This study compared the protective effect of sodium selenite (SS) and selenomethionine (SeMet) on heat stress (HS) invoked porcine IPEC-J2 cellular damage and integrate potential roles of corresponding selenoprotein. Cells were cultured at 37 oC until 80% conﬂuence, and then subjected to four different conditions for 24 h: at 37 oC (Control), 41.5 oC (HS), 41.5 oC supplied with 0.42 µmol Se/L SS (SS) or SeMet (SeMet). HS significantly decreased cell viability, up-regulated mRNA and protein levels of HSP70, and down-regulated mRNA and protein levels of tight junction-related proteins (CLDN-1 and ZO-1). HS induced cells injury was associated with the up-regulation (P < 0.05) of 6 inflammation-related genes and 14 selenoprotein encoding genes, and down-regulation (P < 0.05) of 2 inflammation-related genes and 5 selenoprotein encoding genes. Compared with the HS group, SS and SeMet supplementation resulted in an increase (P < 0.05) in cell viability, decreases (P < 0.05) mRNA expression of HSP70 and 6 inflammation-related genes, and rescue (P < 0.05) of mRNA and protein levels of CLDN-1 and ZO-1. SS and SeMet supplementation changes expressions of 19 selenoprotein encoding genes in cells affected by HS. Both Se supplementation significantly recovered the protein level of GPX1 and increased SELP in the IPEC-J2 cells under HS, respectively. In summary, Se supplementation alleviated the negative impact of HS on IPEC-J2 cells, and their cellular protective effect was associates with regulation expression of selenoproteins, SeMet exhibited a better protective effect.
Echinococcus granulosus sensu stricto (s.s.), Echinococcus multilocularis and Echinococcus canadensis are the common causes of human echinococcosis in China. An accurate species identification tool for human echinococcosis is needed as the treatments and prognosis are different among species. The present work demonstrates a method for the simultaneous detection of these three Echinococcus species based on multiplex polymerase chain reaction (mPCR). Specific primers of this mPCR were designed based on the mitochondrial genes and determined by extensive tests. The method can successfully detect either separated or mixed target species, and generate expected amplicons of distinct size for each species. Sensitivity of the method was tested by serially diluted DNA, showing a detection threshold as less as 0.32 pg for both E. granulosus s.s. and E. canadensis, and 1.6 pg for E. multilocularis. Specificity assessed against 18 other parasites was found to be 100% except weakly cross-react with E. shiquicus. The assay was additionally applied to 69 echinococcosis patients and 38 healthy persons, confirming the high reliability of the method. Thus, the mPCR described here has high application potential for clinical identification purposes, and can further provide a useful tool for evaluation of serology and imaging method.
Alzheimer’s Disease (AD), characterized by deficits in memory and cognition and by behavioral impairment, is a progressive neurodegenerative disorder that influences more than 47 million people worldwide. Currently, no available drug is able to stop AD progression. Therefore, novel therapeutic strategies need to be investigated.
We analyzed the RNA sequencing data (RNA-seq) derived from the Gene Expression Omnibus (GEO) database to identify the differentially expressed mRNAs in AD. The AD mouse model Tg2576 was used to verify the effects of IGF-2. The Morris Water Maze was administered to test the role of IGF-2 in memory consolidation. In addition, we quantified cell apoptosis by the TUNEL assay. The levels of amyloid plaques and the levels of Aβ40 and Aβ42 in the hippocampus were also determined by immunohistochemistry and ELISA, respectively.
RNA-seq analysis revealed that IGF-2 was remarkably reduced in AD. The expression of the upstream genes PI3K and AKT and the downstream gene CREB in the PI3K signaling pathway was significantly increased in the hippocampus of Tg2576 mice cells treated with IGF-2. The Morris water maze test showed that IGF-2 improved memory consolidation in Tg2576 mice. The activity of caspase-3 was decreased in Tg2576 mice treated with IGF-2. Amyloid plaques in the hippocampus were reduced, and the levels of Aβ40 and Aβ42 were decreased. The above effects of IGF-2 on AD were blocked when the PI3K signaling pathway inhibitor wortmannin was added.
IGF-2 attenuates memory decline, oxidative stress, cell apoptosis and amyloid plaques in the AD mouse model Tg2576 by activating the PI3K/AKT/CREB signaling pathway.
In this contribution, we devoted ourselves to fabricating aggregation-induced emission (AIE) activity copolymers via one-pot combination of RAFT polymerization and Biginelli reaction for the first time. When the feeding ratio of TPB was 33.5%, the molar fraction of TPB was, respectively, about 14.2 and 22.5% in PEG-PTE1 copolymers by two-step strategy and PEG-PTE2 copolymers by one-pot strategy with the similar structure. The Mn of PEG-PTE1 increased to 59,300 from 52,800 of PEG-AE presoma with narrow PDI, which was more than Mn of PEG-PTE2 with 52,300. As compared with PEG-PTE2, when the feeding ratio of TPB was 48.6%, the molar fraction of TPB increased to 32.6% in PEG-PTE3. In aqueous solution, the as-obtained PEG-PTE2 copolymers can self-assemble into fluorescent organic nanoparticles (FONs) with 100–180 nm spherical morphology, the maximal emission peak of which presented at 460 nm with the obvious AIE phenomenon. Moreover, due to the low toxicity and excellent cell dyeing behavior, the as-prepared PEG-PTE2 copolymers displayed great potential for biomedical applications.
The present study investigated the effects of glutamine (GLN) pretreatment on CD4+ T cell polarisation and remote kidney injury in mice with gut-derived polymicrobial sepsis. Mice were randomly assigned to three groups: normal control fed with American Institute of Nutrition (AIN)-93G diet and two sepsis groups provided with either AIN-93G-based diet or identical components, except part of casein was replaced by GLN. Mice were given their respective diets for 2 weeks. Then, mice in the sepsis groups were performed with caecal ligation and puncture and were killed 72 h after the surgery. Blood, spleens and kidneys were collected for further examination. The results showed that sepsis resulted in decreased circulating and splenic total T lymphocyte and CD4+ T cell percentages, whereas IL-4-, and forkhead box p3 (Foxp3)-expressing CD4+ T cells percentages were up-regulated. Compared with the sepsis control group, pretreatment with GLN maintained blood T and CD4+ T cells and reduced percentages of IL-4- and Foxp3-expressing CD4+ T cells. Also, a more pronounced activation and increased anti-apoptotic Bcl-2 gene expression of splenic CD4+ T cells were observed. Concomitant with the decreased plasma IL-6, keratinocyte-derived chemokine (KC) levels, the gene expression of KC, macrophage inflammatory protein-2 and renal injury biomarker kidney injury molecule-1 (Kim-1) were down-regulated when GLN was administered. These findings suggest that antecedent of GLN administration elicit a more balanced blood T helper cell polarisation, sustained T cell populations, prevented splenic CD4+ T cell apoptosis and attenuated kidney injury at late phase of polymicrobial sepsis. GLN may have benefits in subjects at risk of abdominal infection.
Estimating the robot state within a known map is an essential problem for mobile robot; it is also referred to “localization”. Even LiDAR-based localization is practical in many applications, it is difficult to achieve global localization with LiDAR only for its low-dimension feedback, especially in environments with repetitive geometric features. A sensor-fusion-based localization system is introduced in this paper, which has the capability of addressing the global localization problem. Both LiDAR and vision sensors are integrated, making use of the rich information introduced by vision sensor and the robustness from LiDAR. A hybrid grid-map is built for global localization, and a visual global descriptor is applied to speed up the localization convergence, combined with a pose refining pipeline for improving the localization accuracy. Also, a trigger mechanism is introduced to solve kidnapped problem and verify the relocalization result. The experiments under different conditions are designed to evaluate the performance of the proposed approach, as well as a comparison with the existing localization systems. According to the experimental results, our system is able to solve the global localization problem, and the sensor-fusion mechanism in our system has an improved performance.
The excessive use of plastic, especially polystyrene (PS), has caused serious environmental pollution. The efficient utilization of plastics and the conversion of plastics into value-added carbon materials are the concerns of researchers. Herein, we propose novel “pyrolysis–deposition” method to convert one popular plastic substance, PS, into ordered mesoporous carbons (OMCs). During the synthesis process, PS is pyrolyzed into small organic gases under high temperature, which is then adsorbed through capillary adsorption into the mesoporous of SBA-15 in the presence of catalyst. The obtained OMCs have high specific surface area, uniform pore size, and ordered pore structure. The OMCs exhibit specific capacitance of 118 F/g at a current density of 0.2 A/g and electrochemical stability of 87.2% at a current density of 2 A/g after 5000 cycles. The pyrolysis–deposition strategy provides a new idea to convert waste plastics into high-performance carbon materials for electrochemical applications.
Compared with end pumping fiber combiner, one of the advantages for side pumping combiner is the unlimited pumping points, which means multi-point or cascaded side pumping can be realized. However, the loss mechanism of the cascaded structure is rarely discussed. In this paper, we present the numerical and experimental investigation about the loss mechanism of a two-stage-cascaded side pumping combiner based on tapered-fused technique. The influence of loss mechanism on the coupling efficiency and thermal load of the fiber coating is analyzed according to simulations and experiments with different tapering ratios for the first stage. Based on the analysis, a cascaded component with total pump coupling efficiency of 96.4% handling a pump power of 1088 W is achieved by employing 1018 nm fiber laser as the pump source. Future work to further improve the performance of a cascaded side pumping combiner is discussed and prospected.

Inheritance involves the vital interests of thousands of families, the interests of any spouse's creditor(s) and ensuring that property and assets are transferred securely. Unlike other jurisdictions where testaments are widely used, the Chinese are still reluctant to use wills or other testamentary dispositions to deal with property problems after one's death. Therefore, the intestate succession system continues to be an important way of dealing with inheritance disputes in many provinces in China. With the development of China's economy and the generally increasing wealth of its people, private enterprises, including self-employed households, have become an important force in economic development. The owners of these enterprises, that is, private entrepreneurs, tend to have even more wealth. They have more inheritable interests than others, and their heirs are more likely to fall into succession disputes. Civil law codification is currently underway in China. Therefore, we carried out a survey on the property ideas and inheritance habits of Chinese private entrepreneurs during the first two months of 2017. Owing to limited funds and the general Chinese tendency to avoid any discussion of property problems with strangers, we had to conduct this survey in a comparatively unusual way to discover entrepreneurs ‘attitudes. The responses came from students’ face-to-face interviews, which to some extent might influence the scientific validity of the survey. But they are indeed suggestive of what this special group of people with property might think. Owing to space limitations, we only present the data on the intestate inheritance system part of the questionnaire, analyse their responses on intestate inheritance and inheritance habits, summarising the characteristics of the respondents and analysing the main reasons given for their choices. From the analysis of this survey and studies of scholars ‘legislative proposals, we put forward suggestions on intestacy legislation for the Succession Part of the Civil Code based on Chinese individuals’ positions.
BASIC INFORMATION ABOUT THE RESPONDENTS
The survey was carried out by investigators from the Southwest University of Political Science and Law, China. During their winter holiday break in 2017, investigators visited entrepreneurs in their respective hometowns and recorded the questionnaires, eventually collecting 179 sets of responses from Sichuan, Chongqing, Hebei, Fujian, and other provinces and cities.
The accounting information of a firm is analogous to the characteristics of an organism that contain biological information that influences decisions; such characteristics result from organizational routines (genes). Organizational routines result from organizational learning, and learning from an associated company is an efficient approach for a new venture to establish routines. The study results revealed that the subsidiaries inherited routines from the parent companies related to financial ratios, so we suggest that people should judge the adequacy of a firm’s financial situation by not only referring to the standard of its industry but also to its parent company.
Hemostasis, a process which causes bleeding to stop, can be enhanced using chitosan; but the detailed mechanism is unclear. Red blood cells (RBCs) adhere to chitosan because of their opposite charges, but the adhesion force is small, 3.83 pN as measured here using an optical tweezer, such that the direct adhesion cannot be the sole cause for hemostasis. However, it was observed in this study that layer structures of aggregated RBCs were formed next to chitosan objects in both static and flowing environments, but not formed next to cotton and rayon yarns. The layer structure is the clue for the initiation of hemostatsis. Through the supporting measurements of zeta potentials of RBCs and pH's using blood-chitosan mixtures, it is proposed here that the formation of the RBC layer structure next to chitosan objects is due to the reduction of repulsive electric double layer force between RBCs, because of the association of H+ deprotonated from chitosan with COO− on RBC membrane, under the DLVO (Derjaguin-Landau-Verwey-Overbeek) theory. The results are beneficial for designing effective chitosan-based wound dressings, and also for general biomedical applications.
Synthetic photonic materials created by engineering the profile of refractive index or gain/loss distribution, such as negative-index metamaterials or parity-time-symmetric structures, can exhibit electric and magnetic properties that cannot be found in natural materials, allowing for photonic devices with unprecedented functionalities. In this article, we discuss two directions along this line—non-Hermitian photonics and topological photonics—and their applications in nonreciprocal light transport when nonlinearities are introduced. Both types of synthetic structures have been demonstrated in systems involving judicious arrangement of optical elements, such as optical waveguides and resonators. They can exhibit a transition between different phases by adjusting certain parameters, such as the distribution of refractive index, loss, or gain. The unique features of such synthetic structures help realize nonreciprocal optical devices with high contrast, low operation threshold, and broad bandwidth. They provide promising opportunities to realize nonreciprocal structures for wave transport.
A novel luminogen-functionalized SBA-15, denoted as SNT, was developed by incorporating tris(4-bromophenyl)amine (TBPA) into SBA-15 via a “fixation-induced emission” strategy. The emission of TBPA on the matrix of SBA-15 was greatly enhanced, making the SNT possible as a fluorescence sensor. Cefalexin, a typical antibiotic, was chosen as the model analyte to be assayed and sensitive detection performance was achieved. This is the first time for cefalexin to be detected by a fluorescent method. Moreover, the SNT can be recycled by simply washing with proper solvents then used for next detection. This work provides a strategy to greatly improve the emission characteristics of fluorophores, even if a mediocre small fluorophore. It can be extended to design practical fluorescent sensors with high performance and recyclability by this strategy.
The power scaling on mid-infrared Raman fibre lasers (RFLs) is in demand for applications in health, environment and security. In this paper, we present the simulated laser behaviours of the tellurite glass RFLs pumped by 300-W Tm-doped fibre lasers (TDFLs) at
for the first time. By combining the advantages of the TDFLs and tellurite fibre, the output power at
has reached over hundreds of watts by first-order Raman shift. Moreover, the cascaded RFLs have been demonstrated with a wavelength extension greater than
and output power of tens of watts. To maximize the output power and the slope efficiency of the RFLs, we further analyse the interaction between the Raman gain and cavity loss, which are determined by fibre length and output reflectance of the laser cavity.
Sulphur isotopes can be used as a powerful tool to trace fluid evolution and explore the formation of chimneys. To clarify the in situ S isotopic variations of sulphides at the micro-scale, we analyzed a sulphide chimney collected from the hydrothermal field in the East Pacific Rise 1–2° S using a sensitive high-mass-resolution ion micro-probe for stable isotopes (SHRIMP SI). Three mineral zones can be identified in the chimney: an external outer wall of porous anhydrite and colloform pyrite, an internal middle zone of sub-euhedral pyrite and massive chalcopyrite, and an inner zone of massive pyrite. The δ34SV-CDT values of the sulphides fall within the range 1.83–7.51 ‰ (avg. 4.05 ‰, n = 16), and S isotopic values increase from the core (3.09 ‰, n = 3) to the middle (3.78 ‰, n = 11) to the edge (6.99 ‰, n = 2). These results illustrate mineral crystallization processes and the mixing between seawater-derived S and magmatic–hydrothermal fluids during the growth of the chimney. The zones from the edge to the core are characterized by crystal morphologies of colloform/anhedral pyrite to massive pyrite with decreasing δ34S values, revealing multi-stage mineral deposition and sulphur isotopic fractionation. In contrast to the increase in δ34S values from the core to the edge in one profile (profile A), anomalously low δ34S values in fine-grained pyrite relative to chalcopyrite in another profile (profile B) in the middle zone result from S isotopic exchange between seawater SO42− and fluid H2S due to different fluid–seawater mixing, possibly caused by variations in permeability and porosity across the chimney.
A novel synthetic air data estimation method without using air data sensors is presented, and the method only relies on the information from the Navigation System (NS) and Flight Control System (FCS). The aircraft's aerodynamic model is also required to make a connection between the FCS control parameters and the NS measurements. The airspeed, angle of attack and sideslip, angular velocity and wind speed are defined as state vectors, and state equations are established through the aircraft's aerodynamic model and dynamics. Linear velocity and angular velocity provided by the navigation system are considered as the measurement vector. To deal with variable wind fields, a novel Initialised Three-step Extended Kalman Filter (ITEKF), which considers the wind speed as an unknown input, is developed to track the variation of wind speed. Simulation results based on a Generic Hypersonic Vehicle (GHV) model are presented and compared with an existing method. Factors affecting the method's accuracy include the navigation system accuracy and the aerodynamic model error, are also discussed.
This paper reports the measurement of the energy loss of protons at the energy of 100 keV penetrating a partially ionized hydrogen plasma. The plasma of ne ≈ 1015–16 cm−3; Te ≈ 1–2 eV and lifetime of about 8 µs is created by the hydrogen gas discharge. The experimental results show an increase of a factor of 2.8 in the energy loss, which are in good agreement with the Bethe, Standard Stopping Model, Li–Petrasso and Vlasov models’ predictions within the error limit. The Bethe–Bloch Coulomb logarithm term is found to increase by a factor of 4.0 for free electrons as compared with the situation where bound electrons prevail. The potential application of protons energy loss for diagnosing the electron density in plasma is proposed too.
Population decline among Asian horseshoe crabs in Asia is increasingly reported, but knowledge of their population and ecological status in China is limited. We conducted community interviews in 30 fishing villages around Beibu Gulf in Guangxi, China, to collect distribution information about the potential spawning/nursery grounds of Tachypleus tridentatus and Carcinoscorpius rotundicauda, and any imminent threats to their populations. Based on the results from 400 respondents we identified 45 potential spawning/nursery grounds distributed widely along the shores of Beibu Gulf. We visited 10 of these sites and verified the presence of juvenile horseshoe crabs by field surveys. Nearly all respondents reported an overall depletion in horseshoe crab populations from these 45 sites, which they attributed mainly to unsustainable fishing practices. Respondents who reported having seen horseshoe crab mating pairs on shores were mostly older people, which may suggest a considerable reduction in horseshoe crabs coming to the shores to spawn in recent years. The mean daily harvest of adult T. tridentatus offshore, as indicated by fishers, has declined from c. 50–1,000 in the 1990s to 0–30 individuals during 2011–2016. Our Wisdom of Crowds approach, supported by confirmatory field surveys, is a cost-effective method for assessing the population status of horseshoe crabs, and the level of threat they face. Similar approaches with other species are likely to be particularly valuable in the Asia–Pacific region, where well-structured population monitoring is largely unaffordable.
