Horseweed and giant ragweed are competitive, annual weeds that can negatively impact crop yield. Biotypes of glyphosate-resistant (GR) giant ragweed and horseweed were first reported in 2008 and 2010 in Ontario, respectively. GR horseweed has spread throughout the southern portion of the province. The presence of GR biotypes poses new challenges for soybean producers in Canada and the United States. Halauxifen-methyl is a recently registered selective herbicide against broadleaf weeds for preplant use in corn and soybean. There is limited literature on the efficacy of halauxifen-methyl on GR horseweed and giant ragweed when combined with currently registered products in Canada. The purpose of this study was to determine the effectiveness of halauxifen-methyl applied alone and tank-mixed to control GR giant ragweed and GR horseweed in glyphosate and dicamba-resistant (GDR) soybean in southwestern Ontario. Six field experiments were conducted separately for each weed species over 2018 and 2019. Halauxifen-methyl applied alone offered 72% control of GR horseweed at 8 wk after application (WAA). Control was improved to >91% when halauxifen-methyl applied in combination with metribuzin, saflufenacil, chlorimuron-ethyl + metribuzin, and saflufenacil + metribuzin. At 8 WAA, halauxifen-methyl provided 11% control of GR giant ragweed, and 76% to 88% control when glyphosate/2,4-D choline, glyphosate/dicamba, glyphosate/2,4-D choline + halauxifen-methyl, and glyphosate/dicamba + halauxifen-methyl were used. We conclude that halauxifen-methyl applied preplant in a tank-mixture can provide effective control of GR giant ragweed and horseweed in GDR soybean.

Preplant (PP) herbicide applications are an important tool within an integrated weed management system, specifically in no-till production. An understanding of crop tolerance regarding PP applications is important for effectively integrating a new herbicide into no-till cropping systems. Twelve field trials (six in corn and six in soybean) were conducted over a 2-yr period (2018 and 2019) near Exeter and Ridgetown, ON. The purpose of these studies was to evaluate the tolerance of soybean and corn to halauxifen-methyl applied PP, PRE, or POST at the registered rate (5 g a.i. ha−1) and twice the registered rate (10 g a.i. ha−1), hereafter referred to as the 1× and 2× rate, respectively. All trials were kept weed-free throughout the growing season to remove the confounding effect of weed interference. Halauxifen-methyl applied 14 d preplant (DPP), 7 DPP, 1 DPP, and 5 d after seeding (DAS) at the 1× and 2× rates caused ≤10% visible soybean injury. In contrast, halauxifen-methyl applied POST (cotyledon–unifoliate stage, VE-VC) caused 67% to 87% visible soybean injury, a 50% to 53% reduction in height, 65% to 81% decrease in population, 56% to 67% lower biomass, and 53% to 63% decline in yield. Halauxifen-methyl applied 10 DPP, 5 DPP, 1 DPP, 5 DAS, and POST (spike–one leaf stage, VE-V1) at the 1× and 2× rate caused ≤3% visible corn injury and caused no effect on corn height or biomass. Halauxifen-methyl applied at VE-V1 at the 2× rate reduced corn yield 10%. Based on these studies, the current application restriction of 7 DPP in soybean and 5 DPP in corn is conservative and could be expanded. Expanding the application window of halauxifen-methyl would increase the utility of this herbicide for producers.

Horseweed is a competitive summer or winter annual weed that produces up to 230,000 small seeds per plant that are capable of traveling more than 500 km via wind. Giant ragweed is a tall, highly competitive summer annual weed. Glyphosate-resistant (GR) horseweed and GR giant ragweed pose significant challenges for producers in the United States and Ontario, Canada. It is thought that an integrated weed management (IWM) system involving herbicide rotation is required to control GR biotypes. Halauxifen-methyl is a new selective broadleaf POST herbicide registered for use in cereal crops; there is limited information on its efficacy on horseweed and giant ragweed. The purpose of this research was to determine the efficacy of halauxifen-methyl applied POST, alone and in a tank mix, for the control of GR horseweed and GR giant ragweed in wheat across southwestern Ontario. For each weed species, an efficacy study consisting of six field experiments was conducted over a 2-yr period (2018, 2019). At 8 wk after application (WAA), halauxifen-methyl, fluroxypyr/halauxifen-methyl, fluroxypyr/halauxifen-methyl + MCPA EHE, fluroxypyr + MCPA ester, 2,4-D ester, clopyralid, and pyrasulfotole/bromoxynil + ammonium sulfate controlled GR horseweed >95%. Fluroxypyr and MCPA provided only 86% and 37% control of GR horseweed, respectively. At 8 WAA, fluroxypyr, fluroxypyr/halauxifen-methyl, fluroxypyr/halauxifen-methyl + MCPA EHE, fluroxypyr + MCPA ester, fluroxypyr/halauxifen-methyl + MCPA EHE + pyroxsulam, 2,4-D ester, clopyralid, and thifensulfuron/tribenuron + fluroxypyr + MCPA ester controlled GR giant ragweed 87%, 88%, 90%, 94%, 96%, 96%, 98%, and 93%, respectively. Halauxifen-methyl and pyroxsulam provided only 45% and 28% control of GR giant ragweed, respectively. Halauxifen-methyl applied alone POST in the spring controlled GR horseweed but not GR giant ragweed in winter wheat.

Populations of eastern black nightshade suspected of being resistant to acetolactate synthase (ALS) inhibitors have been reported since 1999 in different locations in Ontario, Canada. This event has threatened the use of ALS inhibitors for control of this species. The objectives of this study were to evaluate the spectrum of resistance to different ALS-inhibiting herbicides and to examine the effectiveness of alternative modes of action herbicides. Growth room experiments were conducted to determine the response to imazethapyr and atrazine in seven suspected ALS inhibitor– resistant populations. One resistant and one susceptible population were further characterized for their response to ALS inhibitors and chloroacetamides. Seven populations were able to survive imazethapyr at 100 g ai/ha, while there was no resistance to atrazine. Compared to a susceptible (S) population, resistant (R) population SOLPT 1 had 726-, 31-, 6-, and 4-fold resistance to postemergence (POST) applied imazethapyr, imazamox, primisulfuron, and flumetsulam, respectively. Preemergence (PRE) application of imazethapyr, flumetsulam, cloransulam, nicosulfuron, prosulfuron, and rimsulfuron did not provide control of the R population, whereas they totally controlled the S population. The chloroacetamide herbicides metolachlor, dimethenamid, and flufenacet all provided at least 90% control of both R and S populations when applied PRE at the recommended field rates. The ALS inhibitors will not provide adequate control of these resistant populations, but acceptable control could be achieved with chloroacetamides or with atrazine.

Twelve populations of eastern black nightshade from different locations in Ontario are resistant to imazethapyr. This study aimed at determining the molecular basis of resistance in these populations and the activity of the resistant acetohydroxyacid synthase (AHAS) enzyme compared to that of the sensitive AHAS in response to different herbicides and branched-chain amino acid concentration. The results of partial AHAS sequencing indicated that all resistant populations had a cytosine331 to thymine substitution coding for an alanine205 to valine substitution. In vitro AHAS enzyme assays of one resistant population showed that the specific activity of the resistant enzyme was 56% less than that of the susceptible enzyme. AHAS from the resistant population was 72-, 70-, and 64-fold less sensitive than that of the susceptible population to imazethapyr, imazamox, and primisulfuron, respectively. Furthermore, the resistant enzyme was less sensitive to feedback inhibition from branched-chain amino acids compared to the susceptible enzyme. Results confirmed that resistance in resistant populations of eastern black nightshade was conferred by target-site modification and that the Ala205Val substitution alters the kinetics and regulation of branched-chain amino acid biosynthesis.

Two populations of Palmer amaranth from New Mexico have been confirmed to be resistant to glyphosate. In the present study, the molecular basis of resistance and the mode of inheritance of resistance in those populations were investigated. Quantitative real-time polymerase chain reaction analysis indicated up to an eightfold increase in genomic EPSPS copy number in glyphosate resistant plants compared with susceptible plants. The relative genomic EPSPS copy number of resistant plants was positively correlated with the relative EPSPS cDNA expression levels. Eight hours after treatment with glyphosate, the shikimate accumulation levels in resistant plants were negatively correlated with the genomic EPSPS copy numbers. Multiple sequencing of the EPSPS cDNA of resistant plants did not reveal any glyphosate resistance-conferring mutations. The evaluation of F1, reciprocal F1, and F2 Palmer amaranth families indicated that resistance to glyphosate does not follow a single-gene segregation pattern. Results suggest that the EPSPS amplification is the primary molecular basis of resistance in glyphosate resistant populations of Palmer amaranth from New Mexico.

Two populations of Palmer amaranth suspected of being resistant to glyphosate have been reported since 2007 in pecan orchards in Doña Ana County, New Mexico. The objectives of the study were to confirm and evaluate the level of resistance, to evaluate the effectiveness of alternative herbicide mechanisms of action, and to compare the cost of effective alternative herbicides for weed management in pecan orchards. Greenhouse experiments indicated that the resistant populations were able to survive glyphosate at 736 g ae ha−1. Compared with a susceptible (S) population, one of the resistant (R) populations had sevenfold resistance to glyphosate. POST application of 12 herbicides, with five different mechanisms of action, all provided at least 88% control of both R and S populations when applied at their recommended field rates. PRE application of trifluralin and metolachlor also provided more than 99% control of R and S populations. The results of field studies indicated that the financial benefit of season-long weed management with glyphosate, in pecan orchards, was comparable with some of the tested alternative herbicides.

Many substitutions in the herbicide target enzyme acetohydroxyacid synthase (AHAS) confer whole-plant resistance and may reduce plant fitness. This study was done to determine the impact of different watering and temperature regimes on the germination, growth, and seed production of eastern black nightshade populations resistant (R) to AHAS inhibitors as conferred by an Ala205Val substitution in their AHAS. Growth and reproductive ability of four R and four susceptible (S) populations were determined in growth-cabinet and greenhouse studies. The R populations had lower total berry and viable seed production per plant than S under optimal conditions because of slower berry maturation. Seed production of both S and R populations decreased under lower or higher than optimal watering regimes; however, this reduction was more pronounced for the S populations so that seed production was comparable across S and R. The R populations had significantly higher germination and vegetative growth under cooler alternating temperature regimes. Although there were no differences between R and S plants under stress conditions, under optimal growth conditions, the Ala205Val substitution comes at a significant cost in eastern black nightshade. Under optimal growth conditions and in the absence of herbicide selection, S populations should eventually dominate over R; however, the lack of fitness differences under stress conditions could enhance the persistence of the R individuals.

Resistance to acetohydroxyacid synthase (AHAS) inhibiting herbicides in populations of eastern black nightshade from Ontario has been linked to an Ala205Val substitution in the AHAS enzyme. The aim of this study was to determine the mechanism of inheritance of AHAS inhibitor resistance and the genetic relationships among resistant (R) and susceptible (S) eastern black nightshade populations from Ontario. Homozygous R and S parental populations were crossed and the inheritance was analyzed in F1 (S × R), reciprocal F1 (R × S), F2, and backcross (S × F1) progenies after application of imazethapyr at 150 g ai ha−1. Compared to parental lines, the progenies were rated as R, intermediate (I), and S phenotypes. All the F1 progenies were of the I phenotype. The backcross progenies segregated in a 1:1 (S:I) ratio, and the F2 families segregated in a 1:2:1 (R:I:S) ratio. These results indicate that a single nuclear gene, with incomplete dominance, controls resistance to AHAS-inhibiting herbicides in R population of eastern black nightshade. Random amplified polymorphic DNA (RAPD) markers were screened among 25 R and S populations. The genetic relationship of R and S populations based on RAPD profiles generated from six RAPD primers indicated four groups of populations in which resistance seems to have arisen independently. However, based on similarity coefficients, resistance within three of the groups could have arisen by gene flow. Both similar local selection pressure and gene flow could explain the spread of the Ala205Val substitution in R populations of eastern black nightshade in Ontario.

Theoretical models predict that seed size and seed-bank persistence evolve interdependently, such that strong selection for one trait corresponds with weak selection for the other. This framework has been supported and rejected by empirical data, and thus, conclusive evidence is lacking. We expanded the seed size–persistence framework to include seed-coat thickness, a defence trait previously correlated with seed survival in soil. To do this, we used Abutilon theophrasti accessions with varied evolutionary histories and we quantified associations among seed traits including morphology, size, coat thickness, dormancy (percentage of viable seeds that fail to germinate under optimum conditions) and seed-bank persistence (percentage of viable seeds remaining after 1 year of burial). Statistical models were developed to test the hypothesis that combined measurements of seed-coat thickness and seed size better explain variability in seed-bank persistence than seed-size data alone. Results indicated that measurements of seed size (length, width, mass) were negatively correlated with coat:width ratio (coat thickness relative to seed width) and coat:mass ratio (coat thickness relative to seed mass). Accessions characterized by smaller seeds with proportionally thicker seed coats were more dormant and more persistent in soil than accessions characterized by larger seeds with proportionally thinner seed coats. Seed-coat thickness data improved the explanatory power of logistic regression models for seed-size effects on both seed-bank persistence and dormancy. These results indicate that supplementing seed-size data with seed-defence data may clarify previously reported contradictory results regarding trade-offs between seed size and seed-bank persistence.
