Focused assessment with sonography for trauma (FAST) has been incorporated into the initial evaluation of trauma for decades. It is an important screening tool in the detection of intra-abdominal fluid. The objective of this study was to perform a systematic review of the use and accuracy of FAST as an imaging tool for blunt abdominal trauma in disaster/mass casualty settings. A systematic review of literature was conducted using key words and search terms. Two independent reviewers screened abstracts to determine inclusion using the Quality Assessment of Diagnostic Accuracy Studies (QUADAS). For studies passing QUADAS, a meta-analysis was performed calculating sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV). FAST results were compared with the gold standard, which was a combination of CT scan results, operative findings, and medical records of the clinical course. Initial database screening resulted in 133 articles, of which 21 were selected for QUADAS evaluation. Five studies passed QUADAS and were selected in the final meta-analysis, with a total of 4263 patients. The sensitivity of FAST was 92.1% (87.8–95.6), specificity 98.7% (96.0–99.9), PPV 90.7% (70.0–98.0), and NPV 98.8% (98.1–99.5) for the detection of intra-abdominal injury. In our meta-analysis, FAST was both sensitive and specific in the evaluation of trauma in the disaster setting.

A controversy at the 2016 IUCN World Conservation Congress on the topic of closing domestic ivory markets (the 007, or so-called James Bond, motion) has given rise to a debate on IUCN's value proposition. A cross-section of authors who are engaged in IUCN but not employed by the organization, and with diverse perspectives and opinions, here argue for the importance of safeguarding and strengthening the unique technical and convening roles of IUCN, providing examples of what has and has not worked. Recommendations for protecting and enhancing IUCN's contribution to global conservation debates and policy formulation are given.

Managing agricultural pests with an incomplete understanding of the impacts that tactics have on crops, pests, and other organisms poses risks for loss of short-term profits and longer-term negative impacts, such as evolved resistance and nontarget effects. This is especially relevant for the management of weeds that are viewed almost exclusively as major impediments to crop production. Seldom considered in weed management are the benefits weeds provide in agroecosystems, which should be considered for optimal decision-making. Integration of weed costs and benefits will become increasingly important as management for pests transitions away from nearly complete reliance on herbicides and transgenic crop traits as the predominant approach for control. Here, we introduce a weed-management decision framework that accounts for weed benefits and exemplify how in-crop weed occurrence can increase crop yields in which a highly damaging insect also occurs. We highlight a case study showing how management decision-making for common milkweed, which is currently controlled primarily with glyphosate in herbicide-tolerant corn, can be improved by integrating management of the European corn borer (ECB), which is currently controlled primarily by the transgenic toxin Cry1 in Bacillus thuringiensis corn. Our data reveal that milkweed plants harboring aphids provide a food source (honeydew) for parasitoid wasps, which attack ECB eggs. Especially at high ECB population densities (> 1 egg mass leaf–1), maintaining low milkweed densities (< 1 stem m–2), effectively helps to minimize yield losses from ECB and to increase the economic injury level of this aggressive perennial weed. In addition, milkweed is the host for the monarch butterfly, so breeding-ground occurrences of the plant, including crop fields, may help sustain populations of this iconic insect. Using a more-holistic approach to integrate the management of multiple crop pests has the capacity to improve decision-making at the field scale, which can improve outcomes at the landscape scale.

The IntCal09 and Marine09 radiocarbon calibration curves have been revised utilizing newly available and updated data sets from 14C measurements on tree rings, plant macrofossils, speleothems, corals, and foraminifera. The calibration curves were derived from the data using the random walk model (RWM) used to generate IntCal09 and Marine09, which has been revised to account for additional uncertainties and error structures. The new curves were ratified at the 21st International Radiocarbon conference in July 2012 and are available as Supplemental Material at www.radiocarbon.org. We present an in vitro model of human skin that, together with nonlinear optical microscopy, provides a useful system for characterizing morphological and structural changes in a living skin tissue microenvironment due to changes in oxygen status and proteolytic balance. We describe for the first time the effects of chronic oxygen deprivation on a bioengineered model of human interfollicular epidermis. Histological analysis and multiphoton imaging revealed a progressively degenerating ballooning phenotype of the keratinocytes that manifested after 48 h of hypoxic exposure. Multiphoton images of the dermal compartment revealed a decrease in collagen structural order. Immunofluorescence analysis showed changes in matrix metalloproteinase (MMP)-2 protein spatial localization in the epidermis with a shift to the basal layer, and loss of Ki67 expression in proliferative basal cells after 192 h of hypoxic exposure. Upon reoxygenation MMP-2 mRNA levels showed a biphasic response, with restoration of MMP-2 levels and localization. These results indicate that chronic oxygen deprivation causes an overall degeneration in tissue architecture, combined with an imbalance in proteolytic expression and a decrease in proliferative capacity. We propose that these tissue changes are representative of the ischemic condition and that our experimental model system is appropriate for addressing mechanisms of susceptibility to chronic wounds.

Panspermia, the dissemination of life through space, would require resistance to the conditions found in space, including UV light. All known life forms depend on DNA to store information. In an effort to understand the liabilities of DNA to UV light and modes of DNA protection in terrestrial life forms, we established UV–VUV (125–340 nm) absorption spectra for dry DNA and its polymerized components and mononucleotides, as well as for a selection of potential UV screens ubiquitous in all organisms, including proteins, selected amino acids and amines (polyamines and tyramine). Montmorillonite clay was included as a potential abiotic UV screen. Among the potential screens tested, adenosine triphosphate (ATP) appeared to be particularly attractive, because its UV absorption spectrum was similar to that of DNA. We suggest that the use of ATP in UV protection could have pre-dated its current role in energy transfer. Spectroscopy also showed that UV absorption varied according to nucleotide content, suggesting that base pair usage could be a factor in adaptation to given UV environments and the availability of UV screens.

Dynamic Atomic Force Microscopy (AFM) is typically performed at amplitudes that are quite large compared to the measured interaction range. This complicates the data interpretation as measurements become highly non-linear. A new dynamic AFM technique in which ultra-small amplitudes are used (as low as 0.15 Angstrom) is able to linearize measurements of nanomechanical phenomena in ultra-high vacuum (UHV) and in liquids. Using this new technique we have measured single atom bonding, atomic-scale dissipation and molecular ordering in liquid layers, including water.

At a time of the emergence of drug-resistant bacterial strains, the development of antimicrobial compounds with novel mechanisms of action is of considerable interest. Perhaps the most promising among these is a family of antibacterial peptides originally isolated from insects. These were shown to act in a stereospecific manner on an as-yet unidentified target bacterial protein. One of these peptides, drosocin, is inactive in vivo due to the rapid decomposition in mammalian sera. However, another family member, pyrrhocoricin, is significantly more stable, has increased in vitro efficacy against Gram-negative bacterial strains, and if administered alone, as we show here, is devoid of in vitro or in vivo toxicity. At low doses, pyrrhocoricin protected mice against Escherichia coli infection, but at a higher dose augmented the infection of compromised animals. Analogs of pyrrhocoricin were, therefore, synthesized to further improve protease resistance and reduce toxicity. A linear derivative containing unnatural amino acids at both termini showed high potency and lack of toxicity in vivo and an expanded cyclic analog displayed broad activity spectrum in vitro. The bioactive conformation of native pyrrhocoricin was determined by nuclear magnetic resonance spectroscopy, and similar to drosocin, reverse turns were identified as pharmacologically important elements at the termini, bridged by an extended peptide domain. Knowledge of the primary and secondary structural requirements for in vivo activity of these peptides allows the design of novel antibacterial drug leads.

A new type of AFM is presented which allows for direct measurements of nanomechanical properties in ultra-high vacuum and liquid environments. The AFM is also capable of atomic-scale imaging of force gradients. This is achieved by vibrating a stiff lever at very small amplitudes of less than 1 Å (peak-to-peak) at a sub-resonance amplitude. This linearizes the measurement and makes the interpretation of the data straight-forward. At the atomic scale, interaction force gradients are measured which are consistent with the observation of single atomic bonds. Also, atomic scale damping is observed which rapidly rises with the tip-sample separation. A mechanism is proposed to explain this damping in terms of atomic relaxation in the tip. We also present recent results in water where we were able to measure the mechanical response due to the molecular ordering of water close to an atomically flat surface.
