OBJECTIVES/GOALS: Contingency management (CM) procedures yield measurable reductions in cocaine use. This poster describes a trial aimed at using CM as a vehicle to show the biopsychosocial health benefits of reduced use, rather than total abstinence, the currently accepted metric for treatment efficacy. METHODS/STUDY POPULATION: In this 12-week, randomized controlled trial, CM was used to reduce cocaine use and evaluate associated improvements in cardiovascular, immune, and psychosocial well-being. Adults aged 18 and older who sought treatment for cocaine use (N=127) were randomized into three groups in a 1:1:1 ratio: High Value ($55) or Low Value ($13) CM incentives for cocaine-negative urine samples or a non-contingent control group. They completed outpatient sessions three days per week across the 12-week intervention period, totaling 36 clinic visits and four post-treatment follow-up visits. During each visit, participants provided observed urine samples and completed several assays of biopsychosocial health. RESULTS/ANTICIPATED RESULTS: Preliminary findings from generalized linear mixed effect modeling demonstrate the feasibility of the CM platform. Abstinence rates from cocaine use were significantly greater in the High Value group (47% negative; OR = 2.80; p = 0.01) relative to the Low Value (23% negative) and Control groups (24% negative;). In the planned primary analysis, the level of cocaine use reduction based on cocaine-negative urine samples will serve as the primary predictor of cardiovascular (e.g., endothelin-1 levels), immune (e.g., IL-10 levels) and psychosocial (e.g., Addiction Severity Index) outcomes using results from the fitted models. DISCUSSION/SIGNIFICANCE: This research will advance the field by prospectively and comprehensively demonstrating the beneficial effects of reduced cocaine use. These outcomes can, in turn, support the adoption of reduced cocaine use as a viable alternative endpoint in cocaine treatment trials.

Core KM-3 at Searles Lake, California, comprises 693.4 m of lacustrine sediments deposited over the past 3.2 m.y. The lake water evolved from moderately saline, slightly alkaline, and dominated by Na, Ca, Cl, SO4, and HCO3 + CO3 to a highly alkaline brine dominated by Na and CO, ions. Sediments are chiefly muds and evaporites. Montmorillonite and illite are the principal detrital clay minerals supplied to Searles Lake at present and probably during the late Pliocene and Pleistocene.

The drill core is divided into three diagenetic zones on the basis of clay-mineral reactions. The upper zone (0-291.1 m) contains authigenic Fe-illite, Mg-smectite, K-feldspar, and analcime, which average 60–70% of the <2-μm silicate fraction of mud samples. The principal silicate reactants are detrital montmorillonite and kaolinite, which have been largely consumed. The middle zone (291.1–541.6 m) also contains authigenic Fe-illite, Mg-smectite, K-feldspar, and analcime, but they form only 20 to 30% of the <2-μm silicate fraction, and considerable detrital montmorillonite and kaolinite remain unaltered. In muds of the lower zone (541.6–693.4 m), a small amount of clinoptilolite is the only authigenic silicate mineral identified, although authigenic montmorillonite probably coexists here as well. Vitric ash is the silicate reactant, and detrital clay minerals apparently remain unaltered in the lower zone.

Diagenetic zoning reflects the pore-water chemistry, in which pH may have been the most important parameter. Sediments of the upper zone were deposited in highly alkaline lakes of variable salinity, and sinking brines with a pH of 9.0–10.0 have saturated all sediments. Sediments of the middle zone were deposited in lakes of moderate to high salinity. The pH was overall lower than in the upper zone, although it probably exceeded 9.0, at least locally, during silicate diagenesis. Moderate salinity and a slightly alkaline pH (~ 7.5–8.0) are inferred for both the lake and pore water of the lower zone.

Oxygen-isotope values of authigenic Fe-illite, Mg-smectite, and K-feldspar, and phillipsite in the upper diagenetic zone reflect a high degree of evaporative concentration and presumably of salinity. Equilibrium water values calculated for 22°C from the oxygen-isotopic composition of authigenic phyllosilicates range from −3.5 to +1.9%, averaging −1.2% (SMOW). Higher salinities are suggested for K-feldspar, for which water values range from +1.8 to +4.8%, averaging +3.4‰ The water value for phillipsite is +0.3‰ By comparison, rainfall at Searles Lake has an average δ18O value of about −9.8%, and a brine sample has a value of +4.0‰

Mud samples of the middle and upper diagenetic zones tend to be rich in Fe-illite or Mg-smectite, but not in both, indicating that the two minerals are favored to some extent by different environments. Fe-illite seems to be generally favored by oxidizing conditions and probably by a playa environment, and Mg-smectite seems to be favored by reducing conditions and an open-water environment. One strati-graphic unit of the upper zone does not fit this pattern and contains abundant Fe-illite in sediments of a deep perennial lake.

The Sinya Beds of the Amboseli Basin in Tanzania and Kenya consist largely of carbonate rocks and Mg-rich clays that are intensely deformed where exposed in and near former meerschaum mines. The carbonate rocks consist of limestone and dolomite in Tanzania, but only dolomite has been identified in Kenya. Sepiolite and mixed-layered kerolite/stevensite (Ke/St) are subordinate constituents of the carbonate rocks. The carbonate rocks and overlying bedded sepiolite were deposited in a semiarid lake basin at the foot of the large volcano Kilimanjaro. Calcite and dolomite of the carbonate rocks have δ18O values 4–6‰ lower than calcite and dolomite of the late Pleistocene Amboseli Clays, suggesting that the Sinya Beds were deposited in the middle or early Pleistocene under a different climatic regime when meteoric water had lower δ18O values than at present.

Mg-rich clay minerals form veins and fill cavities in the Sinya Beds. The principal clay minerals are sepiolite and Ke/St, some of which contains substantial Al and Fe (Al-Ke/St). NEWMOD® modeling and other X-ray diffraction (XRD) data suggest that most of the Ke/St contains 25–50 percent kerolite layers, but minor amounts of kerolite-rich Ke/St are present in some samples. Illite with an inferred high content of Fe or Mg is a minor constituent of the samples with Al-Ke/St. The cavity-filling clays were chemically precipitated, as shown by field relationships and SEM study. The early-deposited clays of veins and cavities are principally Ke/St with minor sepiolite, and the latest clay is sepiolite (meerschaum), generally with minor Ke/St.

The δ18O values of cavity-filling Ke/St range from 22.5–25.6‰ and correlate with mineral composition, with the highest values associated with the highest content of stevensite and the lowest values with the highest content of kerolite. This relation suggests that high salinities favored stevensite and low salinities favored kerolite. δ18O values of sepiolite (meerschaum) fall in the middle of the range for Ke/St, suggesting that salinity was not the main control on sepiolite precipitation. High values of $a_{Si{O}_2}/a_{M{g}^{2+}}$ may have been a major factor in sepiolite precipitation.

Different mixtures of dilute ground water and saline, alkaline lake water in pore fluids may largely account for the differences in clay mineralogy of cavity-filling clays. Sepiolite is the dominant clay mineral in lacustrine sediments of the Amboseli Basin, and the cavity-filling sepiolite may reflect a high proportion of lake water. The low-Al Ke/St may have formed from fluids with a higher proportion of ground water. Detrital clay was very likely a factor in forming the Al-Ke/St, for which δ18O values suggest a saline environment.

Recent research has shown the potential of speleothem δ13C to record a range of environmental processes. Here, we report on 230Th-dated stalagmite δ13C records for southwest Sulawesi, Indonesia, over the last 40,000 yr to investigate the relationship between tropical vegetation productivity and atmospheric methane concentrations. We demonstrate that the Sulawesi stalagmite δ13C record is driven by changes in vegetation productivity and soil respiration and explore the link between soil respiration and tropical methane emissions using HadCM3 and the Sheffield Dynamic Global Vegetation Model. The model indicates that changes in soil respiration are primarily driven by changes in temperature and CO2, in line with our interpretation of stalagmite δ13C. In turn, modelled methane emissions are driven by soil respiration, providing a mechanism that links methane to stalagmite δ13C. This relationship is particularly strong during the last glaciation, indicating a key role for the tropics in controlling atmospheric methane when emissions from high-latitude boreal wetlands were suppressed. With further investigation, the link between δ13C in stalagmites and tropical methane could provide a low-latitude proxy complementary to polar ice core records to improve our understanding of the glacial–interglacial methane budget.

The IntCal family of radiocarbon (14C) calibration curves is based on research spanning more than three decades. The IntCal group have collated the 14C and calendar age data (mostly derived from primary publications with other types of data and meta-data) and, since 2010, made them available for other sorts of analysis through an open-access database. This has ensured transparency in terms of the data used in the construction of the ratified calibration curves. As the IntCal database expands, work is underway to facilitate best practice for new data submissions, make more of the associated metadata available in a structured form, and help those wishing to process the data with programming languages such as R, Python, and MATLAB. The data and metadata are complex because of the range of different types of archives. A restructured interface, based on the “IntChron” open-access data model, includes tools which allow the data to be plotted and compared without the need for export. The intention is to include complementary information which can be used alongside the main 14C series to provide new insights into the global carbon cycle, as well as facilitating access to the data for other research applications. Overall, this work aims to streamline the generation of new calibration curves.

Understanding seed moisture desorption and adsorption isotherms is important for seed quality maintenance and better predicting seed storage lifespan. Freshly harvested oilseed rape and barley seeds were dried and then rehydrated twice. Seed equilibrium relative humidity (eRH) and moisture content (MC) were determined at different humidity levels so that two cycles of desorption and adsorption could be constructed. In addition, seeds were dried to 30% RH and then rehydrated to 50% RH for five cycles to determine whether they shift to the adsorption isotherm. Monolayer MC was determined using the Gugenheim-Anderson-de Boer model. Storage experiments were conducted for seeds equilibrated at 30, 40, 50, 60 and 70% RH for two cycles of desorption and adsorption at 45°C. Isotherm curves’ shapes were similar for oilseed rape and barley, although spanning a greater MC range in barley. The hysteresis effect was observed for oilseed rape and barley seeds when dried over silica gel at <10% RH. However, this effect was only observed for barley seeds when dried to 30% RH, but not for oilseed rape seeds. Longevity was greater for adsorbing seeds than desorbing seeds at a given eRH, however, there was no significant difference in σ (the standard deviation of the normal distribution of seed deaths over time)–MC log–log relationship. The relationship shifted for seeds on the second cycle. In conclusion, if seed lots are stored at a specific RH, reaching equilibrium by desorption or adsorption can strongly influence their longevity. Also, when seeds of different species are dried to low RH, they will respond differently to a subsequent increase in RH, which could profoundly affect their longevity.

New stalagmites from Qadisha Cave (Lebanon) located at 1720 m above sea level provide a high-resolution and well-dated record for northern Mount Lebanon. The stalagmites grew discontinuously from 9.2 to 5.7 and at 3.5 ka, and they show a tendency to move from a more negative oxygen isotope signal at ~9.1 ka to a more positive signal at ~5.8 ka. Such a trend reflects a change from a wetter to a drier climate at high altitudes. The δ13C signal shows rapid shifts throughout the record and a decreasing trend toward more negative values in the mid-Holocene, suggesting enhanced soil activity. In the short-term trend, Qadisha stalagmites record rapid dry/wet changes on centennial scales, with a tendency to more rapid dry events toward the mid-Holocene. Such changes are characterized by overall good agreement between both geochemical proxies and stalagmite growth and might be affected by the seasonal variations in snow cover. The Qadisha record is in good agreement with other Levantine records, showing more humid conditions from 9 to 7 ka. After 7 ka, a drier climate seems to affect sites at both low- and high-altitude areas. The Qadisha record reflects uniquely mountainous climate characteristics compared with other records, specifically the effect of snow cover and its duration regulating the effective infiltration.

The 4.2 ka event is widely presumed to be a globally widespread aridity event and has been linked to several episodes of societal changes across the globe. Whether this climate event impacted the cultural development in south-central China remains uncertain due to a lack of regional paleorainfall records. We present here stalagmite stable carbon isotope and trace element–based reconstruction of hydroclimatic conditions from south-central China. Our data reveal a sub–millennial scale (~5.6 to 4.3 ka) drying trend in the region followed by a gradual transition to wetter conditions during the 4.2 ka event (4.3–3.9 ka). Together with the existing archaeological evidence, our data suggest that the drier climate before 4.3 ka may have promoted the Shijiahe culture, while the pluvial conditions during the 4.2 ka event may have adversely affected its settlements in low-lying areas. While military conflicts with the Wangwan III culture may have accelerated the collapse of Shijiahe culture, we suggest that the joint effects of climate and the region's topography also played important causal roles in its demise.

Understanding the relative longevity of different seed lots, perhaps of different species or genotypes, but also following production under different environments or using different cultivation methods, or following different post-harvest treatments, is relevant to anyone concerned with the retention of seed lot viability and vigour during storage. However, different scientists over the years have used different conditions to assess seed lot longevity, as well as different variables as the measure of ‘longevity.’ Here, we give some of the backgrounds to how two standard protocols, with an open and closed system respectively, were derived, and explain why we consider p50, defined as the time during storage when seed lot viability, as measured through a germination test, has declined to 50%, is a suitable longevity trait parameter.

This paper introduces a formal definition of a Cyber-Physical System (CPS) in the spirit of the CPS Framework proposed by the National Institute of Standards and Technology (NIST). It shows that using this definition, various problems related to concerns in a CPS can be precisely formalized and implemented using Answer Set Programming (ASP). These include problems related to the dependency or conflicts between concerns, how to mitigate an issue, and what the most suitable mitigation strategy for a given issue would be. It then shows how ASP can be used to develop an implementation that addresses the aforementioned problems. The paper concludes with a discussion of the potentials of the proposed methodologies.

Fatty acid (FA) levels and profiles are vital for soybean oil quality, while cytokinins (CKs) and abscisic acid (ABA) are potent regulators of plant growth and development. Previous research suggested associations between FA biosynthesis and hormonal signalling networks; however, hormonal regulation of FA accumulation during soybean (Glycine max) seed maturation has never been measured. We analysed hormone and FA profiles obtained from HPLC-(ESI)-MS/MS and GC-FID screening during soybean seed maturation. A multilayered data processing approach, involving heat-maps, principal component analysis (PCA), correlation and multiregression models, suggested a strong relationship between hormone metabolism and FA/oil accumulation during seed maturation. Most strikingly, positive correlations were found between the levels of CK ribosides [transZeatin riboside (tZR), N6-isopentenyladenosine (iPR)] at the early stages of SM (R5-R6) and C18:0, C18:2 and oil content at the R8 stage. Moreover, multiple regression models revealed functional linkages between several CK derivatives and FA and oil content in mature seeds. To further test the significance of hormone regulation in FA metabolism, plants of two soybean accessions with contrasting hormone and FA profiles were sprayed with exogenous ABA and transZeatin (tZ) during the seed-filling period (R5-R6). Depending on the hormone type and concentration, these treatments distinctly modified biosynthesis of all tested FAs, except for C18:0. Most remarkably, tZ (50 nM) promoted production of C16:0, C18:1, C18:2, C18:3, and oil accumulation in maturing seeds. Overall, the results indicate impactful roles for ABA and CKs in FA accumulation during SM and represent a further step towards understanding FA biosynthesis, and potential improvements of soybean oil profiles.

This study examines the first precisely dated and temporally highly resolved speleothem record from Iberia that reconstructs the Oldest Dryas (OD). The onset of cold conditions in the study area, contemporary with the beginning of Heinrich Stadial 1, is recorded at 18.13 ± 0.08 ka, with a pronounced drop of 6.1‰ in δ13C in 250 years. Henceforth, stadial conditions depict a period of instability in the Atlantic Meridional Overturning Circulation, peaking in freshwater input from iceberg melting during Heinrich Event 1. Anomalies in the δ18O of the stalagmite attributed to such a freshwater event are found from 16.17 to 15.89 ka. Such absolute dates given to the onset of the OD in Iberia and to the main iceberg discharges are reliable anchor points for non-absolute chronologies. Two periods are identified in the OD: OD-a (18.13–16.17 ka) is characterized by wet conditions and a faster growth rate, and OD-b (15.89–14.81 ka) exhibits relative dryness and a slower growth rate. The sudden release of fresh water is considered to be the reason for the disruption of rainfall patterns in eastern Iberia. The present study also highlights the existence of heterogeneous and complex hydrological conditions during the OD in Iberia when both Atlantic and Mediterranean realms are considered.

Quantifying tree biomass is an important research and management goal across many disciplines. For species that exhibit predictable relationships between structural metrics (e.g. diameter, height, crown breadth) and total weight, allometric calculations produce accurate estimates of above-ground biomass. However, such methods may be insufficient where inter-individual variation is large relative to individual biomass and is itself of interest (for example, variation due to herbivory). In an East African savanna bushland, we analysed photographs of small (<5 m) trees from perpendicular angles and fixed distances to estimate above-ground biomass. Pixel area of trees in photos and diameter were more strongly related to measured, above-ground biomass of destructively sampled trees than biomass estimated using a published allometric relation based on diameter alone (R2 = 0.86 versus R2 = 0.68). When tested on trees in herbivore-exclusion plots versus unfenced (open) plots, our predictive equation based on photos confirmed higher above-ground biomass in the exclusion plots than in unfenced (open) plots (P < 0.001), in contrast to no significant difference based on the allometric equation (P = 0.43). As such, our new technique based on photographs offers an accurate and cost-effective complement to existing methods for tree biomass estimation at small scales with potential application across a wide variety of settings.

Radiocarbon (14C) ages cannot provide absolutely dated chronologies for archaeological or paleoenvironmental studies directly but must be converted to calendar age equivalents using a calibration curve compensating for fluctuations in atmospheric 14C concentration. Although calibration curves are constructed from independently dated archives, they invariably require revision as new data become available and our understanding of the Earth system improves. In this volume the international 14C calibration curves for both the Northern and Southern Hemispheres, as well as for the ocean surface layer, have been updated to include a wealth of new data and extended to 55,000 cal BP. Based on tree rings, IntCal20 now extends as a fully atmospheric record to ca. 13,900 cal BP. For the older part of the timescale, IntCal20 comprises statistically integrated evidence from floating tree-ring chronologies, lacustrine and marine sediments, speleothems, and corals. We utilized improved evaluation of the timescales and location variable 14C offsets from the atmosphere (reservoir age, dead carbon fraction) for each dataset. New statistical methods have refined the structure of the calibration curves while maintaining a robust treatment of uncertainties in the 14C ages, the calendar ages and other corrections. The inclusion of modeled marine reservoir ages derived from a three-dimensional ocean circulation model has allowed us to apply more appropriate reservoir corrections to the marine 14C data rather than the previous use of constant regional offsets from the atmosphere. Here we provide an overview of the new and revised datasets and the associated methods used for the construction of the IntCal20 curve and explore potential regional offsets for tree-ring data. We discuss the main differences with respect to the previous calibration curve, IntCal13, and some of the implications for archaeology and geosciences ranging from the recent past to the time of the extinction of the Neanderthals.

Vitamin E is known to scavenge lipid peroxy radicals and has a purported role in preventing seed deterioration during storage. In our previous studies using 20 rice varieties from different variety groups, the specific ratio of vitamin E homologues rather than total vitamin E content was associated with seed longevity. To validate this result, we extended the experiment to a rice panel composed of 185 Aus (semi-wild rice) varieties. Seed longevity values were determined through storage experiments at 45°C and 10.9% seed moisture content (MC). Eight types of vitamin E homologues (α-, β-, γ- and δ-tocopherol/tocotrienol) were quantified by ultra-performance liquid chromatography. The theoretical initial viability in NED, Ki, was positively correlated with γ- and δ-tocopherols and negatively correlated with α-tocotrienol. The time for viability to fall to 50% during storage at elevated temperature and relative humidity, p50, was positively correlated with δ-tocopherol. The harvest MC was negatively correlated with all seed longevity traits. Taking this factor into account in a genome-wide association (GWA) analysis, we were able to correct false positives. A consistent major peak on chromosome 4 associated with −σ−1 was detected with a mixed linear analysis. Based on rice genome annotation and gene network ontology databases, we suggest that RNA modification, oxidation–reduction, protein–protein interactions and abscisic acid signal transduction play roles in seed longevity extension of Aus rice. Although major GWA regions were not overlapped across traits, three genetic markers, on chromosomes 1, 3 and 4, were associated with both δ-tocopherol and Ki and two markers on chromosome 1 and 8 were associated with both δ-tocopherol and p50.
