The aim of the present work was to evaluate the distribution of the different clones of the parasite prevailing after treatment with benznidazole (BZ) and clomipramine (CLO), in mice infected with Trypanosoma cruzi, Casibla isolate which consists of a mixture of two discrete typing units (DTUs). Albino Swiss mice were infected and treated with high and low concentrations of BZ (100 or 6.25 mg/kg), CLO (5 or 1.25 mg/kg), or the combination of both low doses (BZ6.25 + CLO1.25), during the acute phase of experimental infection. Treatment efficacy was evaluated by comparing parasitaemia, survival and tissular parasite presence. For DTUs genotyping, blood, skeletal and cardiac muscle samples were analysed by multiplex quantitative polymerase chain reaction. The combined treatment had similar outcomes to BZ6.25; BZ100 was the most effective treatment, but it failed to reach parasite clearance and produced greater histological alterations. Non-treated mice and the ones treated with monotherapies showed both DTUs while BZ6.25 + CLO1.25 treated mice showed only TcVI parasites in all the tissues studied. These findings suggest that the treatment may modify the distribution of infecting DTUs in host tissues. Coinfection with T. cruzi clones belonging to different DTUs reveals a complex scenario for the treatment of Chagas disease and search for new therapies.

Optimizing interfaces between photosynthetic natural photoconverters, like photosynthetic bacterial Reaction Centers (RCs) and electrode surfaces represents a challenge in the progress of bio-optoelectronic devices. The features of the surfaces may result detrimental for the tertiary and quaternary structures of the RC, even resulting in the denaturation of the enzyme. Functional surfaces possessing both confinement capability and conductive features able to preserve the conformation of the biomolecule and its bioelectronic behaviours are highly needed. In this work, the RC is adsorbed on diatomaceous silica and plasma treated hydrophobic silicon based materials. Both the materials are demonstrated to be able to preserve and enhance the RC photoconverting activity. In particular, we evaluate the functioning of isolated bacterial RC interacting with flat pSi electrode through two nanotextured interfaces designed to address the RC: a thin conductive silicon film nanotextured in pillars via plasma treatment, and a cast film of nanostructured dielectric biosilica obtained from diatomaceous earth. The characterization of these interfaces, together with the RC photocurrent production measurements, pave the way to new generation RC based bio-devices for photocurrent investigation.

Sunlight is the most environmental friendly energy source available on Earth; many efforts devoted to design artificial photoconversion systems are ongoing, nevertheless they are still expensive and poorly efficient. Photoconversion devices made with organic-biological hybrids, or biohybrids, based on the photosynthetic reaction center (RC) have been introduced. In these systems, the photoenzyme is garnished with artificial antennas to enhance the photoactivity of the RC. Here we present a newly synthesized heptamethine cyanine dye that fulfills requisites to act as efficient RC light harvesting antenna.

The chemical decoration of biosilica shells (frustules) from microalgae with several classes of organic molecules is a convenient, scalable biotechnological route to silica nanostructures with applications ranging from photonics to biomedicine. Here we report for the first time the in vivo staining of Thalassiosira weissflogii diatoms with a two photon red emitting triphenylamine-based fluorescent dye bearing a triethoxysilyl functional group (tPhA-Silane). In vivo staining of the cells has been investigated with confocal microscopy and hybrid silica structures comprising the dye embedded into the biosilica have been isolated by proper protocols able to remove the organic protoplasm.

Diatoms microalgae represent a natural source of highly porous biosilica shells (frustules) with promising applications in a wide range of technological fields. Functionalization of diatoms’ frustules with tailored luminescent molecules can be envisaged as a convenient, scalable biotechnological route to new light emitting silica nanostructured materials. Here we report a straightforward protocol for the in vivo modification of Thalassiosira weissflogii diatoms’ frustules with a red emitting organic dye based on thienyl, benzothiadiazolyl and phenyl units. The metabolic insertion of the dye molecules into the diatoms shells, combined with an acidic-oxidative isolation protocol of the resulting dye stained biosilica, represents a novel strategy to develop highly porous luminescent biosilica nanostructures with promising applications in photonics.

Starting from polydisperse diatomaceous earth (DE), we proposed an efficient separation method for obtaining different morphologies of biosilica from diatoms. DE is a very low-cost source of silica containing all the differently nanostructured elements. By a glucose gradient/dialysis, three types of biosilica morphologies were achieved: rods, valves, and clusters. We fully characterized the diatom fractions and we used them to produce fluorescent biosilica platforms (“tabs”). These supports exhibited good resistance in water, ethanol, and soft scraping. A preliminary biologic application by testing Saos-2 proliferation was also performed to check osteoblasts-like cells biologic attitude for this scaffolds with tunable nanostructure.

Diatoms represent a natural source of mesoporous silica whose applications range from biomedical to photonic fields. Porous hierarchically organized micro structures, the biosilica shells called frustules, can be obtained by removal of the organic biological matter from the unicellular living algae. Diatoms frustules have been investigated as scaffold for bone tissue growth taking advantage of their nanostructured surface and of the possibility to chemically modify the biosilica. Here we report on an easy way to calcium-doped biosilica supports for bone tissue regeneration by in vivo feeding the algae. FTIR and EDX analyses confirmed the incorporation of calcium into the mesopouros biosilica. Cell viability studies showed an ameliorative effect on the Saos-2 cells spreading compared with the cells grown on non-doped biosilica supports.

Changes in the cardiac β-adrenergic system in early stages of Trypanosoma cruzi infection have been described. Here, we studied an early (135 days post-infection–p.i.) and a late stage (365 days p.i.) of the cardiac chronic form of the experimental infection (Tulahuen or SGO-Z12 strains), determining plasma epinephrine and norepinephrine levels, β-receptor density, affinity and function, cardiac cAMP concentration and phosphodiesterase activity, cardiac contractility, and the presence of β-receptor autoantibodies. Tulahuen-infected mice presented lower epinephrine and norepinephrine levels; lower β-receptor affinity and density; a diminished norepinephrine response and higher cAMP levels in the early stage, and a basal contractility similar to non-infected controls in the early and augmented in the late stage. The Tulahuen strain induced autoantibodies with weak β-receptor interaction. SGO-Z12-infected mice presented lower norepinephrine levels and epinephrine levels that diminished with the evolution of the infection; lower β-receptor affinity and an increased density; unchanged epinephrine and norepinephrine response in the early and a diminished response in the late stage; higher cAMP levels and unchanged basal contractility. The SGO-Z12 isolate induced β-receptor autoantibodies with strong interaction with the β-receptors. None of the antibodies, however, acted a as β-receptor agonist. The present results demonstrate that this system is seriously compromised in the cardiac chronic stage of T. cruzi infection.
