We investigated the drug resistance of Mycobacterium tuberculosis isolates from patients with tuberculosis (TB) and HIV, and those diagnosed with only TB in Sichuan, China. TB isolates were obtained from January 2018 to December 2020 and subjected to drug susceptibility testing (DST) to 11 anti-TB drugs and to GeneXpert MTB/RIF testing. The overall proportion of drug-resistant TB (DR-TB) isolates was 32.1% (n = 10 946). HIV testing was not universally available for outpatient TB cases, only 29.5% (3227/10 946) cases had HIV testing results. The observed proportion of multidrug-resistant TB (MDR-TB) isolates was almost double than that of the national level, with approximately 1.5% and 0.1% of the isolates being extensively drug resistant and universally drug resistant, respectively. The proportions of resistant isolates were generally higher in 2018 and 2019 than in 2020. Furthermore, the sensitivities of GeneXpert during 2018–2020 demonstrated a downward trend (80.9, 95% confidence intervals (CI) 76.8–85.0; 80.2, 95% CI 76.4–84.1 and 75.4, 95% CI 70.7–80.2, respectively). Approximately 69.0% (7557/10 946) of the TB cases with DST results were subjected to GeneXpert detection. Overall, the DR-TB status and the use of GeneXpert in Sichuan have improved, but DR-TB challenges remain. HIV testing for all TB cases is recommended.

The aim of the present study was to examine whether serum Zn concentrations were associated with metabolic risk factors in Chinese children and adolescents. This was a cross-sectional study including 3241 participants, aged 6 to 17 years, from Jiangsu, China. Metabolic risk factors included fasting glucose (FG), total cholesterol (TC), TAG, HDL-cholesterol, LDL-cholesterol, systolic blood pressure and diastolic blood pressure. Data were analysed using multi-variable linear regression and generalised additive models, which were adjusted for age, sex, high-sensitive C-reactive protein, estimated glomerular filtration rate, BMI and region of residence, to assess the associations of serum Zn concentrations with metabolic risk factors. We observed a negative association between serum Zn concentrations and FG (coefficient = −0·532; 95 % CI −0·569, −0·495; P < 0·001). Moreover, TC (coefficient = 0·175; 95 % CI 0·127, 0·222; P < 0·001), HDL-cholesterol (coefficient = 0·137; 95 % CI 0·082, 0·193; P < 0·001) and LDL-cholesterol (coefficient = 0·195; 95 % CI 0·128, 0·263; P < 0·001) were found to be positively associated with Zn levels. A generalised additive model showed that the negative association between serum Zn and FG was weak at lower serum Zn concentrations and was stronger with the increase in serum Zn concentrations. Additionally, a U-shaped association between serum Zn and TAG was observed. Serum Zn concentrations were associated with FG, TC, TAG, HDL-cholesterol and LDL-cholesterol levels in Chinese children and adolescents. Lower levels of serum Zn were more likely related to a poor metabolic status.

The rapid progress in the performance of today's sophisticated transmission electron microscopes also demands computational and educational tools capable of simulating the intricacy of electron image formation. The tools ideally should be widely accessible to the microscopy community and capable of covering both the breadth and depth demanded by modern materials science. Here, a cloud-based microscopy simulation platform, called cloudEMAPS and powered by cloud computing and modern server-client web programming architecture, is described. Compared to the current desktop solutions for electron microscopy simulations, cloud computing offers the unique advantages of on-demand, data sharing, high-performance computation and internet easy access using the state-of-the-art web infrastructure. This article will demonstrate these advantages using examples of interactive simulations of electron diffraction patterns and aberration-corrected electron optics.

We report on the generation of a mid-infrared (mid-IR) frequency comb with a maximum average output power of 250 mW and tunability in the 2.7–4.0 μm region. The approach is based on a single-stage difference frequency generation (DFG) starting from a compact Yb-doped fiber laser system. The repetition rate of the near-infrared (NIR) comb is locked at 75 MHz. The phase noise of the repetition rate in the offset-free mid-IR comb system is measured and analyzed. Except for the intrinsic of NIR comb, environmental noise at low frequency and quantum noise at high frequency from the amplifier chain and nonlinear spectral broadening are the main noise sources of broadening the linewidth of comb teeth, which limits the precision of mid-IR dual-comb spectroscopy.

Underground Nuclear Astrophysics in China (JUNA) will take the advantage of the ultra-low background in Jinping underground lab. High current accelerator with an ECR source and detectors were commissioned. JUNA plans to study directly a number of nuclear reactions important to hydrostatic stellar evolution at their relevant stellar energies. At the first period, JUNA aims at the direct measurements of 25Mg(p,γ)26 Al, 19F(p,α) 16 O, 13C(α, n) 16O and 12C(α,γ) 16O near the Gamow window. The current progress of JUNA will be given.

The effects of high magnetic fields on the solidification structures of ternary Al–Fe–Zr alloy were investigated. The results showed that the primary Al3Fe crystals mainly show bar-like form, whereas the unmelted Al3Zr crystals reveal tabular and the newly crystallized primary Al3Zr crystals have fine/coarse needle-like forms. When a 12 T magnetic field is applied, the primary Al3Fe crystals are distributed homogenously and the fine needle-like primary Al3Zr levitated. Moreover, the primary Al3Fe crystals align horizontally in the upper but vertically in the lower part of the specimen. The needle-like primary Al3Zr crystals align vertically, whereas the tabular ones have their two opposite corners on the large surfaces toward the positive and negative magnetic field direction. Crystallographic analysis indicates that 〈100〉 and 〈110〉 are the preferred axes of the primary Al3Fe and the Al3Zr crystals with respect to the magnetic field, respectively. The redistribution and realignments of the crystals are discussed.

In this paper we consider new perturbation bounds analysis of a kind of generalized saddle point systems. We provide perturbation upper bounds for the solutions of generalized saddle point systems, which extend the corresponding results in [W.-W. Xu, W. Li, New perturbation analysis for generalized saddle point systems, Calcolo., 46(2009), pp. 25-36] to more general cases.

ZrO2:Eu3+ hollow spheres were successfully fabricated with the resin microspheres as the template. The sample characterizations were carried out by means of x-ray diffraction (XRD), scanning electron microscope (SEM), and photoluminescence spectra. XRD results revealed that Eu3+-doped samples were pure t-ZrO2 phase after being calcined at 873 K. SEM results exhibited that this Eu3+ doped ZrO2 was hollow spheres; the diameter and thickness of which were about 450 and 50 nm, respectively. Upon excitation at 394 nm, the orange-red emission bands at the wave length longer than 570 nm were from 5D0 → 7FJ (J = 1, 2) transitions. The asymmetry ratio of (5D0 → 7F2)/(5D0 → 7F1) intensity is about 1.61, 1.26, 1.42, 1.42, 1.40, and 1.38 for the Eu3+ concentration 0.4, 0.7, 1.0, 1.5, 2.0, and 2.5 mol%, respectively. These values suggest that the asymmetry ratio of Eu3+ ions is independent of the doping concentration. The optimal doping concentration of Eu3+ ions in ZrO2 is 1.5 mol%. According to Dexter's theory, the critical distance between Eu3+ ions for energy transfer was determined to be 16 Å.

A defect in the intestinal barrier is one of the characteristics of Crohn's disease (CD). The tight junction (TJ) changes and death of epithelial cells caused by intestinal inflammation play an important role in the development of CD. DHA, a long-chain PUFA, has been shown to be helpful in treating inflammatory bowel disease in experimental models by inhibiting the NF-κB pathway. The present study aimed at investigating the specific effect of DHA on the intestinal barrier function in IL-10-deficient mice. IL-10-deficient mice (IL-10− / −) at 16 weeks of age with established colitis were treated with DHA (i.g. 35·5 mg/kg per d) for 2 weeks. The severity of their colitis, levels of pro-inflammatory cytokines, epithelial gene expression, the distributions of TJ proteins (occludin and zona occludens (ZO)-1), and epithelial apoptosis in the proximal colon were measured at the end of the experiment. DHA treatment attenuated the established colitis and was associated with reduced infiltration of inflammatory cells in the colonic mucosa, lower mean histological scores and decreased levels of pro-inflammatory cytokines (IL-17, TNF-α and interferon-γ). Moreover, enhanced barrier function was observed in the DHA-treated mice that resulted from attenuated colonic permeability, rescued expression and corrected distributions of occludin and ZO-1. The results of the present study indicate that DHA therapy may ameliorate experimental colitis in IL-10− / − mice by improving the intestinal epithelial barrier function.

Effect of combined electromagnetic fields (EMFs) on the structures of a 3004 aluminum alloy ingot produced by horizontal direct chill casting was crystallographically investigated. The results showed that the structure was transformed from a mixture of equiaxed and fine columnar grains to coarse columnar grains with switching off the EMFs. With the EMFs the grain size is small and shows a uniform distribution, whereas without the EMFs it is increased and reveals inhomogeneous distribution on the cross section. Besides, a transition region composed of fine equiaxed grains appeared at the moment the EMFs were switched off (between the mixture and coarse columnar grains). Furthermore, the microstructure transformation is accompanied by a crystallographic orientation change from a preferred <100> orientation to a random orientation, and then to an intense <100> fiber texture. The structural and crystallographic transformations are mainly related to the forced convection in the melt due to the induced Lorentz force by the EMFs.

Outbreak of Mycobacterium tuberculosis infections associated with acupuncture has not been reported. Thirteen patients with a painful swollen lump were referred to our hospital. The index patient received acupuncture and paraspinal muscular injection at a local acupuncture clinic in April 2011 and was diagnosed with M. tuberculosis 1 month later. From May 2011 to August 2011, 12 more patients with a swollen lump on the nuchal region or in the lower back or the buttocks region were referred to our hospital. Tuberculin skin test (TST), T-SPOT.TB, acid-fast stain, M. tuberculosis culture, chest radiograph, and lump magnetic resonance imaging (MRI) were performed and the patients were diagnosed with tuberculous abscess of the lump. All 13 patients received intramuscular injection at the paraspinal muscle by two acupuncturists at a local clinic and reported a swollen lump at the injection site. The needles and syringes were reused after autoclave sterilization. The TST was positive in all patients. Twelve patients had positive acid-fast stains. Mycobacterial cultures of abscess specimens were positive in all 13 patients. T-SPOT.TB tests were positive in all patients who underwent the test. The lesions and biopsies were subjected to polymerase chain reaction (PCR) and gene sequencing by the Disease Control Center of Zhejiang Province, China and the causative agent was identified as M. tuberculosis, Beijing type. In conclusion, physicians should consider the possibility of mycobacterial infections, apart from other bacterial agents, in patients with a swollen paraspinal lump following intramuscular injection.

Approximately 50 % of patients with inflammatory bowel disease (IBD) suffer from anaemia, with Fe deficiency being the most common cause. CD52 monoclonal antibody (mAb) targets the cell surface CD52 and is effective in depleting lymphocytes through cytolytic effects in vivo. The aim of the present study was to investigate the therapeutic effect of anti-mouse CD52 mAb on Fe-deficient anaemia in IBD. IL-10 knockout mice (IL-10− / −) of 12 weeks with established colitis were treated with anti-mouse CD52 mAb once per week for 2 weeks. Severity of colitis, blood T lymphocytes, blood Hb, haematocrit, plasma erythropoietin (EPO), serum Fe concentration, transferrin saturation, splenic Fe stores, expression of liver hepcidin mRNA, Western blotting of the phosphorylated form of Smad1/5/8 and total Smad1 were measured at the end of the experiment. IL-10− / − mice treated with CD52 mAb showed a reduction in the percentage of CD4+ and CD4+CD45+ T cells in blood and weight loss typically associated with colonic inflammation, serum levels of EPO, the expression of liver hepcidin mRNA and total Smad1 protein, while they showed an increase in Hb concentrations, haematocrit, levels of serum Fe, transferrin saturation and splenic Fe stores. The present results indicated that anti-CD52 therapy may ameliorate Fe-deficient anaemia by reducing colonic inflammation. These findings may open novel horizons in the treatment of patients with IBD by resetting of immunological homeostasis in the gut by depleting the activated T cells in the gut mucosa.

Morphological and crystallographic effects of a high magnetic field on the primary Al6Mn phase formed during the solidification of hypereutectic Al–3.25wt%Mn were investigated. Without the field, the primary Al6Mn crystals are mainly concentrated in the lower part and reveal a dispersed needle-like shape. In three dimension, the needles are in the form of a quadrangular prism (laterally bound by {110} and preferentially growing along <001>). When the magnetic field is applied, they tend to be distributed homogenously and show some extra agglomerate- or chain-like forms (preferentially extending along <100>). Furthermore, they also tend to preferentially orient with <100> parallel to the field direction. The homogenous distribution is caused by the magnetic viscosity resistance force. The “agglomerates” or “chains” are the result of a “bifurcation effect” due to the breakdown at the sharp edges of the quadrangular prisms. The preferential orientation should be attributed to the magnetocrystalline anisotropy of Al6Mn.

The aim of the present study was to determine the effect of peroral bifidobacteria on the intestinal microbiota, barrier function and bacterial translocation (BT) in a mouse model of ischaemia and reperfusion (I/R) injury. A total of twenty-four male BALB/c mice were randomly allocated into three groups: (1) sham-operated, (2) I/R and (3) I/R injury and bifidobacteria pretreatment (109 colony-forming units/d). Bifidobacteria were administered daily intragastrically for 2 weeks before induction of I/R. Subsequently, samples of caecal content, intestinal mucosa, ileal segments, blood, mesenteric lymph nodes (MLN) and distant organs (liver, spleen and kidney) were prepared for examination. In the I/R model, barrier dysfunction (caecal microbiota dysbiosis, disruption of tight junction (TJ), increased epithelial cell apoptosis, disruption of mucosa and multiple erosions) in the intestine was observed, associated with increased BT to extraintestinal sites. The ratio of BT to MLN and distant organs in mice exposed to I/R injury was 62·5 %, which was significantly higher than the sham-operated group. However, pretreatment of animals with bifidobacteria prevented I/R-induced BT, reduced pro-inflammatory cytokine release, the levels of endotoxin, intestinal epithelial cell apoptosis, disruption of TJ and increased the concentration of SCFA, resulting in recovered microbiota and mucosal integrity. Bifidobacteria may be beneficial in reducing BT in I/R injury of mice. Therefore, peroral administration of bifidobacteria is a potential strategy to prevent I/R-induced BT and intestinal barrier dysfunction.

The present study investigated the effects of xanthophyll supplementation on production performance, antioxidant capacity (measured by glutathione peroxidase, superoxide dismutase (SOD), catalase, total antioxidant capacity (T-AOC), and reduced glutathione:oxidised glutathione ratio (GSH:GSSG)) and lipid peroxidation (measured by malondialdehyde (MDA)) in breeding hens and chicks. In Expt 1, 432 hens were fed diets supplemented with 0 (control group), 20 or 40 mg xanthophyll/kg diet. Blood samples were taken at 7, 14, 21, 28 and 35 d of the trial. Liver and jejunal mucosa were sampled at 35 d. Both xanthophyll groups improved serum SOD at 21 and 28 d, serum T-AOC at 21 d and liver T-AOC, and serum GSH:GSSG at 21, 28 and 35 d and liver GSH:GSSG. Xanthophylls also decreased serum MDA at 21 d in hens. Expt 2 was a 2 × 2 factorial design. Male chicks hatched from 0 or 40 mg in ovo xanthophyll/kg diet of hens were fed a diet containing either 0 or 40 mg xanthophyll/kg diet. Liver samples were collected at 0, 7, 14 and 21 d after hatching. Blood samples were also collected at 21 d. In ovo-deposited xanthophylls increased antioxidant capacity and decreased MDA in the liver mainly within 1 week after hatching. Maternal effects gradually vanished during 1–2 weeks after hatching. Dietary xanthophylls increased antioxidant capacity and decreased MDA in the liver and serum mainly from 2 weeks onwards. Data suggested that xanthophyll supplementation enhanced antioxidant capacity and reduced lipid peroxidation in different tissues of hens and chicks.
