaSR played a significant role in the EIA of ageing oocytes. In conclusion, CFA impaired EIA and the developmental potential of mouse oocytes by decreasing sCa and downregulating CaSR expression. Because mouse oocytes routinely treated for activation (18 h post hCG) are equipped with a full sCa complement and CaSR, the present results suggest that, while calcium influx is not essential, CaSR is required for the EIA of oocytes.

For a non-conformal repeller $\Lambda $ of a $C^{1+\alpha }$ map f preserving an ergodic measure $\mu $ of positive entropy, this paper shows that the Lyapunov dimension of $\mu $ can be approximated gradually by the Carathéodory singular dimension of a sequence of horseshoes. For a $C^{1+\alpha }$ diffeomorphism f preserving a hyperbolic ergodic measure $\mu $ of positive entropy, if $(f, \mu )$ has only two Lyapunov exponents $\unicode{x3bb} _u(\mu )>0>\unicode{x3bb} _s(\mu )$ , then the Hausdorff or lower box or upper box dimension of $\mu $ can be approximated by the corresponding dimension of the horseshoes $\{\Lambda _n\}$ . The same statement holds true if f is a $C^1$ diffeomorphism with a dominated Oseledet’s splitting with respect to $\mu $ .

For a $C^1$ non-conformal repeller, this paper proves that there exists an ergodic measure of full Carathéodory singular dimension. For an average conformal hyperbolic set of a $C^1$ diffeomorphism, this paper constructs a Borel probability measure (with support strictly inside the repeller) of full Hausdorff dimension. If the average conformal hyperbolic set is of a $C^{1+\alpha }$ diffeomorphism, this paper shows that there exists an ergodic measure of maximal dimension.

The highly nonlinear evolution of the single-mode stratified compressible Rayleigh–Taylor instability (RTI) is investigated via direct numerical simulation over a range of Atwood numbers ($A_T=0.1$–$0.9$) and Mach numbers ($Ma=0.1$–$0.7$) for characterising the isothermal background stratification. After the potential stage, it is found that the bubble is accelerated to a velocity which is well above the saturation value predicted in the potential flow model. Unlike the bubble re-acceleration behaviour in quasi-incompressible RTI with uniform background density, the characteristics in the stratified compressible RTI are driven by not only vorticity accumulation inside the bubble but also flow compressibility resulting from the stratification. Specifically, in the case of strong stratification and high $A_T$, the flow compressibility dominates the bubble re-acceleration characters. To model the effect of flow compressibility, we propose a novel model to reliably describe the bubble re-acceleration behaviours in the stratified compressible RTI, via introducing the dilatation into the classical model that takes into account only vorticity accumulation.

Since embryonic stem cells (ESCs) were first identified, significant progress has been achieved. However, the establishment of buffalo ESCs (bESCs) is still unclear. This study was undertaken to explore the effect of the blastocyst stage on the isolation of bESCs. Firstly, our results indicated that the pluripotent genes were mainly expressed at the early stages of blastocyst, and the attachment and colony formation rates of bESCs derived from expanded blastocyst and hatched blastocyst were significantly higher than early blastocyst and blastocyst. In the meantime, bESCs showed positive alkaline phosphatase activity and expressed genes like OCT4, NANOG, SOX2, c-MYC, CDH1, KLF4, and TBX3. Immunofluorescence also confirmed the expression of OCT4, SOX2. Embryoid bodies expressing three marker genes were generated from the differentiation experiment, and fibroblast, epithelial, and neuron-like cells were induced. Moreover, naive-related genes KLF4, TBX3, primed-related genes FGF5, ACTA2 were expressed in the cells, but not REX-1. Immunofluorescence and western blot confirmed the FGF5 expression. Furthermore, bESCs could maintain pluripotency with the signal of LIF and bFGF. In summary, our results indicated that expanded blastocyst and hatched blastocyst are more suitable for bESCs isolation.

A new system for preparing 14C samples was established for a compact accelerator mass spectrometer (GXNU-AMS) at Guangxi Normal University. This sample preparation system consists of three units: a vacuum maintenance unit, a CO2 purification unit, and a CO2 reduction unit, all of which were made of quartz glass. A series of radiocarbon (14C) preparation experiments were conducted to verify the reliability of the system. The recovery rate of graphite obtained was more than 80%. The carbon content in the commercial toner and wood sample was linearly fitted to the CO2 pressure in the measurement unit of the system. The results showed a good linear relationship, indicating that the reliability of the sample preparation system. AMS measurements were conducted on a batch of standard, wood, and dead graphite samples prepared using this system. The results showed that the beam current of 12C- for each sample was more than 40 μA, the carbon contamination introduced during the sample preparation process was ∼ 2 × 10–15, and that the new sample preparation system is compact, low-contamination, and efficient and meets the GXNU-AMS requirements for 14C samples.

Many waterflooding oil fields, injecting water into an oil-bearing reservoir for pressure maintenance, are in their middle to late stages of development. To explore the geological conditions and improve oilfield recovery of the most important well group of the Hu 136 block, located on the border areas of three provinces (Henan, Shandong, and Hebei), Zhongyuan Oilfield, Sinopec, central China, a 14C cross-well tracer monitoring technology was developed and applied in monitoring the development status and recognize the heterogeneity of oil reservoirs. The tracer response in the production well was tracked, and the water drive speed, swept volume of the injection fluid were obtained. Finally, the reservoir heterogeneity characteristics, such as the dilution coefficient, porosity, permeability, and average pore-throat radius, were fitted according to the mathematical model of the heterogeneous multi-layer inter-well theory. The 14C-AMS technique developed in this work is expected to be a potential analytical method for evaluating underground reservoir characteristics and providing crucial scientific guidance for the mid to late oil field recovery process.

The gut microbiota plays an important role in animals’ survival in their local environments. The intertidal rocky shore is a key interface of oceanic, atmospheric and terrestrial environments, and the transmission modes of microbes between an intertidal host and the environment are complex and largely ignored. In the present study, we characterized the gut microbiota of the intertidal snail Nerita yoldii, which is experiencing a northward range shift under the combined impacts of climate change and anthropogenic seascape transformation, and also determined the nearby environmental microbiota on the rock and in the seawater at five locations along the snail's distribution range in China. The gut microbial communities were significantly different from the environmental microbial communities, and the dominant phyla were Tenericutes, Cyanobacteria and Bacteroidetes, and Proteobacteria in the gut, rock and seawater microbial communities, respectively. At the genus level, Mycoplasma, with a relative abundance of 48.0 ± 10.2%, was the dominant genus in the gut microbial community, however, the relative abundances of this genus on the rock and in the water were low. These results imply that the gut microbial community of the intertidal snail N. yoldii is relatively independent from the environmental microbial community, and the dominant genus Mycoplasma in the gut, that is rare in the environment, can potentially assist the snail living in the harsh intertidal environment, especially at its northernmost distribution range edge.

While the detrimental effect of interparental conflict on adolescent depression is well-established, the underlying mechanisms linking the two continue to be inadequately understood. This study investigated the mediating role of family functioning and the moderating role of cultural beliefs about adversity in the association between interparental conflict and adolescent depression. The samples included 651 Chinese adolescents (mean age at Time 1 = 13.27 years; 56.5% girls) from a two-wave longitudinal study with data spanning 1 year. The findings from path modeling analyses provided evidence for the mediating role of family functioning; these findings indicated that interparental conflict can damage family functioning, which in turn exacerbates the risk of adolescent depression. The moderating role of cultural beliefs about adversity was also demonstrated by interactions between interparental conflict and cultural beliefs about adversity, as well as, family functioning and cultural beliefs about adversity. The results indicated a buffering role of cultural beliefs about adversity on the deleterious effect of interparental conflict on adolescent depression. They also suggested that lower levels of family functioning was associated with increased depression among adolescents were lower in cultural beliefs about adversity.

Athetis lepigone Möschler (Lepidoptera, Noctuidae) is a common maize pest in Europe and Asia. However, there is no long-term effective management strategy is available yet to suppress its population. Adults rely heavily on olfactory cues to locate their optimal host plants and oviposition sites. Pheromone-binding proteins (PBPs) are believed to be responsible for recognizing and transporting different odorant molecules to interact with receptor membrane proteins. In this study, the ligand-binding specificities of two AlepPBPs (AlepPBP2 and AlepPBP3) for sex pheromone components and host plant (maize) volatiles were measured by fluorescence ligand-binding assay. The results demonstrated that AlepPBP2 had a high affinity with two pheromones [(Z)-7-dodecenyl acetate, Ki = 1.11 ± 0.1 μM, (Z)-9-tetradecenyl acetate, Ki = 1.32 ± 0.15 μM] and ten plant volatiles, including (-)-limonene, α-pinene, myrcene, linalool, benzaldehyde, nonanal, 2-hexanone, 3-hexanone, 2-heptanone and 6-methyl-5-hepten-2-one. In contrast, we found that none of these chemicals could bind to AlepPBP3. Our results clearly show no significant differences in the functional characterization of the binding properties between AlepPBP2 and AlepPBP3 to sex pheromones and host plant volatiles. Furthermore, molecular docking was employed for further detail on some crucial amino acid residues involved in the ligand-binding of AlepPBP2. These findings will provide valuable information about the potential protein binding sites necessary for protein-ligand interactions which appear as attractive targets for the development of novel technologies and management strategies for insect pests.

The majority of plant viral disease is transmitted and spread by insect vectors in the field. The small brown planthopper, Laodelphax striatellus (Fallén), is the only efficient vector for rice black-streaked dwarf virus (RBSDV), a devastating plant virus that infects multiple grain crops, including rice, maize, and wheat. Adenosine triphosphate (ATP)-binding cassette (ABC) transporters participate in various biological processes. However, little is known about whether ABC transporters affect virus infection in insects. In this study, RBSDV accumulation was significantly reduced in L. striatellus after treatment with verapamil, an effective inhibitor of ABC transporters. Thirty-four ABC transporter genes were identified in L. striatellus and expression analysis showed that LsABCF2 and LsABCG9 were significantly upregulated and downregulated, respectively, after RBSDV infection. LsABCF2 and LsABCG9 were expressed during all developmental stages, and LsABCG9 was highly expressed in the midgut of L. striatellus. Knockdown of LsABCF2 promoted RBSDV accumulation, while knockdown of LsABCG9 suppressed RBSDV accumulation in L. striatellus. Our data showed that L. striatellus might upregulate the expression of LsABCF2 and downregulate LsABCG9 expression to suppress RBSDV infection. These results will contribute to understanding the effects of ABC transporters on virus transmission and provide theoretical basis for virus management in the field.

Vitamin D, Ca and dairy products are negatively associated with colorectal cancer (CRC) incidence, but little is known of their influence on CRC survival. To investigate prediagnostic intakes of vitamin D, Ca and dairy products for their relevance to CRC prognosis, we analysed 504 CRC patients enrolled in the Newfoundland Colorectal Cancer Registry Cohort Study who were diagnosed for the first time with CRC between 1999 and 2003. Follow-up for mortality and cancer recurrence was through April 2010. Data on diet and lifestyle factors were gathered via a validated, semi-quantitative FFQ and a Personal History Questionnaire. Multivariate Cox models estimated hazard ratios (HR) and 95 % CI for the relationship of prediagnostic intakes of vitamin D, Ca and dairy products with all-cause mortality (overall survival, OS) and disease-free survival (DFS) among CRC patients. We found that prediagnostic Ca intake from foods, but not total Ca intake, was negatively associated with all-cause mortality (HR for Q2 v. Q1, 0·44; 95 % CI, 0·26, 0·75). An inverse relationship was also seen in a dose–response fashion for prediagnostic cheese intake (HR for Q4 v. Q1, 0·57, 95 % CI, 0·34, 0·95, P trend = 0·029). No evidence for modification by sex, physical activity, alcohol drinking and cigarette smoking was observed. In summary, high prediagnostic intakes of cheese and Ca from foods may be associated with increased survival among CRC patients. By manipulating diet, this study may contribute to the development of novel therapies that add to the armamentarium against CRC. Replication studies are required before any nutritional interventions are made available.

The present study evaluated effects of dietary supplementation with tryptophan (Trp) on muscle growth, protein synthesis and antioxidant capacity in hybrid catfish Pelteobagrus vachelli♀ × Leiocassis longirostris♂. Fish were fed six different diets containing 2·6 (control), 3·1, 3·7, 4·2, 4·7 and 5·6 g Trp/kg diet for 56 d, respectively. Results showed that dietary Trp significantly (1) improved muscle protein content, fibre density and frequency of fibre diameter; (2) up-regulated the mRNA levels of PCNA, myf5, MyoD1, MyoG, MRF4, IGF-I, IGF-II, IGF-IR, PIK3Ca, TOR, 4EBP1 and S6K1; (3) increased phosphorylation levels of AKT, TOR and S6K1; (4) decreased contents of MDA and PC, and increased activities of CAT, GST, GR, ASA and AHR; (5) up-regulated mRNA levels of CuZnSOD, CAT, GST, GPx, GCLC and Nrf2, and decreased Keap1 mRNA level; (6) increased nuclear Nrf2 protein level and the intranuclear antioxidant response element-binding ability, and reduced Keap1 protein level. These results indicated that dietary Trp improved muscle growth, protein synthesis as well as antioxidant capacity, which might be partly related to myogenic regulatory factors, IGF/PIK3Ca/AKT/TOR and Keap1/Nrf2 signalling pathways. Finally, based on the quadratic regression analysis of muscle protein and MDA contents, the optimal Trp requirements of hybrid catfish (21·82–39·64 g) were estimated to be 3·94 and 3·93 g Trp/kg diet (9·57 and 9·54 g/kg of dietary protein), respectively.

Somatic cell nuclear transfer (SCNT) holds vast potential in agriculture. However, its applications are still limited by its low efficiency. Histone 3 lysine 9 trimethylation (H3K9me3) was identified as an epigenetic barrier for this. Histone demethylase KDM4D could regulate the level of H3K9me3. However, its effects on buffalo SCNT embryos are still unclear. Thus, we performed this study to explore the effects and underlying mechanism of KDM4D on buffalo SCNT embryos. The results revealed that compared with the IVF embryos, the expression level of KDM4D in SCNT embryos was significantly lower at 8- and 16-cell stage, while the level of H3K9me3 in SCNT embryos was significantly higher at 2-cell, 8-cell, and blastocyst stage. Microinjection of KDM4D mRNA could promote the developmental ability of buffalo SCNT embryos. Furthermore, the expression level of ZGA-related genes such as ZSCAN5B, SNAI1, eIF-3a, and TRC at the 8-cell stage was significantly increased. Meanwhile, the pluripotency-related genes like POU5F1, SOX2, and NANOG were also significantly promoted at the blastocyst stage. The results were reversed after KDM4D was inhibited. Altogether, these results revealed that KDM4D could correct the H3K9me3 level, increase the expression level of ZGA and pluripotency-related genes, and finally, promote the developmental competence of buffalo SCNT embryos.

Kinetic energy and enstrophy transfer in compressible Rayleigh–Taylor (RT) turbulence were investigated by means of direct numerical simulation. It is revealed that compressibility plays an important role in the kinetic energy and enstrophy transfer based on analyses of transport and large-scale equations. For the generation and transfer of kinetic energy, some findings have been obtained as follows. The pressure-dilatation work dominates the generation of kinetic energy in the early stage of flow evolution. The baropycnal work and deformation work handle the kinetic energy transfer from large to small scales on average for RT turbulence. The baropycnal work is mainly responsible for the kinetic energy transfer on large scales, and the deformation work for the kinetic energy transfer on small scales. The baropycnal work is also disclosed to be related to the compressibility from the finding that the expansion motion enhances the positive baropycnal work and the compression motion strengthens the negative baropycnal work. For the generation and transfer of enstrophy, the horizontal enstrophy is generated by the baroclinic effect and the vertical enstrophy by vortex stretching and tilting. Then the enstrophy is strengthened by the vortex stretching and tilting during the evolution of RT turbulence and the vorticity tends to be isotropic in the turbulent mixing region. The large-scale enstrophy equation in compressible flow has also been derived to deal with the enstrophy transfer. It is identified that the enstrophy is transferred from large to small scales on average and tends to stabilize for RT turbulence.

A one-dimensional steady-state model for stimulated Raman backscatter (SRS) and stimulated Brillouin backscatter (SBS) processes in laser-irradiated plasmas is presented. Based on a novel “predictor-corrector” method, the model is capable to deal with broadband scattered light and inhomogeneous plasmas, exhibiting robustness and high efficiency. Influences of the electron density and temperature on the linear gains of both SRS and SBS are investigated, which indicates that the SRS gain is more sensitive to the electron density and temperature than that of the SBS. For the low-density case, the SBS dominates the scattering process, while the SRS exhibits much higher reflectivity in the high-density case. The nonlinear saturation mechanisms and competition between SRS and SBS are included in our model by a phenomenological method. The typical anti-correlation between SRS and SBS versus electron density is reproduced in the model. Calculations of the reflectivities are qualitatively in agreement with the typical results of experiments and simulations.
