OBJECTIVES/GOALS: To truly improve health equity and accessibility, we must develop a diverse and inclusive workforce. The BUILD EXITO program developed as a collaboration between a network of undergraduate programs and a CTSA hub and now has become a sustainable resource that will outlive NIH funding. We will disseminate our successful model. METHODS/STUDY POPULATION: The BUILD EXITO program has completed 10 years of NIH funding, a partnership between OCTRI and Portland State University (PSU), to create a new model of research training for underrepresented and disadvantaged students. This model provides an opportunity to learn about clinical and translational research academic careers; participate in a research enhancement and professional development curriculum;have a long-term authentic research experience; and receive enhanced mentorship. BUILD EXITO includes PSU, and local and 3 US Pacific territory 2-year colleges. We have developed a sustainable plan that includes these core elements after NIH support for the program ends. We have tracked long-term student outcomes for entry into graduate programs and the research workforce. RESULTS/ANTICIPATED RESULTS: We will describe the experimental model and the network of university and community colleges in BUILD EXITO, including PSU, U of Alaska, and colleges in US territories of Guam, Northern Mariana Islands, and American Samoa. All these universities and colleges have high proportions of underrepresented and disadvantaged students. We will present data on characteristics of the >600 students who have participated in BUILD EXITO to demonstrate the diversity of the cohort. We will also describe 4-year degree completion, engagement in the research workforce, and entry into graduate or professional programs. We will show how this has positively affected faculty inclusion of students in research, institutional policies at the 2-year and 4-year programs, and how this model has become sustainable. DISCUSSION/SIGNIFICANCE: The BUILD EXITO program developed as a collaboration of the CTSA hub at OHSU and a highly diverse undergraduate programs. We have developed a successful model for training a diverse research workforce and will disseminate this sustainable model.

OBJECTIVES/GOALS: Supported by the State of Alabama, the Alabama Genomic Health Initiative (AGHI) is aimed at preventing and treating common conditions with a genetic basis. This joint UAB Medicine-HudsonAlpha Institute for Biotechnology effort provides genomic testing, interpretation, and counseling free of charge to residents in each of Alabama’s 67 counties. METHODS/STUDY POPULATION: Launched in 2017, as a state-wide population cohort, AGHI (1.0) enrolled 6,331 Alabamians and returned individual risk of disease(s) related to the ACMG SF v2.0 medically actionable genes. In 2021, the cohort was expanded to include a primary care cohort. AGHI (2.0) has enrolled 750 primary care patients, returning individual risk of disease(s) related to the ACMG SF v3.1 gene list and pre-emptive pharmacogenetics (PGx) to guide medication therapy. Genotyping is done on the Illumina Global Diversity Array with Sanger sequencing to confirm likely pathogenic / pathogenic variants in medically actionable genes and CYP2D6 copy number variants using Taqman assays, resulting in a CLIA-grade report. Disease risk results are returned by genetic counselors and Pharmacogenetics results are returned by Pharmacists. RESULTS/ANTICIPATED RESULTS: We have engaged a statewide community (>7000 participants), returning 94 disease risk genetic reports and 500 PGx reports. Disease risk reports include increased predisposition to cancers (n=38), cardiac diseases (n=33), metabolic (n=12), other (n=11). 100% of participants harbor an actionable PGx variant, 70% are on medication with PGx guidance, 48% harbor PGx variants and are taking medications affected. In 10% of participants, pharmacists sent an active alert to the provider to consider/ recommend alternative medication. Most commonly impacted medications included antidepressants, NSAIDS, proton-pump inhibitors and tramadol. To enable the EMR integration of genomic information, we have developed an automated transfer of reports into the EMR with Genetics Reports and PGx reports viewable in Cerner. DISCUSSION/SIGNIFICANCE: We share our experience on pre-emptive implementation of genetic risk and pharmacogenetic actionability at a population and clinic level. Both patients and providers are actively engaged, providing feedback to refine the return of results. Real time alerts with guidance at the time of prescription are needed to ensure future actionability and value.

The goose barnacle Conchoderma virgatum is a highly opportunistic epibiont, with observations of attachment to a wide range of marine organisms. Attachment to perciform fishes, however, is rarely observed. Observations of Conchoderma-osteichthyan associations in general are also rare in the southern hemisphere, especially in Australian waters. We present the first record of C. virgatum as an epibiont on an ephippid fish (Platax teira). This is also only the second known record of C. virgatum attached to a perciform fish in the Tasman Sea. We also summarize all previous known observations of C. virgatum in association with perciform fishes.

Using recently developed adjoint methods for computing the shape derivatives of functions that depend on magnetohydrodynamic (MHD) equilibria (Antonsen et al., J. Plasma Phys., vol. 85, issue 2, 2019; Paul et al., J. Plasma Phys., vol. 86, issue 1, 2020), we present the first example of analytic gradient-based optimization of fixed-boundary stellarator equilibria. We take advantage of gradient information to optimize figures of merit of relevance for stellarator design, including the rotational transform, magnetic well and quasi-symmetry near the axis. With the application of the adjoint method, we reduce the number of equilibrium evaluations by the dimension of the optimization space (${\sim }50\text {--}500$) in comparison with a finite-difference gradient-based method. We discuss regularization objectives of relevance for fixed-boundary optimization, including a novel method that prevents self-intersection of the plasma boundary. We present several optimized equilibria, including a vacuum field with very low magnetic shear throughout the volume.

The National Cryo-Electron Microscopy Facility (NCEF) at the National Cancer Institute was launched in May of 2017 to provide free and rapid access to high-resolution cryo-EM data collection to United States researchers working on problems of broad general relevance to cancer biology. The decision about suitability of projects for data collection is made on a first-come, first-served basis by NCEF staff and is based solely on the quality of the screening images provided, without need for a scientific proposal. Here we provide an overview of the operation of the facility, typical data collection procedures, and some insights that have emerged from the structures reported from data collected at the facility.

We examine the spatial patterns of near-surface air temperature (Ta) over a melting glacier using a multi-annual dataset from McCall Glacier, Alaska. The dataset consists of a 10-year (2005–2014) meteorological record along the glacier centreline up to an upper glacier cirque, spanning an elevation difference of 900 m. We test the validity of on-glacier linear lapse rates, and a model that calculates Ta based on the influence of katabatic winds and other heat sources along the glacier flow line. During the coldest hours of each summer (10% of time), average lapse rates across the entire glacier range from −4.7 to −6.7°C km−1, with a strong relationship between Ta and elevation (R2 > 0.7). During warm conditions, Ta shows more complex, non-linear patterns that are better explained by the flow line-dependent model, reducing errors by up to 0.5°C compared with linear lapse rates, although more uncertainty might be associated with these observations due to occasionally poor sensor ventilation. We conclude that Ta spatial distribution can vary significantly from year to year, and from one glacier section to another. Importantly, extrapolations using linear lapse rates from the ablation zone might lead to large underestimations of Ta on the upper glacier areas.

The shape gradient is a local sensitivity function defined on the surface of an object which provides the change in a characteristic quantity, or figure of merit, associated with a perturbation to the shape of the object. The shape gradient can be used for gradient-based optimization, sensitivity analysis and tolerance calculations. However, it is generally expensive to compute from finite-difference derivatives for shapes that are described by many parameters, as is the case for typical stellarator geometry. In an accompanying work (Antonsen, Paul & Landreman J. Plasma Phys., vol. 85 (2), 2019), generalized self-adjointness relations are obtained for magnetohydrodynamic (MHD) equilibria. These describe the relation between perturbed equilibria due to changes in the rotational transform or toroidal current profiles, displacements of the plasma boundary, modifications of currents in the vacuum region or the addition of bulk forces. These are applied to efficiently compute the shape gradient of functions of MHD equilibria with an adjoint approach. In this way, the shape derivative with respect to any perturbation applied to the plasma boundary or coil shapes can be computed with only one additional MHD equilibrium solution. We demonstrate that this approach is applicable for several figures of merit of interest for stellarator configuration optimization: the magnetic well, the magnetic ripple on axis, the departure from quasisymmetry, the effective ripple in the low-collisionality $1/\unicode[STIX]{x1D708}$ regime $(\unicode[STIX]{x1D716}_{\text{eff}}^{3/2})$ (Nemov et al. Phys. Plasmas, vol. 6 (12), 1999, pp. 4622–4632) and several finite-collisionality neoclassical quantities. Numerical verification of this method is demonstrated for the magnetic well figure of merit with the VMEC code (Hirshman & Whitson Phys. Fluids, vol. 26 (12), 1983, p. 3553) and for the magnetic ripple with modification of the ANIMEC code (Cooper et al. Comput. Phys. Commun., vol. 72 (1), 1992, pp. 1–13). Comparisons with the direct approach demonstrate that, in order to obtain agreement within several per cent, the adjoint approach provides a factor of $O(10^{3})$ in computational savings.

The shape gradient quantifies the change in some figure of merit resulting from differential perturbations to a shape. Shape gradients can be applied to gradient-based optimization, sensitivity analysis and tolerance calculation. An efficient method for computing the shape gradient for toroidal three-dimensional magnetohydrodynamic (MHD) equilibria is presented. The method is based on the self-adjoint property of the equations for driven perturbations of MHD equilibria and is similar to the Onsager symmetry of transport coefficients. Two versions of the shape gradient are considered. One describes the change in a figure of merit due to an arbitrary displacement of the outer flux surface; the other describes the change in the figure of merit due to the displacement of a coil. The method is implemented for several example figures of merit and compared with direct calculation of the shape gradient. In these examples the adjoint method reduces the number of equilibrium computations by factors of $O(N)$, where $N$ is the number of parameters used to describe the outer flux surface or coil shapes.

Background and aims: Many sustaining traumatic brain injury (TBI) suffer ongoing post-concussion symptoms (PCS). The Rivermead Post-Concussion Symptoms Questionnaire (RPQ) is widely used, although there is disagreement about its structure. This study compared the fit of published RPQ structures with a four-factor structure derived from a large adult sample with TBI in Tasmania.

Method: 661 adults with TBI completed the RPQ at approximately one month post injury. Exploratory factor analysis (EFA), using the first half of the sample (n = 330), suggested a four-factor solution. This was compared with models reported in the literature with the second half of the sample (n = 331), using structural equation modelling. Trajectory of recovery across these factors was examined within subsamples at 1, 3, 6 and 12 months following TBI.

Results: Inter-correlations between items were strongest for somatic, cognitive and emotional functioning items and the EFA identified a four-factor model. Fit was examined utilising bootstrapping for model comparison. The data at 1 month following TBI best fitted the four-factor model (CFI = .95, RMSEA = .060 (.049–.071) and factors had adequate internal consistency (r = .61–.89). This model appeared a good fit and clinically useful across time points to 12 months post injury.

Conclusions: Data best fitted a four-factor model, identified using a rigorous statistical approach. Clinicians and clinical researchers may use this preferred model to provide more specific measurement of the severity of PCS. Future research could attempt replication within international samples.

Whether monozygotic (MZ) and dizygotic (DZ) twins differ from each other in a variety of phenotypes is important for genetic twin modeling and for inferences made from twin studies in general. We analyzed whether there were differences in individual, maternal and paternal education between MZ and DZ twins in a large pooled dataset. Information was gathered on individual education for 218,362 adult twins from 27 twin cohorts (53% females; 39% MZ twins), and on maternal and paternal education for 147,315 and 143,056 twins respectively, from 28 twin cohorts (52% females; 38% MZ twins). Together, we had information on individual or parental education from 42 twin cohorts representing 19 countries. The original education classifications were transformed to education years and analyzed using linear regression models. Overall, MZ males had 0.26 (95% CI [0.21, 0.31]) years and MZ females 0.17 (95% CI [0.12, 0.21]) years longer education than DZ twins. The zygosity difference became smaller in more recent birth cohorts for both males and females. Parental education was somewhat longer for fathers of DZ twins in cohorts born in 1990–1999 (0.16 years, 95% CI [0.08, 0.25]) and 2000 or later (0.11 years, 95% CI [0.00, 0.22]), compared with fathers of MZ twins. The results show that the years of both individual and parental education are largely similar in MZ and DZ twins. We suggest that the socio-economic differences between MZ and DZ twins are so small that inferences based upon genetic modeling of twin data are not affected.

The effect of annual defoliation on leafy spurge-infested rangeland was investigated over a 5-yr period. Artificial defoliation was conducted once or twice annually at various phenological growth stages of leafy spurge to simulate grazing by herded Angora goats. Single defoliation treatments did not reduce total leafy spurge stem densities. Defoliation twice in a growing season for 4 consecutive yr reduced total leafy spurge stem densities by 55% over nondefoliated controls. Grass foliar cover and yield increased in all defoliation treatments. Despite the increased grass yield on single defoliation treatments, cattle use would likely be limited in these treatments due to the high density of leafy spurge stems. Stem densities of leafy spurge in twice-defoliated treatments should not deter cattle from grazing these sites, thereby increasing the available forage supply. The data suggest that repeated grazing of leafy spurge-infested rangeland within a growing season would be required to reduce stem densities adequately and increase cattle use and production from these sites.

Smooth brome and Kentucky bluegrass are introduced cool-season perennial grasses known to invade grasslands throughout North America. During the fall of 2005 and spring of 2006, we implemented a restoration study at six native prairie sites in eastern South Dakota that have been invaded by smooth brome and Kentucky bluegrass. Treatments included five herbicide combinations, a fall prescribed burn, and an untreated control to determine the potential of each for renovation of invaded native grasslands. Herbicide treatments tested were sulfosulfuron, imazapyr, imazapic + sulfosulfuron, and imazapyr + imazapic, and were applied in late September 2005 and mid-May 2006. Untreated control plots averaged 64% (± 3.1) smooth brome cover and 38% (± 5.5) Kentucky bluegrass cover after the third growing season. Smooth brome cover in herbicide treated plots ranged from 6 to 23% and Kentucky bluegrass cover ranged from 15 to 35% after the third growing season. Smooth brome cover was 20% (± 2.9) and Kentucky bluegrass cover was 19% (± 4.0) in burned plots after the third growing season. Spring and fall treatments had similar native plant cover after three growing seasons. Spring and fall application of 0.33 kg ai ha−1 imazapyr and 0.10 kg ai ha−1 imazapic + 0.16 kg ai ha−1 imazapyr had ≤ 10% smooth brome cover and increased native species cover after three growing seasons. Herbicides were effective at reducing cover of smooth brome and Kentucky bluegrass, and can be incorporated with other management strategies to restore prairie remnants.

Hoop structures bedded with crop residues are becoming increasingly popular for swine production in the northcentral United States. Compost made from bedding materials and swine manure can be used as a soil amendment. A 3-yr field experiment was conducted in Boone, IA, to determine how composted swine manure affected selected soil characteristics and nutrient uptake, growth, and seed production of corn and three weed species (giant foxtail, velvetleaf, and common waterhemp) grown in mixture with corn. Two soil management systems, designed to provide equivalent amounts of N to corn, were compared: one that received composted manure and an average of 118 kg N ha−1 as synthetic fertilizer and another that received no composted manure and an average of 143 kg N ha−1 as synthetic fertilizer. Soil organic matter, P, K, and early-season NO3-N levels were greater in the (+) compost system. The N concentration of velvetleaf shoots, the P concentration of giant foxtail and common waterhemp shoots, and the K concentration of shoots of all three weed species also were greater in the (+) compost system. Compost application consistently increased common waterhemp height, common waterhemp biomass, and velvetleaf height, but increased velvetleaf biomass in only 1 yr and had no effect on giant foxtail height or biomass. Measurements of weed seed production, conducted in the final year of the study, showed that compost increased velvetleaf and common waterhemp seed production but had no effect on giant foxtail seed production. Compost consistently increased corn height and leaf K concentration but generally had no effect on corn yield. Results of this study indicate that large differences can exist among crop and weed species in their response to soil amendments. Depending on the weed species present, use of composted swine manure may increase requirements for weed management in corn production systems.

Smooth brome (Bromus inermis) is an introduced, cool-season perennial, sod-forming grass that has been shown to invade both native cool-and warm-season grasslands throughout North America. During the fall of 2005 through spring 2007, we implemented a smooth brome removal study at five sites in eastern South Dakota. Sites were selected to represent a range of soil and environmental conditions. Seven fall herbicide treatments, five spring herbicide treatments, an untreated plot that was planted with a native seed mix, and an untreated control that received no herbicide or seed addition were applied at each location in fall 2005/spring 2006 and fall 2006/spring 2007. Based upon first-year results, three fall herbicide treatments and two spring herbicide treatments were added in fall 2006/spring 2007. Sites were seeded with a native plant mix within 2 wk following spring herbicide treatment. Smooth brome cover in untreated plots ranged from 73 to 99% at the conclusion of the study. Smooth brome cover on herbicide-treated plots ranged from 0 to 84% on 2005/2006 plots and 0 to 98% on 2006/2007 plots after three growing seasons. Native plant response varied by site and treatment, possibly due to competition from exotic weeds. Although several herbicides show promise for control of smooth brome, future response of native plants will be important in determining the proper timing and herbicide combination.

Multiplicative interaction models are common in the quantitative political science literature. This is so for good reason. Institutional arguments frequently imply that the relationship between political inputs and outcomes varies depending on the institutional context. Models of strategic interaction typically produce conditional hypotheses as well. Although conditional hypotheses are ubiquitous in political science and multiplicative interaction models have been found to capture their intuition quite well, a survey of the top three political science journals from 1998 to 2002 suggests that the execution of these models is often flawed and inferential errors are common. We believe that considerable progress in our understanding of the political world can occur if scholars follow the simple checklist of dos and don'ts for using multiplicative interaction models presented in this article. Only 10% of the articles in our survey followed the checklist.
