Dates from the mammal fauna of Crystal Caverns, El Dorado County, California confirm that it is a Pleistocene deposit from the Last Glacial Maximum. Results indicate it is similar in age to other comparable California caves and has a high degree of time averaging.

Samples of bone and teeth from the Middle Paleolithic open-air site of Rozhok I, located on the Gulf of Taganrog, were dated by radiocarbon (14C). We obtained 26 accelerator mass spectrometry (AMS) dates on a large sample of nonhuman bone and teeth recovered by N. D. Praslov during 1961–1962 from 5 of the 6 occupation horizons at Rozhok I. Two rigorous pretreatment methods were applied to the samples: ultra-filtration and XAD resin. The 14C ages from the uppermost two layers (Horizons 1 and 2) are infinite with the exception of one age near the limit of the 14C method. By contrast, the dates from Horizon 3 and the two lowermost layers (Horizons 5 and 6) are all finite ages and some are less than 45,000 BP (uncalibrated). While there is evidence of some post-depositional disturbance, there is no evidence of a stratigraphic reversal of the cultural layers. The simplest explanation for the pattern is that bones and teeth in the lower layers have been contaminated by groundwater containing young carbon. We conclude that Rozhok I probably antedates 50,000 cal BP and is beyond the range of 14C dating.

The radiocarbon (14C) calibration curve so far contains annually resolved data only for a short period of time. With accelerator mass spectrometry (AMS) matching the precision of decay counting, it is now possible to efficiently produce large datasets of annual resolution for calibration purposes using small amounts of wood. The radiocarbon intercomparison on single-year tree-ring samples presented here is the first to investigate specifically possible offsets between AMS laboratories at high precision. The results show that AMS laboratories are capable of measuring samples of Holocene age with an accuracy and precision that is comparable or even goes beyond what is possible with decay counting, even though they require a thousand times less wood. It also shows that not all AMS laboratories always produce results that are consistent with their stated uncertainties. The long-term benefits of studies of this kind are more accurate radiocarbon measurements with, in the future, better quantified uncertainties.

Radiocarbon (14C) ages cannot provide absolutely dated chronologies for archaeological or paleoenvironmental studies directly but must be converted to calendar age equivalents using a calibration curve compensating for fluctuations in atmospheric 14C concentration. Although calibration curves are constructed from independently dated archives, they invariably require revision as new data become available and our understanding of the Earth system improves. In this volume the international 14C calibration curves for both the Northern and Southern Hemispheres, as well as for the ocean surface layer, have been updated to include a wealth of new data and extended to 55,000 cal BP. Based on tree rings, IntCal20 now extends as a fully atmospheric record to ca. 13,900 cal BP. For the older part of the timescale, IntCal20 comprises statistically integrated evidence from floating tree-ring chronologies, lacustrine and marine sediments, speleothems, and corals. We utilized improved evaluation of the timescales and location variable 14C offsets from the atmosphere (reservoir age, dead carbon fraction) for each dataset. New statistical methods have refined the structure of the calibration curves while maintaining a robust treatment of uncertainties in the 14C ages, the calendar ages and other corrections. The inclusion of modeled marine reservoir ages derived from a three-dimensional ocean circulation model has allowed us to apply more appropriate reservoir corrections to the marine 14C data rather than the previous use of constant regional offsets from the atmosphere. Here we provide an overview of the new and revised datasets and the associated methods used for the construction of the IntCal20 curve and explore potential regional offsets for tree-ring data. We discuss the main differences with respect to the previous calibration curve, IntCal13, and some of the implications for archaeology and geosciences ranging from the recent past to the time of the extinction of the Neanderthals.

We performed a new series of measurements on samples that were part of early measurements on radiocarbon (14C) dating made in 1948–1949. Our results show generally good agreement to the data published in 1949–1951, despite vast changes in technology, with only two exceptions where there was a discrepancy in the original studies. Our new measurements give calibrated ages that overlap with the known ages. We dated several samples at four different laboratories, and so we were also able to make a small intercomparison at the same time. In addition, new measurements on samples from other Egyptian materials used by Libby and co-workers were made at UC Irvine. Samples of tree rings used in the original studies (from Broken Flute Cave and Centennial Stump) were obtained from the University of Arizona Laboratory of Tree-Ring Research archive and remeasured. New data were compared to the original studies and other records.

While the North American archaeological record signals the presence of early humans along the northeastern Pacific coast by the Late Pleistocene, we know little about the technological systems employed by these coastally oriented colonizing groups. We here report the discovery of the earliest unequivocal evidence for the use and manufacture of shell fishhooks in the western hemisphere. Four single-piece shell fishhooks dating to the terminal Pleistocene/early Holocene transition (between ~11,300 and 10,700 cal B.P.) have been excavated on Isla Cedros, Baja California, Mexico. One hook is directly dated at 9495 ± 25 B.P. with a marine reservoir–corrected age of 11,165–9185 cal B.P. Radiocarbon ages associated with three other shell fishhooks range between 8900 ± 25 B.P. and 10,415 ± 25 B.P, while median ages for the earliest contexts confirm occupation of the island by at least 12,600–12,000 cal B.P. The stratigraphic levels from which the fishhooks were recovered contained a diverse assemblage of fish remains, including deepwater species, indicative of boat use. Thus, some of the earliest known inhabitants of the Pacific coast of the Americas employed shell hook and line technology for offshore marine fishing at least by the Pleistocene-Holocene transition, if not earlier.

Late-glacial macrofossils from a 10-m core from Alpine Swamp, New Jersey, were radiocarbon dated using accelerator mass spectrometry (AMS). The arrival of the first trees to the area following deglaciation is indicated by maximum percentages of spruce pollen and a date of 12,290 ± 440 yr B.P. on a single spruce needle. Subsequent spread of deciduous hardwoods was followed by the expansion of boreal taxa, including spruce (Picea), fir (Abies), larch (Larix laricina), paper birch (Betula papyrifera), and alder (Alnus). Three AMS dates on paper birch seeds and a spruce needle during this boreal expansion indicate that it took place between 11,000 and 10,000 yr B.P. The timing of this vegetational shift and its correlation with late-glacial pollen stratigraphy from many sites in southern New England indicate that a climatic reversal correlative with the Younger Dryas characterized the North Atlantic seaboard of the United States.

Combining atmospheric Δ14CO2 data sets from different networks or laboratories requires secure knowledge on their compatibility. In the present study, we compare Δ14CO2 results from the Heidelberg low-level counting (LLC) laboratory to 12 international accelerator mass spectrometry (AMS) laboratories using distributed aliquots of five pure CO2 samples. The averaged result of the LLC laboratory has a measurement bias of –0.3±0.5‰ with respect to the consensus value of the AMS laboratories for the investigated atmospheric Δ14C range of 9.6 to 40.4‰. Thus, the LLC measurements on average are not significantly different from the AMS laboratories, and the most likely measurement bias is smaller than the World Meteorological Organization (WMO) interlaboratory compatibility goal for Δ14CO2 of 0.5‰. The number of intercomparison samples was, however, too small to determine whether the measurement biases of the individual AMS laboratories fulfilled the WMO goal.

This paper presents the first successful methods for accelerator mass spectrometry (AMS) dating of asphalt-impregnated insect chitin from the Rancho La Brea Tar Pits in southern California. A persistent problem with stratigraphic correlation at this site is that asphalt flows are characteristically intermittent and are really discontinuous, which can result in mixing fossils of quite different ages. Direct 14C dating of specimens circumvents this difficulty but requires a pretreatment method that can produce dates from relatively small samples (<10 mg) of insect cuticle, while successfully removing residual asphalt and sample preparation solvents as well as soil carbon contamination. 14C dating accuracy was verified by comparing dates on insect chitin with ages for seeds and twigs compacted during a rapid entrapment event within a separately dated skull of the Western Camel, Camelops hesternus Leidy. All dates fell within a relatively narrow range of ~40,000–44,000 14C yr BP, suggesting that such methods can be used with confidence on other insect material from this site. Insects are often superior paleoenvironmental indicators for establishing precise data points for climate fluctuations. This is because their lifecycles and present-day climate-restricted geographic distributions are well documented, and unlike migrating mammals and birds, insects offer crucial information about the local environment. Our results are therefore potentially significant for studies of paleoecological and paleoclimatic change within the Los Angeles Basin and coastal southern California, as well as reconstruction of entrapment events at Rancho La Brea.

The IntCal09 and Marine09 radiocarbon calibration curves have been revised utilizing newly available and updated data sets from 14C measurements on tree rings, plant macrofossils, speleothems, corals, and foraminifera. The calibration curves were derived from the data using the random walk model (RWM) used to generate IntCal09 and Marine09, which has been revised to account for additional uncertainties and error structures. The new curves were ratified at the 21st International Radiocarbon conference in July 2012 and are available as Supplemental Material at www.radiocarbon.org. The database can be accessed at http://intcal.qub.ac.uk/intcal13/.

Four accelerator mass spectrometry (AMS) facilities undertook an interlaboratory exercise designed to examine the reliability and reproducibility of radiocarbon determinations on bone by dating a sample of elk (Alces alces) from Miesenheim IV. This specimen is derived from a secure geological context directly beneath the Laacher See tephra, which provides a precise terminus ante quern of ∼11,060 yr BP (∼13,050 cal yr BP). Regrettably, the results of the intercomparison exercise were complicated by evident contamination of the bone sample by exogenous organic material. This contaminant, probably humic acid, resulted in a wide span of ages (10,010 ± 30 to 11,100 ± 45 BP). The only method that yielded an accurate determination, consistent with the age of the tephra, was Oxford's single amino acid technique, which targets hydroxyproline. An acid hydrolysis step seems to have been crucial in breaking the bonds between the bone collagen and the contaminant.

High-quality data from appropriate archives are needed for the continuing improvement of radiocarbon calibration curves. We discuss here the basic assumptions behind 14C dating that necessitate calibration and the relative strengths and weaknesses of archives from which calibration data are obtained. We also highlight the procedures, problems, and uncertainties involved in determining atmospheric and surface ocean 14C/12C in these archives, including a discussion of the various methods used to derive an independent absolute timescale and uncertainty. The types of data required for the current IntCal database and calibration curve model are tabulated with examples.

We establish a precision accelerator mass spectrometry (AMS) radiocarbon chronology for the Archaic period Tlacuachero shellmound (Chiapas, Mexico) within a Bayesian statistical framework. Carbonized twig samples were sequentially selected from well-defined stratigraphic contexts based on iterative improvements to a probabilistic chronological model. Analytical error for these measurements is ±15 to 20 14C yr. This greater precision and the absence of stratigraphic reversals eclipses previous 14C work at the site. Based on this, we establish a chronological framework for a sequence of 3 clay floors dating to between 4930 and 4270 cal BP and determine that the bedded shell deposits that formed the mound accumulated rapidly during 2 episodes: a lower 2-m section below the floors that accumulated over a 0–150 cal yr period at 5050–4875 cal BP and, an upper 3.5-m section above the floors that accumulated over a 0–80 cal yr period at 4380–4230 cal BP.

We have compared accelerator mass spectrometry (AMS) radiocarbon results on wood samples at or near the limit of 14C dating, pretreated with a standard acid-base-acid (ABA) protocol, with those obtained from cellulose prepared from the same samples by several modifications of the Jayme-Wise cellulose extraction method (Green 1963). These tests were carried out to determine the most efficient way to ensure low backgrounds in 14C measurements of well-preserved ancient wood samples.

The Keck Carbon Cycle AMS facility at the University of California, Irvine (KCCAMS/UCI) has developed protocols for analyzing radiocarbon in samples as small as ∼0.001 mg of carbon (C). Mass-balance background corrections for modern and 14C-dead carbon contamination (MC and DC, respectively) can be assessed by measuring 14C-free and modern standards, respectively, using the same sample processing techniques that are applied to unknown samples. This approach can be validated by measuring secondary standards of similar size and 14C composition to the unknown samples. Ordinary sample processing (such as ABA or leaching pretreatment, combustion/graphitization, and handling) introduces MC contamination of ∼0.6 ± 0.3 μg C, while DC is ∼0.3 ± 0.15 μg C. Today, the laboratory routinely analyzes graphite samples as small as 0.015 mg C for external submissions and ≅0.001 mg C for internal research activities with a precision of ∼1% for ∼0.010 mg C. However, when analyzing ultra-small samples isolated by a series of complex chemical and chromatographic methods (such as individual compounds), integrated procedural blanks may be far larger and more variable than those associated with combustion/graphitization alone. In some instances, the mass ratio of these blanks to the compounds of interest may be so high that the reported 14C results are meaningless. Thus, the abundance and variability of both MC and DC contamination encountered during ultra-small sample analysis must be carefully and thoroughly evaluated. Four case studies are presented to illustrate how extraction chemistry blanks are determined.

The IntCal04 and Marine04 radiocarbon calibration curves have been updated from 12 cal kBP (cal kBP is here defined as thousands of calibrated years before AD 1950), and extended to 50 cal kBP, utilizing newly available data sets that meet the IntCal Working Group criteria for pristine corals and other carbonates and for quantification of uncertainty in both the 14C and calendar timescales as established in 2002. No change was made to the curves from 0–12 cal kBP. The curves were constructed using a Markov chain Monte Carlo (MCMC) implementation of the random walk model used for IntCal04 and Marine04. The new curves were ratified at the 20th International Radiocarbon Conference in June 2009 and are available in the Supplemental Material at www.radiocarbon.org.

A preliminary determination of the absolute 14C/12C ratio of the oxalic acid I standard (NBS SRM 4990B) has been made. Using an accelerator mass spectrometry (AMS) system, a known number of radiocarbon ions were implanted in a thin copper foil. The foil was then combusted with the addition of 14C-free carrier material. This resulting gas (which had a known 14C/12C ratio) was graphitized and compared to OX-I material. This comparison yielded an absolute 14C/12C ratio of OX-I. The absolute 14C/12C ratio of OX-I, coupled with knowledge of the specific activity of OX-1, provides an alternative determination of the 14C half-life.

We present an overview of accelerator mass spectrometry (AMS) radiocarbon sample preparation and measurements, describing the technical upgrades that now allow us to routinely obtain 0.2–0.3% precision for 1-mg carbon samples. A precision of ∼1% on samples with 100 μg of carbon can also be achieved. We have also developed graphitization techniques and AMS procedures for ultra-small samples (down to 0.002 mg of carbon). Detailed time series are presented for large and small aliquots of standards such as NIST OX-I and OX-II; FIRI-C and -D; IAEA-C6, -C7 and -C8; and 14C-free samples.

We demonstrate variable radiocarbon content within 2 historic (AD 1936) and 2 prehistoric (about 8200 BP and 3500 BP) Mytilus californianus shells from the Santa Barbara Channel region, California, USA. Historic specimens from the mainland coast exhibit a greater range of intrashell variability (i.e. 180–240 14C yr) than archaeological specimens from Daisy Cave on San Miguel Island (i.e. 120 14C yr in both shells). δ13C and δ18O profiles are in general agreement with the up welling of deep ocean water depleted in 14C as a determinant of local marine reservoir correction (ΔR) in the San Miguel Island samples. Upwelling cycles are difficult to identify in the mainland specimens, where intrashell variations in 14C content may be a complex product of oceanic mixing and periodic seasonal inputs of 14C-depeleted terrestrial runoff. Though the mechanisms controlling ΔR at subannual to annual scales are not entirely clear, the fluctuations represent significant sources of random dating error in marine environments, particularly if a small section of shell is selected for accelerator mass spectrometry (AMS) dating. For maximum precision and accuracy in AMS dating of marine shells, we recommend that archaeologists, paleontologists, and 14C lab personnel average out these variations by sampling across multiple increments of growth.

Nutrigenomics is the study of how constituents of the diet interact with genes, and their products, to alter phenotype and, conversely, how genes and their products metabolise these constituents into nutrients, antinutrients, and bioactive compounds. Results from molecular and genetic epidemiological studies indicate that dietary unbalance can alter gene–nutrient interactions in ways that increase the risk of developing chronic disease. The interplay of human genetic variation and environmental factors will make identifying causative genes and nutrients a formidable, but not intractable, challenge. We provide specific recommendations for how to best meet this challenge and discuss the need for new methodologies and the use of comprehensive analyses of nutrient–genotype interactions involving large and diverse populations. The objective of the present paper is to stimulate discourse and collaboration among nutrigenomic researchers and stakeholders, a process that will lead to an increase in global health and wellness by reducing health disparities in developed and developing countries.
