Molecular techniques are an alternative for the diagnosis of strongyloidiasis, produced by Strongyloides stercoralis. However, it is necessary to determine the best amplification target for the populations of this parasite present in a geographical area and standardize a polymerase chain reaction (PCR) protocol for its detection. The objectives of this work were the comparison of different PCR targets for molecular detection of S. stercoralis and the standardization of a PCR protocol for the selected target with the best diagnostic results. DNA extraction was performed from parasite larvae by saline precipitation. Three amplification targets of the genes encoding ribosomal RNA 18S (18S rDNA) and 5.8S (5.8S rDNA) and cytochrome oxidase 1 (COX1) of S. stercoralis were compared, and the PCR reaction conditions for the best target were standardized (concentration of reagents and template DNA, hybridization temperature, and number of cycles). The analytical sensitivity and specificity of the technique were determined. DNA extraction by saline precipitation made it possible to obtain DNA of high purity and integrity. The ideal target was the 5.8S rDNA, since the 18S rDNA yielded non-reproducible results and COX1 never amplified under any condition tested. The optimal conditions for the 5.8S rDNA-PCR were: 1.5 mM MgCl2, 100 μM dNTPs, 0.4 μM primers, and 0.75 U DNA polymerase, using 35 cycles and a hybridization temperature of 60 °C. The analytical sensitivity of the PCR was 1 attogram of DNA, and the specificity was 100%. Consequently, the 5.8S rDNA was shown to be highly sensitive and specific for the detection of S. stercoralis DNA.

In the present work, we study the flow field around, and forces acting on, a circular cylinder with an attached flexible splitter plate/flap. Two cases of flap length ($L/D$), namely, $L/D = 5$ and $L/D = 2$ have been investigated focusing on the effect of variations in flap flexural rigidity, $EI$. We find that for a range of $EI$ and Reynolds numbers $Re = UD/\nu$, a non-dimensional bending stiffness $K^{\ast }=EI/((1/2)\rho U^2 L^3)$ collapses flap motion and forces on the system well, as long as $Re>5000$. In the $L/D = 5$ flap case, two periodic flap deformation regimes in the form of travelling waves are identified (modes I and II), with mode I occurring at $K^{\ast } \approx 1.5\times 10^{-3}$ and mode II at lower $K^{\ast }$ values ($K^{\ast }<3\times 10^{-5}$). In the $L/D = 2$ flap case, we find a richer set of flapping modes (modes A, B, C and D) that are differentiated by their flapping characteristics (symmetric/asymmetric and amplitude). Force measurements show that the largest drag reduction occurs in mode I ($L/D = 5$) and mode C ($L/D = 2$), which also correspond to the lowest lift and wake fluctuations, with the mode C wake fluctuations being lower than even the rigid splitter plate case. In contrast, the highest fluctuating lift, in both $L/D$ cases, occurs at higher $K^{\ast }$, when the wake frequency is close to the first structural bending mode frequency of the flap. The observed rich range of flap/splitter plate dynamics could be useful for applications such as drag reduction, vibration suppression, reduction of wake fluctuations and energy harvesting.

Using a multiple-scale analysis, Walker et al. (J. Fluid Mech., vol. 944, 2022, R2) obtain the long-time behaviour of a shape-changing swimmer in a Poiseuille flow. They show that the behaviour falls into one of three categories: endless tumbling at increasing distance from the midline of the flow; preserved initial behaviour of the swimmer; or convergence to upstream rheotaxis, where the swimmer is situated at the midline of the flow. Furthermore, a single swimmer-dependent constant is identified that determines which of the three behaviours is realised.

Supercritical pressure fluids are widely used in heat transfer and energy systems. The benefit of high heat transfer performance and the successful avoidance of phase change from the use of supercritical pressure fluids are well-known, but the complex behaviours of such fluids owing to dramatic thermal property variations pose strong challenges to the design of heat transfer applications. In this paper, the turbulent flow and heat transfer of supercritical pressure $\textrm {CO}_2$ in a small vertical tube influenced by coupled effects of buoyancy and thermal acceleration are numerically investigated using direct numerical simulation. Both upward and downward flows with an inlet Reynolds number of 3540 and pressure of 7.75 MPa have been simulated and the results are compared with corresponding experimental data. The flow and heat transfer results reveal that under buoyancy and thermal acceleration, the turbulent flow and heat transfer exhibit four developing periods in which buoyancy and thermal acceleration alternately dominate. The results suggest a way to distinguish the dominant factor of heat transfer in different periods and a criterion for heat transfer degradation under the complex coupling of buoyancy and thermal acceleration. An analysis of the orthogonal decomposition and the generative mechanism of turbulent structures indicates that the flow acceleration induces a stretch-to-disrupt mechanism of coherent turbulent structures. The significant flow acceleration can destroy the three-dimensional flow structure and stretch the vortices resulting in dissipation.

Analogy is a core cognition process used to produce inferences as well as new ideas using previous knowledge and experience. Ontology is a formal representation of a set of domain concepts and their relationships. The use of analogy and ontology in design activities to support design creativity have previously been explored. This paper explores an approach to construct ontologies with sufficient richness and coverage to support reasoning over real-world datasets for prompting creative idea generation. This approach has been implemented into a computational tool for assisting designers in generating creative ideas during the early stages of design. The tool, called “the Retriever”, has been developed based on ontology by embracing the aspects of analogical reasoning. A case study has indicated that the tool can be effective and useful for idea generation. The results have indicated that the tool, in its current formulation, can significantly improve the fluency and flexibility of idea generation and the usefulness of ideas, as well as slightly increase the originality of ideas, for the case study concerned.

The influence of calcium arsenate (48% w/w tricalcium arsenate) rate, physical form, time of application, and phosphorus (P) rate on the growth of annual bluegrass (Poa annua L.) and Bray P1 extractable arsenic (As) were investigated. Both powder and granular formulations reduced clipping yields and enhanced extractable As levels. Higher initial soil As activity and significantly better control were obtained in one study when the powder formulation was used. Similar levels of control were achieved whether the As was applied prior to germination or to the mature turf. Soil P-levels did not influence the effectiveness of either As formulation.

Flavonoids are natural compounds derived from fruits, vegetables and medicinal plants. These compounds have been reported to possess various biological actions including anti-inflammatory, antioxidant, growth promoter, antiviral, hepatoprotective, antibacterial, antiallergic, anticarcinogenic, antithrombotic and immunomodulator activities in various animals and poultry species. Quercetin is found in various foods such as vegetables, tea, fruits, wine apples and onions and is known to exert positive effects on poultry production and health. Since, quercetin enhances the immune system by stimulating lymphocytes, macrophages and IgY antibody production and improving natural killer cell activity, lymphoid organs (spleen, thymus and bursa) weights as well as activating the cytokines profile, its supplementation can lead to a state of immune alertness and a lower incidence of infections and diseases. In addition, it affects the dendritic cells (DCs) that play an important role in adaptive and innate immunity. Quercetin revokes the ability of lipopolysaccharide (LPS)-stimulated dendritic cells to induce specific T cell activation and reduce the cytotoxicity in both in vivo and in vitro studies, suggesting quercetin works as an immunosuppressive agent. As immune function is crucial for poultry productivity and prosperity, this review elucidates the potential nutritional effects and health benefits of quercetin on poultry as a replacer for traditional immune boosters and growth promoters. It includes the antiviral activity of quercetin against influenza A virus, rhinovirus (caused common cold), adenovirus (source of necrotic pancreatitis in broilers) and suggests its role in counteracting heavy metal toxicity. However, further studies on molecular basis are required to understand mode of action, beneficial applications as a natural feed additive, growth promoter and immune enhancer, beside the side effects of this compound in poultry.

Seismic profiles across the southwest end of Jeffreys Ledge, a bathymetric high north of Cape Ann, Massachusetts, reveal two end moraines. The moraines overlie upper Wisconsinan glacialmarine silty clay and are composed mostly of subaqueous ice-contact deposits and outwash. They were formed below sea level in water depths of as much as 120 m during fluctuations of a calving ice front. The moraines are late Wisconsinan in age and were formed after the Cambridge readvance, about 14,000 yr B.P., and before the Kennebunk readvance, about 13,000 yr B.P. They represent fluctuations of the ice front during overall retreat of Laurentide ice from the Gulf of Maine and New England.

Having accurate data for ileal amino acid digestibility of a feed ingredient is one of the ways to enhance protein and nitrogen utilisation, improve poultry feeding efficiency, and mitigate ammonia emission and pollution in the environment. Moreover, the precise estimation of endogenous amino acid losses (EAAL) depends on formulating diets on digestible amino acid (DAA) basis. Numerous methods have been reported to determine the endogenous fractions including the regression method, the protein-free diet, and the total digestible nitrogen diet. The EAAL determined by these techniques can help in calculated corrected values for amino acid digestibility, termed true digestibility. Certain dietary components that can be called ‘specific losses’ could lead to higher losses than the basal calculation would indicate. To determine the basal plus specific losses (total endogenous losses), some researchers suggested the 15N-dilution method, which allows the determination of the actual digestibility. This method needs more studies and evaluations because there are some controversial issues about this methodology, such as the validity of the 15N-dilution technique when applied to any amino acid or to total nitrogen, the reference pool for the estimation of the labelling of endogenous fractions/secretions, anti-nutritional agents, the effect of bird age, mucin, different cereal grains, as well as the marker itself and the proper assessment. This review will be focused on the above issues, finding that the recently reported 15N isotopes single injection method could be an easy, time-saving, consistent, and reliable methodology for EAAL estimation in poultry.
