Among nursing home outbreaks of coronavirus disease 2019 (COVID-19) with ≥3 breakthrough infections when the predominant severe acute respiratory coronavirus virus 2 (SARS-CoV-2) variant circulating was the SARS-CoV-2 δ (delta) variant, fully vaccinated residents were 28% less likely to be infected than were unvaccinated residents. Once infected, they had approximately half the risk for all-cause hospitalization and all-cause death compared with unvaccinated infected residents.

In recent years, unmanned aerial vehicle (UAV) technology has expanded to include UAV sprayers capable of applying pesticides. Very little research has been conducted to optimize application parameters and measure the potential of off-target movement from UAV-based pesticide applications. Field experiments were conducted in Raleigh, NC during spring 2018 to characterize the effect of different application speeds and nozzle types on target area coverage and uniformity of UAV applications. The highest coverage was achieved with an application speed of 1 m s−1 and ranged from 30% to 60%, whereas applications at 7 m s−1 yielded 13% to 22% coverage. Coverage consistently decreased as application speed increased across all nozzles, with extended-range flat-spray nozzles declining at a faster rate than air-induction nozzles, likely due to higher drift. Experiments measuring the drift potential of UAV-applied pesticides using extended-range flat spray, air-induction flat-spray, turbo air–induction flat-spray, and hollow-cone nozzles under 0, 2, 4, 7, and 9 m s−1 perpendicular wind conditions in the immediate 1.75 m above the target were conducted in the absence of natural wind. Off-target movement was observed under all perpendicular wind conditions with all nozzles tested but was nondetectable beyond 5 m away from the target. Coverage from all nozzles exhibited a concave-shaped curve in response to the increasing perpendicular wind speed due to turbulence. The maximum target coverage in drift studies was observed when the perpendicular wind was 0 and 8.94 m s−1, but higher turbulence at the two highest perpendicular wind speeds (6.71 and 8.94 m s−1) increased coverage variability, whereas the lowest variability was observed at 2.24 m s−1 wind speed. Results suggested that air-induction flat-spray and turbo air–induction flat-spray nozzles and an application speed of 3 m s−1 provided an adequate coverage of target areas while minimizing off-target movement risk.

A cementitious backfill has been proposed in many geological disposal concepts for intermediate-level waste and low-level waste in the UK and elsewhere. In this paper, the main features of the chemical evolution of backfill and the associated changes in the near-field pH are illustrated with results from recent work. For example, interaction of the groundwater with calcium silicate hydrate (C-S-H) phases in a backfill is expected to play an important role in the long-term pH-buffering behaviour. Existing experimental data for the dissolution of C-S-H gels are compared with recent experimental results from leach tests on gels of a lower calcium to silicon ratio (C/S) to provide a consistent set of data across the full C/S range. The results confirm that a congruent dissolution point around C/S = 0.8 is approached by leaching from below (i.e. for gels with 0.29 < C/S < 0.8), as well as from above, as reported elsewhere. In addition, a spreadsheet model has been developed to calculate the volume of backfill required at the vault scale to meet specified pH performance criteria. This model includes the major reactions of the backfill with the groundwater, waste encapsulants and waste components. It can also consider the effects of specific waste packages on local pH performance to allow comparison with the vault-scale calculations.

Drug-resistant Mycobacterium tuberculosis bacterium (MTB) is spreading worldwide. Three drug-resistant isolates were detected in Burmese, Hmong, and Indian immigrants currently residing in Milwaukee, Wisconsin, USA. Ion Torrent full-gene sequencing and complete genetic analysis was performed within 5 days and compared to results from traditional drug sensitivity testing (DST). Genetic characterization of seven, full-length resistance-associated genes revealed two MDR and one highly resistant strain with important drug-resistant mutations that were confirmed by traditional DST. The rapid turnaround from sample-to-sequence underscores the public health value of Ion Torrent full-gene sequencing of MDR/XDR genes from epidemiologically significant clinical isolates.

United Kingdom Nirex Limited develops and advises on safe, environmentally sound and publicly acceptable options for the long-term management of radioactive waste. One option Nirex has developed is a phased geological repository concept for intermediate level waste and some low level wastes that makes use of a combination of engineered and natural barriers. Physical containment of radionuclides would be achieved by immobilisation and packaging of wastes (mostly) in stainless steel containers.

Existing models of the migration of dissolved radionuclides from packaged wastes suggest that radionuclide release is determined largely by the rate of diffusion through the encapsulation grout used to immobilise the waste. The use of such models requires diffusion coefficient data for radionuclides in waste encapsulation grouts. This paper describes a programme of through-diffusion experiments, and modelling interpretation, aimed at deriving diffusion coefficients for some radionuclides in two types of encapsulation grout.

An intrinsic diffusion coefficient of HTO of around 1×10−13 to 2×10−13 m2s−1 was determined for a 3:1 mix of blast furnace slag to ordinary Portland cement, compared to around 4×10−13 to 5×10−13 m2s−1 for a 3:1 mix of pulverised fuel ash to ordinary Portland cement. These values are lower than that assumed for a non-sorbing radionuclide in an earlier modelling exercise. Porosity values around 0.3 were obtained in each case. For 36Cl as chloride, the experiments showed no significant breakthrough over the experimental timescale of about one year, suggesting an intrinsic diffusion coefficient below 5×10−13 m2s−1. One possibility is that chlorine-containing solids are precipitating in the cement. An intrinsic diffusion coefficient for 137Cs in the 3:1 mix of pulverised fuel ash to ordinary Portland cement of 4×10−15 m2s−1 was estimated, significantly lower than that determined for HTO.

The results from three of the sixteen experiments could not be fitted with a simple diffusion model, and for a further five experiments there was some doubt as to whether simple diffusion behaviour had been observed. It is suggested that this may have been due to cracks in the grouts that were sufficiently large to affect the diffusion properties of the grouts, although none was visible to the naked eye. Cracking of the waste encapsulation grouts could provide a mechanism for enhanced migration of radionuclides from waste packages, compared with diffusion in a homogeneous porous medium alone.

The gap junction (GJ) is an aggregate of intercellular channels that facilitates cytoplasmic interchange of ions, second messengers, and other molecules of less than 1000 Da between cells. In excitable organs such as heart and brain, GJs configure extended intercellular pathways for stable and long-term propagation of action potential. In a previous study in adult rat heart, we have shown that the Drosophila disks-large related protein ZO-1 shows low to moderate colocalization at myocyte borders with the GJ protein Cx43. In the present study, we detail a protocol for characterizing the pattern and level of colocalization of ZO-1 with Cx43 in cultures of neonatal myocytes at the level of individual GJ plaques. The data indicate that ZO-1 shows on average a partial 26.6% overlap (SD = 11.3%) with Cx43 GJ plaques. There is a strong positive correlation between GJ plaque size and area of ZO-1 colocalization, indicating that the level of associated ZO-1 scales with the area of the GJ plaque. Qualitatively, the most prominent colocalization occurs at the plaque perimeter. These studies may provide insight into the presently unknown biological function of ZO-1 interaction with Cx43.

Silicon carbide (SiC) based gas sensors have the ability to meet the needs of a range of aerospace applications including leak detection, environmental control, emission monitoring, and fire detection. While each of these applications require that the sensor and associated packaging be tailored for that individual application, they all require sensitive detection. The sensing approach taken to meet these needs is the use of SiC as a semiconductor in a Schottky diode configuration due to the demonstrated high sensitivity of Schottky diode-based sensors. However, Schottky diode structures require good control of the interface between the gas sensitive metal and SiC in order to meet required levels of sensitivity and stability. Two examples of effort to better control the SiC gas sensitive Schottky diode interface will be discussed. First, the use of chrome carbide as a barrier layer between the metal and SiC is discussed. Second, we report the first use of atomically flat SiC to provide an improved SiC semiconductor surface for gas sensor deposition. An example of the demonstration of a SiC gas sensor in an aerospace applications is given. It is concluded that, while significant progress has been made, the development of SiC gas sensor systems is still at a relatively early level of maturity for a number of applications.

Between 1 August and 15 September 2000, 361 cases of Salmonella enterica serotype Typhimurium definitive phage type (DT) 104, resistant to ampicillin, chloramphenicol, streptomycin, sulphonamides, spectinomycin and tetracycline (R-type ACSSuSpT), were identified in England and Wales residents. Molecular typing of 258 isolates of S. Typhimurium DT104 R-type ACSSuSpT showed that, although isolates were indistinguishable by pulsed-field gel electrophoresis, 67% (174/258) were characterized by a particular plasmid profile. A statistically significant association between illness and consumption of lettuce away from home was demonstrated (OR=7·28; 95% CI=2·25–23·57; P=0·0006) in an unmatched case–control study. Environmental investigations revealed that a number of food outlets implicated in the outbreak had common suppliers of salad vegetables. No implicated foods were available for microbiological testing. An environmental audit of three farms that might have supplied salad vegetables to the implicated outlets did not reveal any unsafe agricultural practices. The complexity of the food supply chain and the lack of identifying markers on salad stuffs made tracking salad vegetables back to their origin extremely difficult in most instances. This has implications for public health since food hazard warnings and product withdrawal are contingent on accurate identification of the suspect product.

It has been hypothesized that autism results from an ‘opioid peptide excess’. The aims of this study were to (1) confirm the presence of opioid peptides in the urine of children with autism and (2) determine whether dipeptidyl peptidase IV (DPPIV/CD26) is defective in children with autism. Opioid peptides were not detected in either the urine of children with autism (10 children; nine males, one female; age range 2 years 6 months to 10 years 1 month) or their siblings (10 children; seven males, three females; age range 2 years 3 months to 12 years 7 months) using liquid chromatography-ultraviolet-mass spectrometric analysis (LC-UV-MS). Plasma from 11 normally developing adults (25 years 5 months to 55 years 5 months) was also tested. The amount and activity of DPPIV in the plasma were quantified by an ELISA and DPPIV enzyme assay respectively; DPPIV was not found to be defective. The percentage of mononuclear cells expressing DPPIV (as CD26) was determined by flow cytometry. Children with autism had a significantly lower percentage of cells expressing CD3 and CD26, suggesting that they had lower T-cell numbers than their siblings. In conclusion, this study failed to replicate the findings of others and questions the validity of the opioid peptide excess theory for the cause of autism.

Data recording the course of scrapie outbreaks in 4 sheep flocks (2 in Cheviot sheep and 2 in Suffolks) are compared. For each outbreak the data on scrapie incidence and sheep demography and pedigrees cover periods of years or decades. A key finding is that the incidence of clinical cases peaks in sheep 2–3 years old, despite very different forces-of-infection. This is consistent with age-specific susceptibility of sheep to scrapie, as has been reported for cattle to bovine spongiform encephalopathy and for humans to variant Creutzfeldt–Jakob disease. Scrapie incidence was higher in ewes than rams and at certain times of years, though these effects were not consistent between flocks. There was no evidence for high levels of vertical transmission.

In order to establish a material system for packaging 500°C SiC microsystems, aluminum nitride (AlN) and aluminum oxide (Al2O3) were selected as packaging substrates, and gold (Au) thick-film materials were selected as substrate metallization material for electrical interconnection system (thick-film printed wires and thick-film metallization based wire-bond) and conductive die-attach interlayer. During a 1500-hour test in atmospheric oxygen with and without electrical bias, the electrical resistance of Au thick-film based interconnection system demonstrated low and stable electrical resistance at 500°C. The electrical interconnection system was also tested in extreme dynamic thermal environment. A silicon carbide (SiC) Schottky diode was attached to ceramic substrate using Au thick-film material as the conductive bonding layer and was successfully tested at 500°C in air for more than 1000 hours. In addition to the electrical test of die-attach in static thermal environments, nonlinear finite element analysis (FEA) was used for thermal mechanical evaluation and optimization of the die-attach in a wide temperature range.

Single crystal silicon carbide (SiC) has such excellent physical, chemical, and electronic properties that SiC based semiconductor electronics can operate at temperatures in excess of 600°C well beyond the high temperature limit for Si based semiconductor devices. SiC semiconductor devices have been demonstrated to be operable at temperatures as high as 600°C, but only in a probe-station environment partially because suitable packaging technology for high temperature (500°C and beyond) devices is still in development. One of the core technologies necessary for high temperature electronic packaging is semiconductor die-attach with low and stable electrical resistance. This paper discusses a low resistance die-attach method and the results of testing carried out at both room temperature and 500°C in air. A 1 mm2 SiC Schottky diode die was attached to aluminum nitride (AlN) and 96% pure alumina ceramic substrates using precious metal based thick-film material. The attached test die using this scheme survived both electronically and mechanically performance and stability tests at 500°C in oxidizing environment of air for 550 hours. The upper limit of electrical resistance of the die-attach interface estimated by forward I-V curves of an attached diode before and during heat treatment indicated stable and low attach-resistance at both room-temperature and 500°C over the entire 550 hours test period. The future durability tests are also discussed.
