OBJECTIVES/GOALS: The effect of immunosuppressive metabolites on anti-tumor immunity in human papillomavirus (HPV)-associated vs carcinogen-driven head and neck cancer is unknown. The objective of this study is to define the extent to which metabolites impair this response and identify novel metabolic targets for enhancing anti-tumor immunity. METHODS/STUDY POPULATION: HPV-associated and carcinogen-driven head and neck squamous cell carcinoma specimens were frozen following surgical excision, and tumor sections were cut onto glass slides. Slides were coated in alpha-cyano-4-hydroxy-cinnamic acid (CHCA) matrix and subjected to mass spectrometry imaging using matrix-assisted laser desorption ionization (MALDI) on a Bruker SolariX XR 12T Hybrid QqFT-ICR mass spectrometer run in positive mode. Slides were then stained for immunohistochemistry (IHC) using markers of CD8 T cells, macrophages (CD163), B cells (CD20), and tumor cells (panCK). Mass spectrometry imaging and IHC spatially resolved data will be co-registered and metabolite intensity in regions of interest (cell types) quantified. RESULTS/ANTICIPATED RESULTS: A total of seven HPV-associated (three metastatic lymph nodes and four primary tumors) and six carcinogen-driven (primary tumors) HNSC specimens were subjected to MALDI and IHC. Metabolites significantly enriched in HPV-associated HNSC relative to carcinogen-driven HNSC include 2,3-diphosphoglyceric acid, xanthine, 2,3,5-Trichloromaleylacetate, and indole-3-carboxyaldehyde. Metabolites significantly enriched in carcinogen-driven HNSC relative to HPV-associated HNSC include hesperetin 3'-O-sulfate, hypoxanthine, phosphorylcholine, and L-homocysteine sulfonic acid. In ongoing analyses, we anticipate identifying a relationship between CD8+ T cell enriched vs depleted regions and immunosuppressive metabolites (e.g., kynurenine, adenosine monophosphate). DISCUSSION/SIGNIFICANCE: Defining the extent to which CD8+ T cells interact with the metabolic milieu of the microenvironment will provide a foundation for metabolic Precision Medicine. Strategically targeting metabolic pathways to enhance the anti-tumor immune response will be leveraged for the design and implementation of immune modulatory metabolic therapy.

In this paper, we describe the system design and capabilities of the Australian Square Kilometre Array Pathfinder (ASKAP) radio telescope at the conclusion of its construction project and commencement of science operations. ASKAP is one of the first radio telescopes to deploy phased array feed (PAF) technology on a large scale, giving it an instantaneous field of view that covers $31\,\textrm{deg}^{2}$ at $800\,\textrm{MHz}$. As a two-dimensional array of 36$\times$12 m antennas, with baselines ranging from 22 m to 6 km, ASKAP also has excellent snapshot imaging capability and 10 arcsec resolution. This, combined with 288 MHz of instantaneous bandwidth and a unique third axis of rotation on each antenna, gives ASKAP the capability to create high dynamic range images of large sky areas very quickly. It is an excellent telescope for surveys between 700 and $1800\,\textrm{MHz}$ and is expected to facilitate great advances in our understanding of galaxy formation, cosmology, and radio transients while opening new parameter space for discovery of the unknown.

Winter cover crops (CCs) provide soil conservation benefits for strip-tillage tobacco producers, but soil-residual herbicides may interfere with their establishment and growth. Tobacco is planted later than many agronomic crops, but growers often terminate CCs early to minimize CC residue at planting, and this may reduce weed suppression potential. We examined residual herbicide effects on CCs across two seasons and the potential for CC-based weed suppression within strip-tilled tobacco. Mixtures of wheat plus crimson clover and cereal rye plus crimson clover were examined, with a no-CC control. Herbicides included two rates of PRE sulfentrazone (177 or 354 g ai ha–1) plus carfentrazone (20 or 40 g ai ha–1); the higher rate was also followed by POST clomazone (840 g ai ha–1) or mixed with PRE pendimethalin (1,400 g ai ha–1). Controls with no weed management and hand weeding were also included. CC density and biomass were not reduced by weed management (WM) treatments with residual herbicides. However, CCs did not reduce density of annual grasses, small-seeded broadleaves, or perennials in the tilled in-row or untilled between-row zones. Cereal rye plus crimson clover resulted in lower weed biomass at tobacco harvest in the untilled between-row zone in 2017. WM effects were variable between the years, weed groups, and zones. Adding clomazone or pendimethalin was more consistent for reducing weed density and biomass compared to the low rate of sulfentrazone plus carfentrazone. Tobacco yield was unaffected by CCs in 2017 but lower in some WM treatments in 2018. In this study, tobacco herbicides did not interfere with wheat, cereal rye, or crimson clover establishment, but additional research should determine if these results apply to other environments and soil types. However, when these CC species were terminated 5 to 6 wk before transplanting, they did not consistently contribute to weed control.

Weed management in tobacco (Nicotiana tabacum L.) is accomplished primarily with soil-residual herbicides, cultivation, and hand removal. Management practices that reduce weed emergence, like reduced tillage and cover crop mulches, may improve weed management efficacy. Depending on cover-cropping goals, growers face trade-offs in species selection and management priorities—producing weed-suppressive mulches may lead to transplanting difficulties and soil-residual herbicide interception. Managing more complex cover crop mixtures may result in different challenges. We established on-farm trials across 4 site-years to study impacts of cover crop composition [wheat (Triticum aestivum L.) monoculture or mixture], termination treatment (early or late chemical termination or removing aboveground biomass), and soil-residual herbicides on weed density and biomass. The cover crop mixture contained cereal rye (Secale cereale L.), crimson clover (Trifolium incarnatum L.), and hairy vetch (Vicia villosa Roth.), with canola (Brassica napus L.) at 1 site-year. The mixture typically produced more biomass than monoculture wheat, although composition had few impacts on weed density or biomass. With residual herbicides, termination treatment had few impacts on weed density, suggesting that residues did not adversely affect herbicide efficacy. Without residual herbicides, early-season weed density was often higher following the late-terminated cover crop compared with other termination treatments, though midseason weed density was typically lower. When termination treatment affected final weed biomass, it was lower following late termination, with one exception—crop establishment was reduced at 1 site-year, leading to reduced weed–crop competition and greater weed biomass. Our results suggest that growers can use mixtures and, if well-timed to a rainfall event for incorporation, still effectively use soil-residual herbicides to maintain adequate weed control in tobacco regardless of how the cover crop is managed. Later termination, resulting in more residue, may lead to less weed biomass accumulation in the absence of herbicide use.

The Kulshan caldera formed at ∼1.15 Ma on the present-day site of Mt. Baker, Washington State, northwest USA and erupted a compositionally zoned (dacite-rhyolite) magma and a correlative eruptive, the Lake Tapps tephra. This tephra has previously been described, but only from the Puget Lowland of NW Washington. Here an occurrence of a Kulshan caldera correlative tephra is described from the Quaternary Palouse loess at the Washtucna site (WA-3). Site WA-3 is located in east-central Washington, ∼340 km southeast of the Kulshan caldera and ∼300 km east-southeast of the Lake Tapps occurrence in the Puget Lowland. Major- and trace element chemistry and location of the deposit at Washtucna within reversed polarity sediments indicates that it is not correlative with the Mesa Falls, Rockland, Bishop Ash, Lava Creek B or Huckleberry Ridge tephras. Instead the Washtucna deposit is related to the Lake Tapps tephra by fractional crystallisation, but is chemically distinct, a consequence of its eruption from a compositionally zoned magma chamber. The correlation of the Washtucna occurrence to the Kulshan caldera-forming eruption indicates that it had an eruptive volume exceeding 100 km3, and that its tephra could provide a valuable early-Pleistocene chronostratigraphic marker in the Pacific Northwest.

This paper describes the system architecture of a newly constructed radio telescope – the Boolardy engineering test array, which is a prototype of the Australian square kilometre array pathfinder telescope. Phased array feed technology is used to form multiple simultaneous beams per antenna, providing astronomers with unprecedented survey speed. The test array described here is a six-antenna interferometer, fitted with prototype signal processing hardware capable of forming at least nine dual-polarisation beams simultaneously, allowing several square degrees to be imaged in a single pointed observation. The main purpose of the test array is to develop beamforming and wide-field calibration methods for use with the full telescope, but it will also be capable of limited early science demonstrations.

Fibroblast growth factor (FGF)-2 (basic) is a potent angiogenic molecule involved in tumor progression, and is one of several growth factors with a central role in ovarian carcinogenesis. We hypothesized that common single nucleotide polymorphisms (SNPs) in the FGF2 gene may alter angiogenic potential and thereby susceptibility to ovarian cancer. We analyzed 25 FGF2 tgSNPs using five independent study populations from the United States and Australia. Analysis was restricted to non-Hispanic White women with serous ovarian carcinoma (1269 cases and 2829 controls). There were no statistically significant associations between any FGF2 SNPs and ovarian cancer risk. There were two nominally statistically significant associations between heterozygosity for two FGF2 SNPs (rs308379 and rs308447; p < .05) and serous ovarian cancer risk in the combined dataset, but rare homozygous estimates did not achieve statistical significance, nor were they consistent with the log additive model of inheritance. Overall genetic variation in FGF2 does not appear to play a role in susceptibility to ovarian cancer.

A useful result when dealing with backward stochastic differential equations is the comparison theorem of Peng (1992). When the equations are not based on Brownian motion, the comparison theorem no longer holds in general. In this paper we present a condition for a comparison theorem to hold for backward stochastic differential equations based on arbitrary martingales. This theorem applies to both vector and scalar situations. Applications to the theory of nonlinear expectations are also explored.

Studies have been carried out on the kinetics of light induced changes in thin films and solar cells fabricated in different laboratories. Unlike the many studies on undiluted a-Si:H films the kinetics reported here are for 1 sun illuminations carried out to the degraded steady state (DSS). The light induced changes at temperatures from 25 to 100°C were characterized with electron mobility lifetime products and subgap absorption in films and fill factors in solar cells. The significant changes in the degradation kinetics that occur in these materials in this t mperature range have further confirmed the importance of charged defects, not only in the annealed state, but also in the degraded steady states. Results are also presented on corresponding solar cell structures, which also indicate that defects other than neutral dangling bonds have to be taken into account if any meaningful evaluation is to be made of their solar cell properties.

Current concepts of the geological disposal of low- and intermediate-level radioactive wastes in the UK envisage the construction of a mined facility (incorporating cementitious engineered barriers) in chlorite-bearing rocks. To model accurately the fluid-rock reactions within the ‘disturbed zone’ surrounding a repository requires functions that describe mineral dissolution kinetics under pH conditions that vary from near neutral to highly alkaline.

Therefore, an experimental study to determine the dissolution rates of Fe-rich chlorite has been undertaken as part of the Nirex Safety Assessment Research Programme. Four experiments have been carried out at 25 °C and four at 70 °C, both sets using a range of NaCl/NaOH solutions of differing pH (of nominal pH 9.0,10.3, 11.6 and 13.0 [at 25 °C]).

Dissolution rates have been calculated and were found to increase with increasing pH and temperature. However, increased pH resulted in non-stoichiometric dissolution possibly due to preferential dissolution of part of the chlorite structure relative to another, or reprecipitation of some elements as thin hydroxide or oxyhydroxide surface coatings on the chlorite.

These results also show that chlorite dissolution is appreciably slower than that of albite and quartz at both 25 and 70 °C, but slightly faster than that of muscovite at 70 °C.

The effects of the Second World War on British colonial Africa were far-reaching and unexpected. Colonial governments, hitherto concerned with establishing and preserving law and order, with administration, the collection of taxation, and with slow economic development, had perforce to divert their energies into more varied and unorthodox channels. Troops were recruited from the colonies and in addition local Home Guards were formed. African colonies acquired a new strategic importance and became the scene for new intelligence and counter-espionage activities: this was especially so after the French government signed the armistice with Germany on 22 June 1940. On the following day the British government promised that if French overseas territories continued the war against the Nazis Britain would guarantee their integrity and economic stability, as well as providing funds to pay the salaries and pensions of French colonial officials. The governor of Nigeria, Sir Bernard Bourdillon, did much to encourage the rallitmcnt of neighbouring Vichy colonies to the Free French cause of General de Gaulle. It was part of French propaganda to exaggerate British involvement in the French colonies and to paint de Gaulle as simply a tool of British imperialism; and it was part of Britain's propaganda to minimize her own role and to stress that the British were simply supporting the initiatives of the Free French. As a result we have not yet been presented with a balanced and scholarly assessment of the contribution which the British in Nigeria made to the ralliement of French Equatorial Africa and the Cameroons in August 1940.
