Background. Calcific aortic valve disease (CAVD) is the most common native valve disease. Valvular interstitial cell (VIC) osteogenic differentiation and valvular endothelial cell (VEC) dysfunction are key steps in CAVD progression. Circular RNA (circRNAs) is involved in regulating osteogenic differentiation with mesenchymal cells and is associated with multiple disease progression, but the function of circRNAs in CAVD remains unknown. Here, we aimed to investigate the effect and potential significance of circRNA-miRNA-mRNA networks in CAVD. Methods. Two mRNA datasets, one miRNA dataset, and one circRNA dataset of CAVD downloaded from GEO were used to identify DE-circRNAs, DE-miRNAs, and DE-mRNAs. Based on the online website prediction function, the common mRNAs (FmRNAs) for constructing circRNA-miRNA-mRNA networks were identified. GO and KEGG enrichment analyses were performed on FmRNAs. In addition, hub genes were identified by PPI networks. Based on the expression of each data set, the circRNA-miRNA-hub gene network was constructed by Cytoscape (version 3.6.1). Results. 32 DE-circRNAs, 206 DE-miRNAs, and 2170 DE-mRNAs were identified. Fifty-nine FmRNAs were obtained by intersection. The KEGG pathway analysis of FmRNAs was enriched in pathways in cancer, JAK-STAT signaling pathway, cell cycle, and MAPK signaling pathway. Meanwhile, transcription, nucleolus, and protein homodimerization activity were significantly enriched in GO analysis. Eight hub genes were identified based on the PPI network. Three possible regulatory networks in CAVD disease were obtained based on the biological functions of circRNAs including: hsa_circ_0026817-hsa-miR-211-5p-CACNA1C, hsa_circ_0007215-hsa-miR-1252-5p-MECP2, and hsa_circ_0007215-hsa-miR-1343-3p- RBL1. Conclusion. The present bionformatics analysis suggests the functional effect for the circRNA-miRNA-mRNA network in CAVD pathogenesis and provides new targets for therapeutics.

Our research question was to evaluate the chromosome concordance of trophectoderm (TE) biopsy with noninvasive chromosome screening (NICS) using embryo culture medium renewed twice on Day 3 (D3) and Day 4 (D4). In this study, we evaluated 64 cycles with 223 biopsied blastocysts. These were categorized into two groups based on replacing embryo culture medium on D3 (control group) or on D3 and D4 (experimental group). The fundamental characteristics and main outcomes were compared. The concordance rates of NICS results with TE biopsy were determined according to next generation sequencing results. In total, 103 experimental and 120 control embryo cultures were collected, and the euploid status was analyzed using NICS technology. The overall concordance rates with TE biopsy of the experimental and control groups were 0.86 and 0.75, respectively. Statistically significant difference was found between the two groups. An additional medium renewal of the D4 embryo culture can improve the concordance of NICS with TE biopsy.

This work studies the detachment of a micron-sized spherical particle from a surface with concave roughness in a linear shear flow. The concave roughness is described as regularly spaced hollow hemispheres below a flat surface and is characterised by two dimensionless parameters, i.e. dimensionless asperity distance and asperity size ratio. The hydrodynamic force and torque on the particle are calculated by performing lattice Boltzmann simulations for particle Reynolds numbers ranging from 0.02 to 40. Empirical correlations of the drag, lift and torque coefficients of the particle as functions of the particle Reynolds number and the asperity size ratio are proposed. For detachment by lifting, sliding and rolling, a numerical approach to calculate the critical particle Reynolds number (i.e. above which the particle can detach from the surface) is proposed. It is found that the dimensionless asperity distance and the distribution of asperities on the rough surface have a minor influence on the hydrodynamic force and torque on the particle, and the detachment of the particle becomes more difficult as the particle sits deeper in a larger hole. Both the empirical correlations and the numerical approach can be implemented into Lagrangian particle tracking and can accurately predict the detachment of particles from the surface with concave roughness or the detachment of particles embedded in a flat surface.

Varicella is an acute respiratory infectious diseases, with high transmissibility and quick dissemination. In this study, an SEIR (susceptible-exposed-infected-recovered) dynamic model was established to explore the optimal prevention and control measures according to the epidemiological characteristics about varicella outbreak in a school in a central city of China. Berkeley Madonna 8.3.18 and Microsoft Office Excel 2010 software were employed for the model simulation and data management, respectively. The result showed that the simulated result of SEIR model agreed well with the reported data when β (infected rate) equal to 0.067. Models showed that the cumulative number of cases was only 13 when isolation adopted when the infected individuals were identified (assuming isolation rate was up to 100%); the cumulative number of cases was only two and the TAR (total attack rate) was 0.56% when the vaccination coefficient reached 50%. The cumulative number of cases did not change significantly with the change of efficiency of ventilation and disinfection, but the peak time was delayed; when δ (vaccination coefficient) = 0.1, m (ventilation efficiency) = 0.7 or δ = 0.2, m = 0.5 or δ = 0.3, m = 0.1 or δ = 0.4 and above, the cumulative number of cases would reduce to one case and TAR would reduce to 0.28% with combined interventions. Varicella outbreak in school could be controlled through strict isolation or vaccination singly; combined interventions have been adopted when the vaccination coefficient was low.

In seeking to respond to the environmental challenges of the Anthropocene era, our research team of five scholars, including faculty and advanced graduate students, along with each of their collaborators in their respective research sites, has come together to explore the possibilities of a methodology that we call Field to Media. Field to Media involves using video production to study and amplify ecomusical responses to climate change, pollution, deforestation, and other environmental challenges. This methodology is intended as a pragmatic process that blends participant observation with participatory action research and applied or activist engagement. Specific to this project, our efforts have involved the co-creation of five different music videos to address a range of pressing environment-related matters in USA/Canada, Tanzania, Bangladesh, China, and Haiti. In this article, we consider some of the potential successes and challenges that we have each experienced in the course of producing these music videos.

Low-density lipoproteins (LDL) is known to protect boar sperm during freezing–thawing, but little information is known about the effects of LDL extracted from different avian egg yolks on post-thaw boar semen quality. The purpose of this study was to compare and analyze the effects of LDL at various concentrations and different species on boar sperm quality after freezing–thawing. LDL extracted from the yolk of hen egg, duck egg, quail egg, pigeon egg or ostrich egg was added to the extender at the concentrations of 0.06, 0.07, 0.08, 0.09 and 0.1 g/ml, respectively, and their effects on frozen–thawed boar sperm quality were assessed. According to all measured parameters, the results showed that sperm motility, acrosome integrity and plasma membrane integrity were 43.20%, 52.57% and 48.13%, respectively, after being frozen–thawed with 0.09 g/ml LDL extracted from pigeon egg yolk. All these quality parameters were higher than that of other groups (P < 0.05). In conclusion, our results confirmed that LDL extracted from pigeon egg yolk had the best cryoprotective effects on frozen–thawed boar sperm among all of the groups supplemented with LDL from five kinds of avian egg in extender. The optimum concentration of LDL extracted from pigeon egg in boar semen freezing extender was 0.09 g/ml.

Health is a multidimensional and continual concept. Traditional latent analytic approaches have inherent deficits in capturing the complex nature of the concept; however, the Grade of Membership (GoM) model is well suited for this problem. We applied the GoM method to a set of 31 indicators to construct ideal profiles of health status based on physical, mental and social support items among 848 adult twins from Qingdao, China. Four profiles were identified: healthy individuals (pure type I), individuals with personality disorders (pure type II), individuals with mental impairments (pure type III) and individuals with physical impairments (pure type IV). The most frequently occurring combination in this population was profiles I, II, IV (14.74%), followed by profiles I, II, III, IV (13.44%), and then type I (11.08%). Only 13.56% of subjects fell completely into one single pure type, most individuals exhibited some of the characteristics of two or more pure types. Our results indicated that, compared to conventional statistical methods, the GoM model was more suited to capture the complex concept of health, reflecting its multidimensionality and continuity, while also exhibiting preferable reliability. This study also made an important contribution to research on GoM application in non-independent samples.

A gradient structure was synthesized on the surface of Zr55Al10Ni5Cu30 alloy with high glass-forming ability by laser surface melting (LSM). Along the laser incident direction, the surface remelted alloy exhibits gradient microstructure distributed in the sequence of amorphous structure, nanocrystal- reinforced amorphous matrix composite (transitional layer A), dendrites–amorphous phase composite (transitional layer B), and crystalline phases from the top surface to the substrate. The formation mechanism of this gradient structure is discussed based on the experimental results of the microstructure together with the finite volume simulation of the process of LSM treatment. The friction coefficient of the transitional layer A is ∼2.5 times lower than those of the other layers under the same sliding friction condition, and possible reasons for this phenomenon are discussed in connection with the rolling motion and material transfer mechanism. The transitional layer B exhibits the best wear resistance among all the structures studied here, which is related to the optimized ratio of microhardness to reduced Young’s modulus (H/Er).

This study was conducted to establish pig embryonic stem (ES)-like cell lines from nuclear transfer blastocysts. A green fluorescent protein (GFP)-expressing cell line was used as the source of donor cells injected into the enucleated oocytes. Blastocysts were collected at D5 (the fifth day), D7 (the seventh day) and D9 (the ninth day). Differential staining was used to assay the viability and development of blastocysts from the 3 days. The number of inner cell mass (ICM) cells increased from 1.83 ± 0.8 (D5) to 5.37 ± 1.2 (D7) to 7.56 ± 1.5 (D9). The expression profiles of embryonic stem (ES) cell factors (OCT4, SOX2, KLF4 and c-MYC) correlated best with the undifferentiated ES state and were identified by qPCR. The expression of the four factors was increased from D5 to D7, whereas the expression decreased from D7 to D9. We tried to isolate ES-like cells from these embryos. However, ES-like cells from the D7 blastocysts grew slowly and expressed alkaline phosphatase. The cells from the D9 blastocysts grew rapidly but did not express alkaline phosphatase. ES-like cells were not isolated from the D5 blastocysts. These results show that the cells from the D7 embryos are pluripotent but grow slowly. The cells from the D9 embryos grow rapidly but start to lose pluripotency.

Ni- and Cu-free Zr–Al–Co–Ag bulk metallic glasses (BMGs) with diameters up to 20 mm were synthesized by copper mold casting. The effects of Ag alloying on the superior glass-forming ability (GFA) of Zr–Al–Co–Ag alloys were studied based on the localized atomic structure and crystallization behavior. High-energy synchrotron radiation x-ray diffraction result reveals that Ag addition in Zr–Al–Co system results in a more homogeneous local atomic structure, which could be an origin for the improved GFA of the Zr–Al–Co–Ag alloy. Crystallization products of the Zr–Al–Co–Ag glassy alloy are more complex than those of the Zr–Al–Co glassy alloy. The Zr–Al–Co–Ag BMGs free from highly toxic elements Ni and Cu exhibited a combination of superior GFA, high compressive fracture strength over 2000 MPa, low Young’s modulus of 93 to 94 GPa, and good corrosion resistance in phosphate-buffered solution (PBS), inspiring their potential biomedical applications.

Miniature pigs have been recognized as valuable experimental animals in medical research. However, porcine models related to gene knockout of human diseases are not widely available. The objective of this study was to establish Mx1-Cre pigs using somatic cell nuclear transfer. In this study, we created transgenic pigs using somatic cell nuclear transfer (SCNT). Transfer of 210, 230, 250 and 215 zygotes to four surrogates produced 10 piglets. The Cre recombinase expression in transgenic pigs was studied using reverse transcriptase (RT)-PCR and immunohistochemistry. Mx1-Cre swine were shown to harbor the Cre gene in their genomic DNA using the PCR. In conclusion, Mx1-Cre transgenic piglets were successfully produced by SCNT. These transgenic swine, in conjunction with inducible systems for controlling Cre expression and function, are likely to have a profound impact on the study of human diseases.

Using a cold graphite mold casting method, bulk AlNiY chill-zone alloys were prepared at hypereutectic compositions with Al content from 85 at.% to 94 at.%. It was found that ultra-hard surface layers with a thickness of about 200 μm and submicron grain size form when the melt can be undercooled without heterogeneous nucleation at the mold contact surface. This hard chill-zone forming in contact with the mold possesses Vickers microhardness Hv about 350–420 and is thus harder than fully amorphous Al alloys. In compression, ultimate strength more than 1.1 GPa and true strain more than 150% without failure were achieved simultaneously. The combination of high strength and good plasticity will be discussed in relation to the special structure of the chill-zone alloy.

Dendrobium Sw., a member of the family Orchidaceae, includes approximately 1100 species distributed in different parts of the world. In China, the genus is represented by 76 species and two varieties, of which D. loddigesii, D. fimbriatum, D. chrysanthum, D. officinale (=D. candidum) and D. nobile are listed in the Chinese Pharmacopoeia as the source material for Herba Dendrobii (Shihu). Because of increased demand and high price, Herba Dendrobii is often adulterated in the trade by other related species. Many Dendrobium species are over-collected from the wild and are listed under endangered taxa in the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). Therefore, a reliable authentication method is needed to regulate the trade and help conserve the species from unscrupulous collections. This review describes the present status of using medicinal Dendrobium in China, current approaches to authenticate medicinal Dendrobium plants and Herba Dendrobii, and the efforts under way towards the development of DNA microarrays to facilitate differentiation/identification of genuine material in complex Chinese medicinal formulations.

Piezoelectric devices generally require film thicknesses in the range 2-200µm and have a response which is perturbed by the substrate. Sol gel and sol gel composite technology provide a manufacturable process for many purposes with a piezoelectric coupling coefficient for 40ptm thick PZT approaching kt= 0.34. Techniques such as laser machining or micromolding are required for patterning thick devices. Measurement and applications of piezoelectric coatings in medical imaging, high temperature ultrasonics and flexure devices are discussed.
