The present study was undertaken to investigate the antiparasitic activity of extracellular products of Streptomyces albus. Bioactivity-guided isolation of chloroform extracts affording a compound showing potent activity. The structure of the compound was elucidated as salinomycin (SAL) by EI-MS, 1H NMR and 13C NMR. In vitro test showed that SAL has potent anti-parasitic efficacy against theronts of Ichthyophthirius multifiliis with 10 min, 1, 2, 3 and 4 h (effective concentration) EC50 (95% confidence intervals) of 2.12 (2.22–2.02), 1.93 (1.98–1.88), 1.42 (1.47–1.37), 1.35 (1.41–1.31) and 1.11 (1.21–1.01) mg L−1. In vitro antiparasitic assays revealed that SAL could be 100% effective against I. multifiliis encysted tomonts at a concentration of 8.0 mg L−1. In vivo test demonstrated that the number of I. multifiliis trophonts on Erythroculter ilishaeformis treated with SAL was markedly lower than that of control group at 10 days after exposed to theronts (P < 0.05). In the control group, 80% mortality was observed owing to heavy I. multifiliis infection at 10 days. On the other hand, only 30.0% mortality was recorded in the group treated with 8.0 mg L−1 SAL. The median lethal dose (LD50) of SAL for E. ilishaeformis was 32.9 mg L−1.
Northeastern China is a region of high tick abundance, multiple tick-borne pathogens and likely human infections. The spectrum of diseases caused by tick-borne pathogens has not been objectively evaluated in this region for clinical management and for comparison with other regions globally where tick-transmitted diseases are common. Based on clinical symptoms, PCR, indirect immunofluorescent assay and (or) blood smear, we identified and described tick-borne diseases from patients with recent tick bite seen at Mudanjiang Forestry Central Hospital. From May 2010 to September 2011, 42% (75/180) of patients were diagnosed with a specific tick-borne disease, including Lyme borreliosis, tick-borne encephalitis, human granulocytic anaplasmosis, human babesiosis and spotted fever group rickettsiosis. When we compared clinical and laboratory features to identify factors that might discriminate tick-transmitted infections from those lacking that evidence, we revealed that erythema migrans and neurological manifestations were statistically significantly differently presented between those with and without documented aetiologies (P < 0.001, P = 0.003). Twelve patients (6.7%, 12/180) were co-infected with two tick-borne pathogens. We demonstrated the poor ability of clinicians to identify the specific tick-borne disease. In addition, it is necessary to develop specific laboratory assays for optimal diagnosis of tick-borne diseases.
Eating away from home is associated with poor diet quality, in part due to less healthy food choices and larger portions. However, few studies account for the potential additional contribution of differences in food composition between restaurant- and home-prepared dishes. The present study aimed to investigate differences in nutrients of dishes prepared in restaurants v. at home.
Eight commonly consumed dishes were collected in twenty of each of the following types of locations: small and large restaurants, and urban and rural households. In addition, two fast-food items were collected from ten KFC, McDonald’s and food stalls. Five samples per dish were randomly pooled from every location. Nutrients were analysed and energy was calculated in composite samples. Differences in nutrients of dishes by preparation location were determined.

On average, both the absolute and relative fat contents, SFA and Na:K ratio were higher in dishes prepared in restaurants than households (P < 0·05). Protein was 15 % higher in animal food-based dishes prepared in households than restaurants (P<0·05). Quantile regression models found that, at the 90th quantile, restaurant preparation was consistently negatively associated with protein and positively associated with the percentage of energy from fat in all dishes. Moreover, restaurant preparation also positively influenced the SFA content in dishes, except at the highest quantiles.
These findings suggest that compared with home preparation, dishes prepared in restaurants in China may differ in concentrations of total fat, SFA, protein and Na:K ratio, which may further contribute, beyond food choices, to less healthy nutrient intakes linked to eating away from home.
Some convergence bounds of the minimal residual (MINRES) method are studied when the method is applied for solving Hermitian indefinite linear systems. The matrices of these linear systems are supposed to have some properties so that their spectra are all clustered around ±1. New convergence bounds depending on the spectrum of the coefficient matrix are presented. Some numerical experiments are shown to demonstrate our theoretical results.
Based on biophysical ice-core data collected in the landfast ice off Barrow, Alaska, USA, in 2002 and 2003, a one-dimensional ice–ocean ecosystem model was developed to determine the factors controlling the bottom-ice algal community. The data and model results revealed a three-stage ice-algal bloom: (1) onset and early slow growth stage before mid-March, when growth is limited by light; (2) fast growth stage with increased light and sufficient nutrients; and (3) decline stage after late May as ice algae are flushed out of the ice bottom. Stages 2 and 3 are either separated by a transition period as in 2002 or directly connected by ice melting as in 2003, when in situ light and nutrient enrichment experiments showed only light limitations. The modeled net primary production of ice algae (NPPAi) from March to June is 1.2 and 1.7 g Cm–2 for 2002 and 2003, respectively, within the range of previous observations. Model sensitivity studies found that overall NPPAi increased almost proportionally to the initial nutrient concentrations in the water column. A phytoplankton bloom (if it occurs as in 2002) would compete with ice algae for nutrients and lead to reduced NPPAi. About 45% of the NPPAi was exported to the shallow benthos.
Kawasaki disease, which is characterised by systemic vasculitides accompanied by acute fever, is regularly treated by intravenous immunoglobulin to avoid lesion formation in the coronary artery; however, the mechanism of intravenous immunoglobulin therapy is unclear. Hence, we aimed to analyse the global expression profile of serum exosomal proteins before and after administering intravenous immunoglobulin.
Two-dimensional electrophoresis coupled with mass spectrometry analysis was used to identify the differentially expressed proteome of serum exosomes in patients with Kawasaki disease before and after intravenous immunoglobulin therapy.
Our analysis revealed 69 differential protein spots in the Kawasaki disease group with changes larger than 1.5-fold and 59 differential ones in patients after intravenous immunoglobulin therapy compared with the control group. Gene ontology analysis revealed that the acute-phase response disappeared, the functions of the complement system and innate immune response were enhanced, and the antibacterial humoral response pathway of corticosteroids and cardioprotection emerged after administration of intravenous immunoglobulin. Further, we showed that complement C3 and apolipoprotein A-IV levels increased before and decreased after intravenous immunoglobulin therapy and that the insulin-like growth factor-binding protein complex acid labile subunit displayed reverse alteration before and after intravenous immunoglobulin therapy. These observations might be potential indicators of intravenous immunoglobulin function.
Our results show the differential proteomic profile of serum exosomes of patients with Kawasaki disease before and after intravenous immunoglobulin therapy, such as complement C3, apolipoprotein A-IV, and insulin-like growth factor-binding protein complex acid labile subunit. These results may be useful in the identification of markers for monitoring intravenous immunoglobulin therapy in patients with Kawasaki disease.
A sediment core (14DH-C01) obtained from the mouth of Gomso Bay, on the west coast of South Korea, was used to obtain high-resolution palynomorph, grain-size, and 14C age data to investigate the Holocene sedimentary environment. The results indicated a transgressive depositional process with four stages controlled by sea-level change, as follows: river-dominated fluvial deposition from the early Holocene to 8.48 cal ka BP; tide-dominated tidal channel fill transgression from 8.48 to 8.08 cal ka BP; tide- to wave-dominated tidal channel fill transgression from 8.08 to 6.98 cal ka BP; and wave-dominated marine transgression from 6.98 cal ka BP to the present. Tidal channel filling was the primary mid-Holocene depositional process, accounting for the high sedimentation rate observed. The different hydrodynamics of the river-dominated, tide-dominated, tide- to wave-dominated, and wave-dominated processes following the changes in sea level may have controlled the transgressive depositional process. This transgressive sedimentary model differs from those of other large river mouth areas (e.g., the Changjiang River) since the mid-Holocene, perhaps resulting from the limited sediment supply in the study area.
In this paper we present new petrological and whole-rock geochemical data for the Palaeoproterozoic metasedimentary rocks in the upper part of the Ji'an Group within the Jiao–Liao–Ji Belt, China, as well as zircon U–Pb age dates and in situ Lu–Hf isotope data. The new data improve our understanding of the original nature of the metasedimentary rocks, further providing insights into their tectonic setting and the evolutionary history of the northern segment of the Jiao–Liao–Ji Belt. The zircons can be divided into two groups, viz., one of magmatic origin and the other of metamorphic origin. Zircon U–Pb dating gave mean or statistical peak ages for the magmatic zircons at 2035, 2082, 2178, 2343–2421, 2451–2545, 2643–2814 and 2923–3446 Ma, and mean peak ages for the metamorphic zircons at 1855 and 1912 Ma, which indicate a maximum depositional age of 2.03 Ga and two-stage metamorphic events at c. 1.91 and 1.85 Ga for the metasedimentary rocks. Geochemical data show that (1) the protoliths of these rocks were mainly sandstones, greywackes and claystones, together with some shales; (2) the main sources of the sedimentary material were Palaeoproterozoic granites and acid volcanic rocks, with minor contributions from Archaean granitic rocks; and (3) the sediments were deposited in an active continental margin setting. Moreover, along the northeastern margin of the Eastern Block of the North China Craton there is evidence of ancient crustal materials as old as 3.76 Ga, and multiple crustal growth events at 3.23–3.05, 2.80–2.65, 2.54–2.45 and 2.28–2.08 Ga.
The mitochondrial genome is maternally inherited in animals, despite the fact that paternal mitochondria enter oocytes during fertilization. Autophagy and ubiquitin-mediated degradation are responsible for the elimination of paternal mitochondria in Caenorhabditis elegans; however, the involvement of these two processes in the degradation of paternal mitochondria in mammals is not well understood. We investigated the localization patterns of light chain 3 (LC3) and ubiquitin in mouse and porcine embryos during preimplantation development. We found that LC3 and ubiquitin localized to the spermatozoon midpiece at 3 h post-fertilization, and that both proteins were colocalized with paternal mitochondria and removed upon fertilization during the 4-cell stage in mouse and the zygote stage in porcine embryos. Sporadic paternal mitochondria were present beyond the morula stage in the mouse, and paternal mitochondria were restricted to one blastomere of 4-cell embryos. An autophagy inhibitor, 3-methyladenine (3-MA), did not affect the distribution of paternal mitochondria compared with the positive control, while an autophagy inducer, rapamycin, accelerated the removal of paternal mitochondria compared with the control. After the intracytoplasmic injection of intact spermatozoon into mouse oocytes, LC3 and ubiquitin localized to the spermatozoon midpiece, but remnants of undegraded paternal mitochondria were retained until the blastocyst stage. Our results show that paternal mitochondria colocalize with autophagy receptors and ubiquitin and are removed after in vitro fertilization, but some remnants of sperm mitochondrial sheath may persist up to morula stage after intracytoplasmic spermatozoon injection (ICSI).
This study was to analyze the microstructure, microhardness, tensile and fatigue performance of the welded joints performed by a fiber laser on 22MnB5 and dual-phase steels (DP590, DP980) in similar and dissimilar combinations. The result shows that the weld zone (WZ) basically consisted of lath martensite. The HAZ in these steels can be divided into 3 parts: quenched, incomplete quenched, and tempered region. The WZ had the highest hardness, and a soft zone existed in the HAZ of all steels. Inside the WZ of the dissimilar welded joints, two hardness subregions were observed due to the difference in the alloying elements of these steels. Tensile specimens of the 22MnB5–22MnB5 and 22MnB5–DP980 welded joints were all broken in HAZ, while the 22MnB5–DP590 welded joints failed in the DP590 base metal (BM). The BM had a higher fatigue life than the welded joints, and the fatigue failure of the 22MnB5 similar and 22MnB5–DP980 dissimilar welded joints respectively occurred in the HAZ and DP980 BM. The fatigue fracture contained 3 parts: crack initiation, crack propagation, and the final fast fracture region.
The BiFeO3 (BFO) / PbTiO3 (PT) multiferroic ceramic composites with multilayered structure were prepared from orderly laminated BFO and PT tapes by tape casting method. The dielectric constant εr, loss tanδ, remnant polarization Pr and field-induced strain of BFO/PT ceramic composites were 140 (1 kHz), 5% (1 kHz), 12 µC/cm2 (at 80 kV/cm) and 0.06% (at 80 kV/cm) respectively, which were comparable to those pure BFO ceramics and BFO-based solid solutions
Surface properties affect the attachment of micro- and macroscopic marine organisms. The current study examined the settlement response of the mussel Mytilus coruscus plantigrades to natural biofilms formed on surfaces of different wettability. The percentages of plantigrade settlement were not influenced by the biofilms formed on variously wettable surfaces in the short term, but after 10 days, the plantigrade settlement rates decreased on biofilms formed on lower wettability surfaces. In general, lower wettability of the surfaces resulted in the decrease of the dry weight, bacterial and diatom density and the thickness of natural biofilms when compared to high wettability surfaces. In contrast, chlorophyll-a concentration in biofilms was independent of the initial wettability of the surfaces. Comparative cluster analysis of bacterial denaturing gradient gel electrophoresis patterns revealed that high variability existed between the bacterial community on high wettability surfaces and that on low wettability surfaces. Thus, surface wettability affects the formation of natural biofilms, and this variation in biofilms developed on different wettability surfaces may explain the discrepancy in their corresponding inducing activities on M. coruscus plantigrade settlement. This finding provides new insight into interactions between mussel settlement, biofilm characteristics and surface properties.
The mutualism between fig trees and their wasp pollinators is a model system for many ecological and evolutionary studies. However, the immature stages of pollinating fig wasps have rarely been studied. We monitored developing fig wasps of known ages and performed a series of dissections at 24 h intervals to identify key developmental traits of Ceratosolen solmsi marchali Mayr (Hymenoptera: Agaonidae), a pollinator of Ficus hispida L. (Moraceae). We identified where in the Ficus ovary eggs were deposited and time to hatch. We were also able to identify the timing and key underlying characters of five larval instars, three sub-pupal stages, and a single prepupal stage. We provide detailed morphological descriptions for the key stages and report some behavioral observations of the wasps in the several developmental stages we recorded. Scanning electron microscope images were taken.
The 0.6(Bi0.85La0.15)FeO3-0.4PbTiO3 (BLF-PT) ceramics were prepared by tape casting method. Effects of binder (polyvinylbutyl dibutyl PVB), plasticizer (phthalate-polyethylene glycol DBP-PEG) and dispersant (triethylolamine, TEA) concentration on the rheological properties of BLF-PT slurry were investigated. The optimized component ratio for ceramics powders, binder, plasticizer, dispersant and solvent (ethanol, EtOH) in the slurry was 50 wt.%, 4 wt.%, 6 wt.%, 1 wt.% and 39 wt.%. The dielectric constant εr, loss tanδ, and remnant polarization Pr of BLF-PT ceramics laminated from the tapes were 525 (1 kHz), 1.7% (1 kHz) and 30 μC/cm2 (45 kV/cm), respectively, which were comparable to those of BLF-PT ceramics prepared by traditional solid state reaction method.
