The relationship between intensity and site of infection of Diorchis brevis, D. inflata and D. ransomi in Fulica atra from central Poland has been analysed; of 173 adult birds 152 (87·9%) were infected with one or more of these three species. From (1) the incidence of the three species (D. brevis, 59·5%; D. inflata, 69·9%; D. ransomi, 61·3%), (2) the high total incidence and high total intensity of infection (118·2 worms/bird) in hosts harbouring the three species concurrently and (3) the frequent co-occurrence of all three species in the same site in the host gut, it is concluded that there are no antagonistic interactions among these species. There are, however, some inter-dependencies between the number of parasites and the site occupied by particular species. In heavy infections there was a tendency to specific segregation and extension of the site of each species. This suggests the existence of inter- and intra-specific competition at higher densities of infection, regulating the spatial distribution of these parasites in the host gut.

The circadian migration of Hymenolepis diminuta in the small intestine of the rat may be correlated with a circadian variation in 5-hydroxytryptamine levels present in worm tissue, in the intestinal lumen, in the intestinal mucosa, with the amount of food present in the small intestine and in arterial blood. The 5-HT and food levels in uninfected animals were also determined. The 16.00 h stage in the circadian cycle marks both the commencement of host feeding, followed by rising 5-HT levels in both worm and host tissues, and initiation of an anteriad migration of worm biomass. It was found that 5-HT levels in the intestine of parasitized animals were significantly higher than in the intestine of uninfected controls. This is the first report of circadian variation in mucosal and luminal 5-HT levels. The similarity in the circadian patterns of worm migration and worm luminal, mucosal and blood 5-HT levels were striking. Fasting eliminated the circadian rise in intestinal 5-HT levels and the worms did not migrate. Luminal 5-HT levels were significantly lower in fasted animals than in the comparable rats fed ad libitum. When the intestine was ligatured at the pyloric sphincter, worm anteriad migration still occurred after feeding, indicating that the presence of exogenous food in the intestine is not a factor in the initial migration of the worms.

A prediction of the hypothesis of Wilson (1977, 1980a, b) to account for larval migration of homogonic Strongyloides ratti in the host is that the pattern of invasion of the mammary gland of a lactating rat will be quantitatively similar on both sides and independent of the point of entry into the body. Twenty-one suckled mother rats in 6 experiments in which live 75Se-labelled 3rd-stage homogonic larvae were injected under the skin of the upper flank had an overall distribution of label 30 h post-injection, as a percentage of the initial dose, in the quadrants, I (rear, injection side), II (rear, opposite injection side), III (front, injection side) and IV (front, opposite injection side) of the mammary gland as follows: 27·4%, 1·27%, 1·89% and 1·24%. Quantitative changes in mammary label between 30 and 48 h post-injection using live larvae, differences between mothers and virgins, and results after injection of heat-killed labelled larvae, confirm that the pattern is representative of the behaviour of normal (unlabelled) worms when injected. The theory is therefore disproved. The findings are put forward as the first quantitative evidence for major lymphatic involvement in migration of a skin-penetrating round worm. They need confirmation in similar experiments in which worms are allowed to penetrate the skin naturally. The role of isotope-labelled larvae versus traditional methods of estimating parasite content of host tissue is discussed.

The survival of the free-living stages of Trichostrongylus colubriformis under defined conditions of temperature and relative humidity was investigated. The survival of embryonated eggs was poor at 0, 33 and 54·5 % relative humidity (rel. hum.) at 20 °C but hatching occurred from a proportion of eggs even after exposure for 104 days to 76 and 98% rel. hum. at 20 °C. Second-stage larvae were desiccation-susceptible and were killed within 6 h even at 98% rel. hum. and 20 °C. Infective larvae, dried separately or in clumps, survived prolonged exposure to desiccation at 33–98% rel. hum. and 20 °C with 50% survival times of 58–164 days. Clump formation did not enhance survival in this range. Infective larvae also survived exposure to vacuum desiccation with 50% survival times of 8·8 h in clumps and 4·5 h when dried separately. The infective larva thus readily survives desiccation and may prove a useful model for the study of anhydrobiosis.

C57B1/6 male mice with chronic trichinellosis, and age-matched uninfected control mice, were inoculated with B16 melanoma cells. Tumour development was inhibited in the infected animals. In the infected tumour-bearing mice, the tumour induction intervals were longer and the tumour size was subsequently smaller than in the control mice. Moreover, when the number of tumour cells in the inoculum was less than that required to produce 100% tumour incidence in the uninfected mice (<1 × 104 cells), significantly more of the mice with an infection of 2 months duration remained tumour-free. These results were significant at the P<0·05 level.

Observations made with the transmission electron microscope are described from the free ovaries of sexually mature female Corynosoma semerme (Palaeacanthocephala) from the ringed seal (Pusa hispida). The structure of the mature ovaries consists of a supporting syncytium, oogonial cells and tissue and appears to be basically similar to that already described for 4 other species of Acanthocephala representing the 3 orders of the phylum. A comparison has been made between the organization of the cytoplasm of the mature oocyte and the zygote with reference to the consequences of fertilization. A detailed description is presented of the heterogeneous inclusions that are observed in the cytoplasm of the mature oocyte but not in the zygote. The fate and possible function of the components of these inclusions is discussed.

Various biological mixtures commonly used as microbiological nutrients were added to a chemically defined medium, containing 0·2% (w/v) albumin, in order to assess their ability to support maturation of eggs deposited by adult Schistosoma mansoni in vitro. Of the supplements tested, only lactalbumin hydrolysate and casein hydrolysate stimulated egg deposition and subsequent egg development in vitro. Upon addition of foetal calf serum, further stimulation was noted only in casein hydrolysate-containing media. The addition of mouse red blood cells did not significantly improve the system. Some miracidia which hatched from eggs derived from casein hydrolysate-fortified cultures displayed normal behavioural characteristics, penetrated susceptible snails and established patent infections. These results provide an experimental system for exploring the nutritional requirements of developing miracidia, and the physiological determinants that influence miracidial penetrative capacity and subsequent ability to develop in a susceptible intermediate host.

The weight gain and growth rate of Trichoplusia ni larvae were severely depressed following parasitization by the insect parasite, Hyposoter exiguae. The development of the host was also delayed and juvenile hormone esterase activity present at the normal time of moulting was absent in parasitized insects. Similar effects on growth and development were achieved by rearing host insects on nutrient-deficient media. Accompanying the above effects of parasitization were alterations in dry/wet weight ratio and host haemolymph osmolality. Parasitization resulted in immediate increases in the concentrations of haemolymph non-glycogen carbohydrate (composed of trehalose, maltose and glucose) and protein. The concentration of fat body glycogen also increased following parasitization. In contrast, the concentration of haemolymph non-glycogen carbohydrate and protein, as well as fat body glycogen, decreased in nutrient-deprived larvae. The significance of the above results as they relate to the insect parasite–host relationship is discussed.

The design and operation of a modular, bacteriological continuous-flow culture system have been adapted for the growth of Trypanosoma cruzi epimastigotes in simple monophasic media. The system was designed to achieve a minimum of 200 days of continuous culture and provision was made for the continuous supply of medium and collection of parasites under sterile conditions. The system provides large quantities of epimastigotes with homogeneous morphology and uniform viability. The system also lends itself tothe analysis of the factors which affect parasite growth. We have examined the effects of changes in environmental parameters on epimastigote growth rate. Optimal growth was observed at 27 °C. The rate ofstirring of the culture had a small but definable effect on the growth rate, which was greatest at 80 r.p.m. Growth was only slightly affected by the level of dissolved oxygen between 10 and 50% saturation, but was inhibited at higher concentrations. Growth was slower at extreme values of pH but showed a broad optimum around pH 7·4.

Haemoproteus kopki (De Mello, 1916) is reported from the geckoes Teratoscincus scincus and T. microlepis collected in three localities on the Afghan border of Pakistan. A detailed description of H. kopki is provided in correlation with the courses of an initial, acute infection and a relapsed, chronic infection. Haemoproteus kopki is characterized by large round or oval gametocytes which show sexual dimorphism in distribution of pigment: macrogametocytes show pigment dispersed in dark granules throughout the cytoplasm, while pigment in microgametocytes is aggregated into large, round golden masses within vacuoles in one, or rarely two foci. Other sexual differences exist in gametocyte dimensions. Gametocytes produced in the relapse infection grew much more slowly than did those of the initial infection, and never attained a comparable size despite being observed for twice the length of time. Despite a maximum parasitaemia of 53·8% in the initial infection, increased erythropoiesis did not occur. No asexual stages were found in tissues examined. The presence of mixed infections of H. kopki, Leishmania sp. and microfilariae in a harsh sand dune habitat with presumably limited dipteran fauna suggests their having a common vector in intimate association with the burrows inhabited by the geckoes.

The study of pathogenicity of haemoprotozoa and the pathogenesis of the diseases they cause requires quantitative descriptions. Statistical and mathematical methods are introduced to describe infectivity, parasitaemia, total body parasitosis and the severity of diseases.

The most efficient procedure for cryopreserving viable Babesia bovis organisms for in vitro cultivation consists of freezing extracellular parasites in a solution of 10% (w/v) polyvinylpyrrolidone (PVP) using a cooling rate of 20 °C/min. Although cultures can be established from thawed infected erythrocytes, the plating efficiency is relatively low. Freezing extracellular parasites resulted in plating efficiency up to 25%, when thawed and placed in culture. Glycerin or dimethyl sulphoxide (Me2SO) can be used successfully in the cryopreservation of B. bovis, but apparent toxic effects greatly decrease their efficiency. B. bovis parasites have been kept at − 196 °C for 60 days with no appreciable reduction in plating efficiency.

The regulation of helminth populations tends to occur primarily as a result of limitations imposed on the build up of parasite subpopulations within individual hosts (Anderson & May, 1979; May & Anderson, 1979). Considering the relevance of these factors to the success or otherwise of intestinal helminth control programmes, it is perhaps surprising that more information is not yet available concerning the particular mechanisms which may be responsible, and in particular, the population consequences of the immune responses which such parasites may precipitate. Density-dependence in a single rate parameter, if operative over the naturally observed numerical range, is sufficient to regulate parasite population flow throughout the life-cycle, whether direct or indirect (Anderson, 1976). For the genera given in Table 1, this could be provided by the observed pattern of parasite mortality and/or fecundity. It is of interest to note, however, that circumstantial evidence cited in the Table suggests that each of the 6 genera is also potentially able to induce host mortality under certain conditions. Whether this acts in a density-dependent manner in natural infections is almost entirely unknown. Rapid reproduction may be of great selective advantage to intestinal helminths, even if it is necessarily accompanied by pathogenicity (see Anderson, 1981). If the manner in which this pathogenicity acts in any way enhances the stability of the host-parasite interaction, then perhaps it may have contributed to the selection pressures which have led so many genera to continue to break the rules of the ‘well-adapted’ parasite (see, for example, Noble & Noble, 1971).
