Acanthamoeba spp. are widely distributed in the environment and cause serious infections in humans. Treatment of Acanthamoeba infections is very challenging and not always effective which requires the development of more efficient drugs against Acanthamoeba spp. The purpose of the present study was to test medicinal plants that may be useful in the treatment of Acanthamoeba spp. Here we evaluated the trophozoital and cysticidal activity of 13 flavonoid glycosides isolated from Delphinium gracile, D. staphisagria, Consolida oliveriana and from Aconitum napellus subsp. Lusitanicum against the amoeba Acanthamoeba castellanii. AlamarBlue Assay Reagent® was used to determine the activity against trophozoites of A. castellanii, and cytotoxic using Vero cells. Cysticidal activity was assessed on treated cysts by light microscopy using a Neubauer chamber to quantify cysts and trophozoites. Flavonoids 1, 2, 3 and 4 showed higher trophozoital activity and selectivity indexes than the reference drug chlorhexidine digluconate. In addition, flavonoid 2 showed 100% cysticidal activity at a concentration of 50 μm, lower than those of the reference drug and flavonoid 3 (100 μm). These results suggest that flavonoids 2 and 3 might be used for the development of novel therapeutic approaches against Acanthamoeba infections after satisfactory in vivo evaluations.

Sperm capacitation includes the reorganization of plasma membrane components and the outstanding modification of the glycocalyx. The α-mannose presence and location during in vitro capacitation have been commonly described in human spermatozoa using Concanavalin A (Con A) lectin. However, it is still unclear to date how in vitro capacitation time affects the α-mannose residues and their topographic spatial distribution on sperm membrane. Here, we characterized the α-mannose density and specific membrane domain locations before and after in vitro capacitation (1–4 h) using high-resolution field emission scanning electron microscopy (FE-SEM). Results showed that α-mannose residues were present preferably on the acrosome domains for all physiological conditions. Uncapacitated sperm comparatively exhibits significant highest labeling densities of α-mannose residues. In addition, as in vitro capacitation takes place, significant and progressive decreasing of sugar residues was combined with their relocation mostly affecting acrosomal domain apical areas. Our findings reveal that combined approach using FE-SEM and gold nanoparticle topographical mapping offers new human sperm biomolecular and structural details during capacitation events.

Insects in the subfamily Ischnorhininae (Hemiptera: Auchenorrhyncha: Cercopidae), known as spittlebugs or froghoppers, are mainly distributed in the Neotropical region. A few genera include pest species of economic relevance, including Prosapia Fennah, 1949. Two new species from Mexico and Costa Rica are now described for this genus, and a key to species is proposed for the P. inferens (Walker, 1858) species group.

The aim of this research was to study the learning process using an objective and computerized task. The performance of 466 schoolchildren aged between 6 and 11 in a category learning task, the Category Learning Test (CLT), was examined. The results showed evidence of category learning throughout the trials for the whole sample, F(7, 469) = 29.979, p <.001. In addition, categorization performance improved with age, H(2) = 48.475, p <.001. However, there were old children that struggled with the task and young children that performed very well. The ability to learn the categories was related to the children’s behavior when trying to solve the task: the response speed (r = –.217, p <.01) and the organization index (r = .247, p <.01). Nevertheless, performance in the task and academic marks were not related. We discuss the impact of these findings on the promotion and improvement of learning in schools: an intervention to promote slowness and organization might help some children to learn.

The aim of this work was to study how machine milking (MM) carried out in suitable conditions affects teat wall thickness and teat canal length and their return after milking to pre-milking conditions, in comparison with other milk extraction methods considered biological referents: lamb suckling (LS), milk removal by catheter (RC) and hand milking (HM). Three Latin square experiments were designed, using 11 ewes in the first experiment (LS), 13 in the second (RC) and 12 in the third (HM). Each of the Latin squares was divided into two periods: in the first, the left gland of each animal was machine milked and the corresponding treatment (LS, RC and HM) was applied to the right gland. Subsequently, in the second period the extraction methods were interchanged. During the experimental period, 4 sampling days were carried out (2 in each experimental period), where ultrasound scans were taken before (B) and immediately after milking (A) and at 1 (1 h), 2 (2 h), 3 (3 h), 4 (4 h), 6 (6 h), 8 (8 h) and 10 (10 h) hours after milking finished. Teat wall thickness (TWT), teat wall area (TWA), teat end area (TEWA) and teat canal length (TCL) were measured in all the ultrasound images. MM increased TWT after milking compared with RC. TWT, TWA, TEWA and TCL were lower (P<0·05) in HM than in MM. No significant differences (P>0·05) were found between LS and MM for any variable. The extraction method affected the recovery time of the variables, with total teat recovery at 6 h after RC and 4 h after HM. In the case of LS, the TEWA and TCL values were recovered sooner, as of 3 h. Teat recovery time after MM was similar to the extraction method with which it was compared in each experiment. Thus, considering the similar increase in wall thickness and their recovery time compared with the reference methods, it was concluded that machine milking, carried out in optimum conditions and respecting the time interval between milkings usually applied on sheep farms (8–12 h), would not affect teat integrity. Moreover, given the variability observed in teat thickness recovery time between the different experiments, further research should be carried out to study which factors intrinsic and extrinsic to the animal may affect the teat wall thickness and recovery time after machine milking.

The aim of this work was to study the effect on teat wall thickness and canal length in sheep and goats of overmilking for 2 min (OM+2) and of milking with used (AL; +3000 milkings) and twisted (TL; 45°) liners in sheep and goats, as well as the effect of milking goats with liners designed for sheep (SL, shorter length and diameter than liners for goats). To this end, we performed four experiments in goats and three in sheep, in a Latin square design with two experimental periods. During the experimental period 4 controls were carried out, performing ultrasound scans before and immediately after milking to determine the teat wall thickness (TWT), teat wall area (TWA), teat end area (TEWA) and teat canal length (TCL). OM+2 caused a significant increase in TWT, TWA, TEWA and TCL in goats and in TWA, TEWA and TCL in sheep. Liner features had a strong influence on the variables studied; aged liners caused significant changes in TWT and TCL in goats and in TWT in sheep; twisted liners produced a significant effect on the increase of TWT and TCL in goats, without reaching significance level in sheep; and milking goats with sheep liners led to a significant increase in TWT, TWA, TEWA and TCL. In practice, it is therefore important to avoid overmilking and the use of worn-out liners. It is also necessary to use liners designed for the morphological features of each species, taking special care to carry out periodic liner positioning revisions to ensure the benefits of pulsation on the teat end. Finally, it would be necessary to carry out long-term experiments to study whether the increase in thickness observed in some experiments is sufficient to affect milking efficiency and mammary gland health status.

In this work, we study CdTe thin films used in CdTe/CdS solar cells with a substrate configuration, which allows for better control in forming the junction, and the possibility for using flexible non-transparent substrates. We studied the properties of CdTe films grown at 450° and 550°C, with and without a CuxTe layer, and before and after CdCl2 treatment. We analyzed the structural and electro-optical properties using electron backscatter diffraction (EBSD), cathodoluminescence (CL) and X-ray diffraction (XRD), and investigated how the film structure, stress, and defect structure changes with the different growth conditions.

The present study analysed the effects of the flavanol ( − )-epicatechin in rats after chronic inhibition of NO synthesis with NG-nitro-l-arginine methyl ester (l-NAME), at doses equivalent to those achieved in the studies involving human subjects. Wistar rats were randomly divided into four groups: (1) control-vehicle, (2) l-NAME, (3) l-NAME-epicatechin 2 (l-NAME-Epi 2) and (4) l-NAME-epicatechin 10 (l-NAME-Epi 10). Rats were daily given by oral administration for 4 weeks: vehicle, ( − )-epicatechin 2 or 10 mg/kg. Animals in the l-NAME groups daily received l-NAME 75 mg/100 ml in drinking-water. The evolution in systolic blood pressure and heart rate, and morphological and plasma variables, proteinuria, vascular superoxide, reactivity and protein expression at the end of the experiment were analysed. Chronic ( − )-epicatechin treatment did not modify the development of hypertension and only weakly affected the endothelial dysfunction induced by l-NAME but prevented the cardiac hypertrophy, the renal parenchyma and vascular lesions and proteinuria, and blunted the prostanoid-mediated enhanced endothelium-dependent vasoconstrictor responses and the cyclo-oxygenase-2 and endothelial NO synthase (eNOS) up-regulation. Furthermore, ( − )-epicatechin also increased Akt and eNOS phosphorylation and prevented the l-NAME-induced increase in systemic (plasma malonyldialdehyde and urinary 8-iso-PGF2α) and vascular (dihydroethidium staining, NADPH oxidase activity and p22phox up-regulation) oxidative stress, proinflammatory status (intercellular adhesion molecule-1, IL-1β and TNFα up-regulation) and extracellular-signal-regulated kinase 1/2 phosphorylation. The present study shows for the first time that chronic oral administration of ( − )-epicatechin does not improve hypertension but reduced pro-atherogenic pathways such as oxidative stress and proinflammatory status of the vascular wall induced by blockade of NO production.

Phenolic compounds are one of the main reasons behind the healthy properties of virgin olive oil (VOO). However, their daily intake from VOO is low compared with that obtained from other phenolic sources. Therefore, the intake of VOO enriched with its own phenolic compounds could be of interest to increase the daily dose of these beneficial compounds. To evaluate the effectiveness of enrichment on their bioavailability, the concentration of phenolic compounds and their metabolites in human plasma (0, 60, 120, 240 and 300 min) from thirteen healthy volunteers (seven men and six women, aged 25 and 69 years) was determined after the ingestion of a single dose (30 ml) of either enriched virgin olive oil (EVOO) (961·17 mg/kg oil) or control VOO (288·89 mg/kg oil) in a cross-over study. Compared with VOO, EVOO increased plasma concentration of the phenol metabolites, particularly hydroxytyrosol sulphate and vanillin sulphate (P < 0·05). After the consumption of VOO, the maximum concentration of these peaks was reached at 60 min, while EVOO shifted this maximum to 120 min. Despite these differences, the wide variability of results indicates that the absorption and metabolism of olive oil phenols are highly dependent on the individual.

Chalcopyrite solar cells based on CuInSe2 and associated alloys have demonstrated high efficiencies, with current annual shipments in the hundreds of megawatts (MW) range and increasing. Largely due to concern over possible indium (In) scarcity, a related set of materials, the kesterites, which comprise Cu2ZnSnS4 and associated alloys, has received increasing attention. Similarities and differences between kesterites and chalcopyrites are discussed as drawn from theory, depositions, and materials characterization. In particular, we discuss predictions from density functional theory, results from vacuum co-evaporation, and characterization via x-ray diffraction, scanning electron microscopy, electron beam-induced current, quantum efficiency, secondary ion mass spectroscopy, and luminescence.

Metamorphic triple-junction solar cells can currently attain efficiencies as high as 41.1%. Using additional junctions could lead to efficiencies above 50%, but require the development of a wide bandgap (2.0-2.2eV) material to act as the top layer. In this work we demonstrate wide bandgap InyGa1-yP grown on GaAsxP1-x via solid source molecular beam epitaxy. Unoptimized tensile GaAsxP1-x buffers grown on GaAs exhibit asymmetric strain relaxation, along with formation of faceted trenches 100-300 nm deep in the [01-1] direction. Smaller grading step size and higher substrate temperatures minimizes the facet trench density and results in symmetric strain relaxation. In comparison, compressively-strained graded GaAsxP1-x buffers on GaP show nearly-complete strain relaxation of the top layers and no evidence of trenches. We subsequently grew InyGa1-yP layers on the GaAsxP1-x buffers. Photoluminescence and transmission electron microscopy measurements show no indication of phase separation or CuPt ordering. Taken in combination with the low threading dislocation densities obtained, MBE-grown InyGa1-yP layers are promising candidates for future use as the top junction of a multi-junction solar cell.

The nurse effect is a positive interaction in which one plant (the nurse) provides conditions that enhance the establishment and growth of another plant species (Callaway 1995). Increased environmental severity appeared to increase the strength of nurse effects (Brooker et al. 2008, Lortie & Callaway 2006). On the one hand, the impact of the nurse effect depends on the magnitude of the environmental changes exerted by the nurse plant. On the other hand, the impact could depend on the number of plant species in the regional pool that respond to such changes. For example, better conditions beneath the crowns of nurse plants might allow the occurrence of species that are sensitive to environmental stress and that occur infrequently in open areas. Thus, if a nurse plant modulates environmental conditions that are critical for the persistence of other plant species, it seems likely that such nurse plants would have greater effects in stressful habitats, where they cause relatively larger environmental mitigation (Badano et al. 2006, Callaway et al. 2002).
