The COllaborative project of Development of Anthropometrical measures in Twins (CODATwins) project is a large international collaborative effort to analyze individual-level phenotype data from twins in multiple cohorts from different environments. The main objective is to study factors that modify genetic and environmental variation of height, body mass index (BMI, kg/m2) and size at birth, and additionally to address other research questions such as long-term consequences of birth size. The project started in 2013 and is open to all twin projects in the world having height and weight measures on twins with information on zygosity. Thus far, 54 twin projects from 24 countries have provided individual-level data. The CODATwins database includes 489,981 twin individuals (228,635 complete twin pairs). Since many twin cohorts have collected longitudinal data, there is a total of 1,049,785 height and weight observations. For many cohorts, we also have information on birth weight and length, own smoking behavior and own or parental education. We found that the heritability estimates of height and BMI systematically changed from infancy to old age. Remarkably, only minor differences in the heritability estimates were found across cultural–geographic regions, measurement time and birth cohort for height and BMI. In addition to genetic epidemiological studies, we looked at associations of height and BMI with education, birth weight and smoking status. Within-family analyses examined differences within same-sex and opposite-sex dizygotic twins in birth size and later development. The CODATwins project demonstrates the feasibility and value of international collaboration to address gene-by-exposure interactions that require large sample sizes and address the effects of different exposures across time, geographical regions and socioeconomic status.
This study aims to investigate the prevalence and genotype distribution of anal human papillomavirus (HPV) infection among men with different sexual orientations with or without human immunodeficiency virus (HIV) in China. A cross-sectional study was conducted during 2016–2017 in Taizhou City, Zhejiang Province. Convenient sampling was used to recruit male participants from HIV voluntary counselling and testing clinics and Center for Disease Control and Prevention. A face-to-face questionnaire interview was administered and an anal-canal swab was collected for HPV genotyping. A total of 160 HIV-positive and 113 HIV-negative men participated in the study. The prevalence of any type HPV was 30.6% for heterosexual men, 74.1% for homosexual and 63.6% for bisexual men among HIV-positive participants, while the prevalence was 8.3%, 29.2% and 23.8% respectively among HIV-negatives. The most prevalent genotypes were HPV-58 (16.9%), HPV-6 (15.6%) and HPV-11 (15.0%) among HIV-positive men, and were HPV-16 (4.4%), HPV-52 (4.4%) and HPV-6 (3.5%) among HIV-negative men. Having ever had haemorrhoids and having ever seen blood on tissue after defaecation was associated with HPV infection. One-fourth of the HPV infections in this study population can be covered by the quadrivalent vaccine in market. The highly prevalent anal HPV infection among men especially HIV-infected men calls for close observation and further investigation for anal cancer prevention.
We numerically study the impact of a compound drop on a hydrophobic substrate using a ternary-fluid diffuse-interface method, aiming to understand how the presence of the inner droplet affects the spreading dynamics and maximal spreading of the compound drop. First, it is interesting to see that the numerical results for an impacting pure drop agree well with the universal rescaling of maximal spreading ratio proposed by Lee et al. (J. Fluid Mech., vol. 786, 2016, R4). Second, two flow regimes have been identified for an impacting compound drop: namely jammed spreading and joint rim formation. The maximal spreading ratio of the compound drop is found to depend on the volume fraction of the inner droplet
, the surface tension ratio
, the Weber number and the flow regime. Moreover, we propose a universal rescaling of maximal spreading ratio for compound drops, by integrating the one for pure drops with a corrected Weber number that takes
and the flow regime into account. The predictions of the universal rescaling are in good agreement with the numerical results for impacting compound drops.
The pressure–strain-rate correlation and pressure fluctuations in convective and near neutral atmospheric surface layers are investigated. Their scaling properties, spectral characteristics, the contributions from the different source terms in the pressure Poisson equation and the effects of the wall are investigated using high-resolution (up to
) large-eddy simulation fields and through spectral predictions. The pressure–strain-rate correlation was found to have the mixed-layer and surface-layer scaling in the strongly convective and near neutral atmospheric surface layers, respectively. Its apparent surface-layer scaling in the moderately convective surface layer is due to the slow variations of the mixed-layer contribution, and is an inherent problem for single-point statistics in a multi-scale surface layer. In the strongly convective surface layer the pressure spectrum has an approximate
scaling range for small wavenumbers (
) due to the turbulent–turbulent contribution, and does not follow the surface-layer scaling, where
are the horizontal wavenumber and the distance from the surface respectively. The pressure–strain-rate cospectrum components have a
scaling range, consistent with our prediction using the surface layer parameters. It is dominated by the buoyancy contribution. Thus the anisotropy in the surface layer is due to the energy redistribution caused by the density fluctuations of the large eddies, rather than the turbulent–turbulent (inertial) effects. In the near neutral surface layer, the turbulent–turbulent and rapid contributions are primarily responsible for redistribution of energy from the streamwise velocity component to the vertical and spanwise components, respectively. The pressure–strain-rate cospectra peak near
, and have some similarities to those in the strongly convective surface layer for
. For the moderately convective surface layer, the pressure–strain-rate cospectra change signs at scales of the order of the Obukhov length, thereby imposing it as a horizontal length scale in the surface layer. This result provides strong support to the multipoint Monin–Obukhov similarity recently proposed by Tong & Nguyen (J. Atmos. Sci., vol. 72, 2015, pp. 4337–4348). We further decompose the pressure into the free-space (infinite domain), the wall reflection and the harmonic contributions. In the strongly convective surface layer, the free-space contribution to the pressure–strain-rate correlation is dominated by the buoyancy part, and is the main cause of the surface-layer anisotropy. The wall reflection enhances the anisotropy for most of the surface layer, suggesting that the pressure source has a large coherence length. In the near neutral surface layer, the wall reflection is small, suggesting a much smaller source coherence length. The present study also clarifies the understanding of the role of the turbulent–turbulent pressure, and has implications for understanding the dynamics and structure as well as modelling the atmospheric surface layer.
Litter size has a great impact on the profit of swine producers. Uterine development is an important determinant of reproduction efficiency and could hence affect litter size. Chinese Erhualian pig is one of the most prolific breeds in the world, even though large phenotypic variation in litter size was observed within Erhualian sows. To dissect the genetic basis of the phenotypic variation, we herein conducted genome-wide association studies for total number born and number born alive (NBA) of Erhualian sows. In total, one significant single nucleotide polymorphism (SNP) (P<1.78e−06) and 11 suggestive SNPs (P<3.57e−05) were identified on 10 chromosomes, confirming seven previously reported quantitative trait loci (QTL) and uncovering six QTL for litter size or uterus length. One locus on Sus scrofa chromosome (SSC) 13 (79.28 to 90.43 Mb) harbored a cluster of suggestive SNPs associated with multiparous NBA. The SNP (rs81447100) within this region was confirmed to be significantly (P<0.05) associated with litter size in Erhualian (n=313), Sutai (n=173) and Yorkshire (n=488) populations. Retinol binding protein 2 and retinol binding protein 1 functionally related to the development of uterus were located in a region of 2 Mb around rs81447100. Moreover, four genes related to embryo implantation and development were also detected around other significant SNPs. Taken together, our findings provide a potential marker (rs81447100) for the genetic improvement of litter size not only in Chinese Erhualian pigs but also in European commercial pig breeds like Yorkshire, and would facilitate the final identification of causative variant(s) underlying the effect of SSC13 QTL on litter size.
Compared with conventional identification methods, DNA-based genetic approaches such as single nucleotide polymorphisms (SNPs) and satellites are much more reliable for pig identification and meat traceability. In this study, multiallelic amplification fragments with multiple SNPs, incorporating the advantages of both SNPs and microsatellites, were explored for the first time for pig identification and meat traceability. Primer pairs for multiallelic fragments and their optimal SNPs were successfully selected and used for identification of individuals from Suzhong and Duroc populations. Meanwhile, the combined panel of the above mentioned primer pairs together with their optimal SNPs for Suzhong and/or Duroc pigs were validated for identification of the hybrids (Suzhong×Duroc). Therefore, we have successfully selected multiallelic amplification fragments with multiple SNPs to identify pigs and their meat samples from Suzhong, Duroc or their hybrids. Our study demonstrates that our method is more powerful for pig identification or meat traceability than SNPs or microsatellites.
Knowledge, attitudes and practices (KAP) of the population regarding severe fever with thrombocytopenia syndrome (SFTS) in endemic areas of Lu'an in China were assessed before and after an intervention programme. The pre-intervention phase was conducted using a sample of 425 participants from the 12 selected villages with the highest rates of endemic SFTS infection. A predesigned interview questionnaire was used to assess KAP. Subsequently, an intervention programme was designed and applied in the selected villages. KAP was re-assessed for each population in the selected villages using the same interview questionnaire. Following 2 months of the programme, 339 participants had completed the re-assessed survey. The impact of the intervention programme was evaluated using suitable statistical methods. A significant increase in the KAP and total KAP scores was noted following the intervention programme, whereas the proportion of correct knowledge, the positive attitudes and the effective practices toward SFTS of respondents increased significantly. The intervention programme was effective in improving KAP level of SFTS in populations that were resident in endemic areas.
Genomic selection has become increasingly important in the breeding of animals and plants. The response variable is an important factor, influencing the accuracy of genomic selection. The de-regressed proof (DRP) based on traditional estimated breeding value (EBV) is commonly used as response variable. In the current study, simulated data from 16th QTL-MAS Workshop and real data from Chinese Holstein cattle were used to compare accuracy and bias of genomic prediction with two methods of calculating DRP. Our results with simulated data showed that the correlation between genomic EBV and true breeding value achieved using the Jairath method (DRP_J) was superior to that achieved using the Garrick method (DRP_G) for simulated trait 1 but the reverse was true for simulated trait 3, and these two methods performed comparably for simulated trait 2. For all three simulated traits, DRP_J yielded larger bias of genomic prediction. However, DRP_J outperformed DRP_G in both accuracy and unbiasedness for four milk production traits in Chinese Holstein. In the estimation of genomic breeding value using genomic BLUP model, two methods for weighting diagonal elements of incidence matrix associated with residual error were also compared. With increasing the proportion of genetic variance unexplained by markers, the accuracy of genomic prediction was decreased and the bias was increased. Weighting by the reliability of DRP produced accuracy comparable to the evaluation where the proportion of genetic variance unexplained by markers was considered, but with smaller bias in general.
The effect of the initial phase distortion of the laser on near-field transmission characteristics in free space is investigated both numerically and theoretically. It is demonstrated and proposed that the near-field modulation and fluence contrast of the output laser beam are changing with the increase of both spatial low- and high-frequency wavefront distortion. The simulation results show that in order to ensure the beam quality in propagation, the Fresnel number should be controlled not <50 generally and the wavefront distortion should also be minimized by controlling both low- and high-frequency phase coefficient not larger than 0.6.
Upgraded heating and current drive (H/CD) systems have been equipped on the Experimental Advanced Superconducting Tokamak (EAST). With the upgraded H/CD systems, the operation space of EAST is extended, and the ability to achieve higher performance is improved. In this paper, a 0.5 dimension transport code named Minute Embedded Tokamak Integrated Simulator (METIS) is applied to predict the EAST operation space and to assess the current drive capability of the 4.6 GHz lower hybrid current drive system. Predictive simulation of several EAST scenarios, including steady-state high confinement mode (H-mode), advanced regime, high normalized beta and high electron temperature, are also performed with the available H/CD systems. The simulation results provide a guidance for forthcoming advanced EAST experiments.
This cross-sectional study aimed to examine and compare prevalence and correlates of whole blood Epstein–Barr virus (EBV) DNA between HIV-positive and HIV-negative men who have sex with men (MSM). Five hundred and four HIV-positive MSM and 504 age-matched HIV-negative MSM were recruited from an HIV counseling and testing clinic in Shanghai, China from November 2014 to November 2015 and were administered with a face-to-face questionnaire interview. Whole blood EBV DNA was tested by nested polymerase chain reaction assays on EBNA-1, EBNA-2, and LMP-1 genes. The prevalence of whole blood EBV DNA was 56·0% (95% CI 51·7–60·3%) among HIV-positive MSM and 26·0% (95% CI 22·4–30·0%) among HIV-negative MSM. Whole blood EBV DNA positivity was significantly associated with HIV infection (adjusted odds ratio (aOR) 3·43, 95% CI 2·58–4·57) and frequent intake of pickled, smoked, or salty food (aOR 1·71, 95% CI 1·02–2·86) in the whole sample, and with <200 cells/μl CD4 cell counts (aOR 1·79, 95% CI 1·05–3·05) and pickled, smoked, or salty food intake (aOR 3·14, 95% CI 1·39–7·08) in HIV-positive group. HIV-infected MSM are at higher risk of active EBV replication than HIV-uninfected MSM, underscoring needs of surveillance and research on EBV-related carcinogenesis in this population.
Studies from high-income countries report moderate-to-strong positive associations between alcohol use disorder (AUD) and other mental disorders, but there is little evidence about the comorbidity of AUD from low-and-middle-income countries.
A sample of 74 752 adults from five provinces that account for >12% of China's adult population was screened using the General Health Questionnaire, and the Structured Clinical Interview for DSM-IV was administered by psychiatrists to a subsample of 9619 males. The associations between AUD and other mental disorders at each site and the characteristics of men with AUD with and without comorbid mental disorders were estimated using logistic regression and summarized across sites using meta-analysis. Generalized estimation equations estimated the associations between the clinical features of alcohol dependence and comorbidity.
Robust inverse associations were found between current AUD and any mood disorder (adjusted OR = 0.6, 95% CI = 0.4–0.8) and any anxiety disorder (OR = 0.5, 95% CI = 0.3–1.0). Compared with men without AUD, men with AUD without comorbid disorders were more likely to be middle-aged, to be currently married, and to have higher family incomes. Men with comorbid AUD and other disorders were more likely to have the clinical features of alcohol dependence than men with AUD without comorbid disorders.
Inverse associations between AUD and other mental disorders and the higher social status of men with AUD than men without AUD found in this large, representative sample of community-dwelling Chinese males highlight the importance of considering the local substance-use culture when designing clinical or preventive interventions for addictive conditions.
The sealed tube Zn reduction method has been applied for small-mass samples ranging from 15 to 100 μg carbon preparation for accelerator mass spectrometry (AMS) radiocarbon (14C) measurements at the AMS-14C Preparation Lab in Guangzhou Institute of Geochemistry, Chinese Academy of Sciences (GIGCAS). The volume of the sealed reactor tube is reduced to ~0.75 cm3 in order to increase the yield of graphite. Graphite targets are measured at the Keck Carbon Cycle AMS Facility at the University of California, Irvine (KCCAMS). The targets generate a maximum 12C+1 current of about 0.5 μA per 1 μg C. The modern-carbon background is estimated to be 0.25–0.60 μg C, and dead-carbon background to be ~0.3–0.9 μg C. Both modern-carbon background and dead-carbon background are size dependent, so the results can be corrected. The precision of the small-mass modern carbon standard samples is±15–25‰ for the size of ~15–20 μg C,±5–10‰ for ~20–50 μg C, and±3–10‰ for 50–100 μg C. Further reduction of dead-carbon and modern-carbon contamination is needed in preparation of small-mass samples at GIGCAS.
An outbreak of acute hepatitis recently occurred in a nursing home in Zhejiang Province, China. The objectives of this study were to confirm the outbreak and identify the aetiology, source and transmission patterns. All residents and staff in or near the nursing home during the period from 1 October 2014 to 21 May 2015 were investigated regarding hygiene and for epidemiological information including water and food (eating meat especially pork products). Serum and stool specimens were collected for detection of hepatitis E virus (HEV) antibodies using ELISA and RNA using RT–PCR. Samples that were RNA positive were genotyped. Of 185 senior residents and 24 staff in the nursing home, there were 37 laboratory-confirmed cases during the outbreak. Of these cases, 12 patients (three deaths) were symptomatic with jaundice, a common clinical symptom for hepatitis E infection. HEV strains were isolated from three cases and they formed a single cluster within genotype 4d. A case-control study was conducted to investigate potential risk factors for the outbreak and the results revealed that cases more often washed their dishes and rinsed their mouths using tap water than the controls (P < 0·05). Based on hygiene investigation and meteorological information, it is likely that HEV-infected sewage and faeces contaminated the water network on rainy days. Collectively, these results suggest that the outbreak of HEV genotype 4 infection was most likely caused by contaminated tap water rather than food.
The historical evolution of an ancient forest that developed at Gaoyao, south China, can be divided into 4 stages of radiocarbon intervals (1.1–1.5, 2.0–3.5, 3.6–4.0, and 4.3–4.9 ka) in which the last 3 stages all developed in a wetland and formed humic layers of 2.0, 0.5, and 0.7 m depth, respectively. The humic layers were interrupted by 2 white-gray silty clay layers that most likely formed during climate fluctuations. Four drought events were identified during the evolution of the ancient forest, occurring around 4.3, 3.6, 2.0, and 1.1 ka, respectively, with durations of ∼1000 14C yr. These events are consistent with other records both in low- and high-latitude areas, in particular with the little ice ages occurring since the mid-Holocene. Precipitation likely increased from 5.0 to 3.6 ka in south China, then decreased, which is probably the main cause for the development as well as the demise of the ancient forest.
This paper examines the carbon isotopes (13C, 14C) of soil organic carbon (SOC) and soil CO2 from an evergreen broadleaf forest in southern China during the rainy season. The distribution of SOC δ13C, and SOC content with depth, exhibits a regular decomposition of SOC compartments with different turnover rates. Labile carbon is the main component in the topsoil (0–12 cm) and has a turnover rate between 0.1 and 0.01 yr–1. In the middle section (12–35 cm), SOC was mainly comprised of mediate carbon with turnover rates ranging between 0.01 and 0.025. Below 35 cm depth (underlayer section), the SOC turnover rate is slower than 0.001 yr–1, indicating that passive carbon is the main component of SOC in this section. The total production of humus-derived CO2 is 123.84 g C m–2 yr–1, from which 88% originated in the topsoil. The middle and underlayer sections contribute only 10% and 2% to the total humus-derived CO2 production, respectively. Soil CO2 δ13C varies from –24.7‰ to –24.0‰, showing a slight isotopic depth gradient. Similar to soil CO2 δ13C, Δ14C values, which range from 100.0‰ to 107.2‰, are obviously higher than that of atmospheric CO2 (60–70‰) and SOC in the middle and underlayer section, suggesting that soil CO2 in the profile most likely originates mainly from SOC decomposition in the topsoil. A model of soil CO2 Δ14C indicates that the humus-derived CO2 from the topsoil contributes about 65–78% to soil CO2 in each soil gas sampling layer. In addition, the humus-derived CO2 contributes ∼81% on average to total soil CO2 in the profile, in good agreement with the field observation. The distribution and origin of soil 14CO2 imply that soil CO2 will be an important source of atmospheric 14CO2 well into the future.
Accurate and reliable dating of paleosols, animal remains, and artifacts is of crucial importance in reconstructing environmental change and understanding the interrelationship between human activities and natural environments. Dating different materials in the same sample can help resolve problems such as soil carbon sources and carbon storage state. Conventional radiocarbon dating of soil (inorganic and organic matter) and accelerator mass spectrometry (AMS) dating of animal remains (fossil bones and teeth) result in different ages for materials from the same sample position in a typical loess section at Xinglong Mountain, Yuzhong County, Gansu Province in NW China. Inorganic matter is ∼3400 yr older than organic matter, 4175 ± 175 cal BP to 3808 ± 90 cal BP. A 1610-yr difference between the 14C ages of fossils (animal bones and teeth) and soil organic matter suggests that a depositional hiatus exists in the studied profile. The varying 14C ages of fossils and soil organic and inorganic matter have important implications for paleoclimate reconstructions from loess sections. It is critical to consider the meaning of the variable 14C ages from different material components from the same sample position in terms of soil organic and inorganic carbon storage, vegetation history reconstruction, archaeology, and the study of ancient civilizations.
While radiocarbon is widely applied in dating ancient samples, recent studies reveal that 14C concentrations in modern samples can also yield precise ages due to the atmospheric testing of thermonuclear devices between 1950 and 1963. 14C concentrations in both enamel and organic matter of 13 teeth from 2 areas in China were examined to evaluate and improve this method of forensic investigation. Choosing enamel near the cervix of the tooth can reduce the error caused by the difference between the sample formation time and whole enamel formation time because tooth enamel formations take a long time to complete. A proper regional data set will be helpful to get an accurate result when calculating the age of the sample (T1) by the CALIBomb program. By subtracting the enamel formation time (t), the birth date of an individual (T2) can be confirmed by enamel F14C from 2 teeth formed at different ages. Calculated enamel formation dates by 14C concentration are basically consistent with corresponding actual values, with a mean error of 1.9 yr for all results and 0.2 yr for the samples formed after AD 1960. This method is more effective for dating samples completed after AD 1960. We also found that 14C concentrations in organic matter of tooth roots are much lower than atmospheric concentrations in root formation years, suggesting that the organic material keeps turning over even after tooth formation is complete. This might be a potential tool for identification of death age to extract a proper component for 14C dating. We also observed that δ13C values between hydroxyapatite and organic matter indicate that isotopic fractionation during the biomineralization is 8–9%‰ more positive in mineral fractions than in organic matter.
CaII K line has been measured regularly nearly every month since 1974 at Kitt Peak. Our study on the data of CaII K profiles over two solar cycles indicates that in center disc spectra, the distance of the K1 component of the CaII K line from the line center and its intensity show periodical variations, but has a time delay of 1.2 years with respect to the sunspot number. Non-LTE computation implies a cyclic temperature variation of about 17 K of the temperature minimum region (TMR) in the quiet-Sun atmosphere and a cyclic variation of about 15-20 km of the position of the TMR.
Pathogens utilize type III secretion systems to deliver effector proteins, which facilitate bacterial infections. The Escherichia coli type III secretion system 2 (ETT2) which plays a crucial role in bacterial virulence, is present in the majority of E. coli strains, although ETT2 has undergone widespread mutational attrition. We investigated the distribution and characteristics of ETT2 in avian pathogenic E. coli (APEC) isolates and identified five different ETT2 isoforms, including intact ETT2, in 57·6% (141/245) of the isolates. The ETT2 locus was present in the predominant APEC serotypes O78, O2 and O1. All of the ETT2 loci in the serotype O78 isolates were degenerate, whereas an intact ETT2 locus was mostly present in O1 and O2 serotype strains, which belong to phylogenetic groups B2 and D, respectively. Interestingly, a putative second type III secretion-associated locus (eip locus) was present only in the isolates with an intact ETT2. Moreover, ETT2 was more widely distributed in APEC isolates and exhibited more isoforms compared to ETT2 in human extraintestinal pathogenic E. coli, suggesting that APEC might be a potential risk to human health. However, there was no distinct correlation between ETT2 and other virulence factors in APEC.
