Co-receptor tropism has been identified to correlate with HIV-1 transmission and the disease progression in patients. A molecular epidemiology investigation of co-receptor tropism is important for clinical practice and effective control of HIV-1. In this study, we investigated the co-receptor tropism on HIV-1 variants of 85 antiretroviral-naive patients with Geno2pheno algorithm at a false-positive rate of 10%. Our data showed that a majority of the subjects harboured the CCR5-tropic virus (81.2%, 69/85). No significant differences in gender, age, baseline CD4+ T-cell counts and transmission routes were observed between subjects infected with CXCR4-tropic or CCR5-tropic virus. The co-receptor tropism appeared to be associated with the virus genotype; a significantly more CXCR4-use was predicted in CRF01_AE infections whereas all CRF07_BC and CRF08_BC were predicted to use CCR5 co-receptor. Sequences analysis of V3 revealed a higher median net charge in the CXCR4 viruses over CCR5 viruses (4.0 vs. 3.0, P < 0.05). The predicted N-linked glycosylation site between amino acids 6 and 8 in the V3 region was conserved in CCR5 viruses, but not in CXCR4 viruses. Besides, variable crown motifs were observed in both CCR5 and CXCR4 viruses, of which the most prevalent motif GPGQ existed in both viral tropism and almost all genotypes identified in this study except subtype B. These findings may offer important implications for clinical practice and enhance our understanding of HIV-1 biology.
Starch digestion in the small intestines of the dairy cow is low, to a large extent, due to a shortage of syntheses of α-amylase. One strategy to improve the situation is to enhance the synthesis of α-amylase. The mammalian target of rapamycin (mTOR) signalling pathway, which acts as a central regulator of protein synthesis, can be activated by leucine. Our objectives were to investigate the effects of leucine on the mTOR signalling pathway and to define the associations between these signalling activities and the synthesis of pancreatic enzymes using an in vitro model of cultured Holstein dairy calf pancreatic tissue. The pancreatic tissue was incubated in culture medium containing l-leucine for 3 h, and samples were collected hourly, with the control being included but not containing l-leucine. The leucine supplementation increased α-amylase and trypsin activities and the messenger RNA expression of their coding genes (P <0.05), and it enhanced the mTOR synthesis and the phosphorylation of mTOR, ribosomal protein S6 kinase 1 and eukaryotic initiation factor 4E-binding protein 1 (P <0.05). In addition, rapamycin inhibited the mTOR signal pathway factors during leucine treatment. In sum, the leucine regulates α-amylase and trypsin synthesis in dairy calves through the regulation of the mTOR signal pathways.
We present results of a supernova lightcurve population synthesis, predicting the range of possible supernova lightcurves arising from a population of progenitor stars that include interacting binary systems. We show that the known diversity of supernova lightcurves can be interpreted as arising from binary interactions. Given detailed models of the progenitor stars, we are able to the determine what parameters within these stars determine the shape of their supernova lightcurve. The primary factors are the mass of supernova ejecta and the mass of hydrogen in the final progenitor. We find that there is a continuum of lightcurve behaviour from type IIP, IIL, to IIb supernovae related to the range of hydrogen and ejecta masses. Most type IIb supernovae arise from a relatively narrow range of initial masses from 10 to 15 M⊙. We also find a few distinct lightcurves that are the result of stellar mergers.
The present study aim to explore temporal–spatial patterns of water use (WU) efficiency and its influencing factors of maize production in China during 1998–2010. WU and productivity (WP) were quantified by taking irrigation loss into account and the links between WP and nine selected indicators were revealed by the partial least squares regression (PLSR) model. Results showed that national WU and WP in maize production were 138.56 cubic gigametres (Gm3; 0.755 green and 0.245 blue) and 1.079 kg/m3, respectively. WP was enhanced in the present study due to an increase in irrigated crop yield. Provinces located in the middle–lower part of the Yellow River had high proportions of green water and WP, while high proportions of irrigation water and low WP were found in Northwest China. The dosage of pesticides per unit area, relative humidity, average temperature and precipitation were the dominant factors that affected WP. However, the relationships between WP and solar radiation, fertilizer, agricultural machinery power, irrigation proportion and irrigated efficiency were not significant. Findings of the present research may also provide a reference for regional agricultural water management.
Prospective data on the associations between vitamin D intake and risk of CVD and all-cause mortality are limited and inconclusive. The aim of the present study was to investigate the associations between vitamin D intake and CVD risk and all-cause mortality in the Caerphilly Prospective Cohort Study.
The associations of vitamin D intake with CVD risk markers were examined cross-sectionally at baseline and longitudinally at 5-year, 10-year and >20-year follow-ups. In addition, the predictive value of vitamin D intake for CVD events and all-cause mortality after >20 years of follow-up was examined. Logistic regression and general linear regression were used for data analysis.

Higher vitamin D intake was associated with increased HDL cholesterol (P=0·003) and pulse pressure (P=0·04) and decreased total cholesterol:HDL cholesterol (P=0·008) cross-sectionally at baseline, but the associations were lost during follow-up. Furthermore, higher vitamin D intake was associated with decreased concentration of plasma TAG at baseline (P=0·01) and at the 5-year (P=0·01), but not the 10-year examination. After >20 years of follow-up, vitamin D was not associated with stroke (n 72), myocardial infarctions (n 142), heart failure (n 43) or all-cause mortality (n 281), but was positively associated with increased diastolic blood pressure (P=0·03).
The study supports associations of higher vitamin D intake with lower fasting plasma TAG and higher diastolic blood pressure.
APETALA2/ethylene-responsive factor (AP2/ERF) family transcription factors play a vital role in plant growth and in response to hormones and abiotic stresses. In the current research, it is reported that OsDERF2, one of the drought-responsive ERF, is a member of the DREB sub-family. OsDERF2 is a nuclear-localized protein and has transcriptional activity in yeast. Expression of OsDERF2 was induced by drought and inhibited by abscisic acid (ABA). However, OsDERF2 RNA interference (RNAi) knock-down transgenic lines enhanced tolerance to drought stress at seedling stage and were much more sensitive to ABA treatment, which may result from the increased ABA level in vivo. The basic leucine zipper (bZIP) transcription factor family plays an important role in the ABA signalling pathway of abiotic stress. Quantitative real-time polymerase chain reaction analysis revealed that the bZIP family gene OsbZIP20 and ABA-response gene OsABA45 were up-regulated 25 times and 120 times, respectively, in OsDERF2 RNAi knock-down lines under drought stress, which were up-regulated five and seven times in wild type under drought stress. The current data reveal that OsDERF2 negatively modulates drought stress response in an ABA-mediated pathway through regulating gene expression of other ABA-response transcription factors.
Worldwide 350 million people suffer from major depression, with the majority of cases occurring in low- and middle-income countries. We examined the patterns, correlates and care-seeking behaviour of adults suffering from major depressive episode (MDE) in China.
A nationwide study recruited 512 891 adults aged 30–79 years from 10 provinces across China during 2004–2008. The 12-month prevalence of MDE was assessed by the Modified Composite International Diagnostic Interview-short form. Logistic regression yielded adjusted odds ratios (ORs) of MDE associated with socio-economic, lifestyle and health-related factors and major stressful life events.
Overall, 0.7% of participants had MDE and a further 2.4% had major depressive symptoms. Stressful life events were strongly associated with MDE [adjusted OR 14.7, 95% confidence interval (CI) 13.7–15.7], with a dose–response relationship with the number of such events experienced. Family conflict had the highest OR for MDE (18.9, 95% CI 16.8–21.2) among the 10 stressful life events. The risk of MDE was also positively associated with rural residency (OR 1.5, 95% CI 1.4–1.7), low income (OR 2.3, 95% CI 2.1–2.4), living alone (OR 2.6, 95% CI 2.3–3.0), smoking (OR 1.4, 95% CI 1.3–1.6) and certain other mental disorders (e.g. anxiety, phobia). Similar, albeit weaker, associations were observed with depressive symptoms. Among those with MDE, about 15% sought medical help or took psychiatric medication, 15% reported having suicidal ideation and 6% reported attempting suicide.
Among Chinese adults, the patterns and correlates of MDE were generally consistent with those observed in the West. The low rates of seeking professional help and treatment highlight the great gap in mental health services in China.
Soluble starch synthase II (SSII) plays an important role in the biosynthesis of starch and in rice it consists of three isoforms encoded by SSII-1, SSII-2 and SSII-3. However, the genetic effects of various SSII alleles on grain quality have not been systematically characterized. In the present study, the japonica alleles on SSII-1, SSII-2 and SSII-3 (SSIIa) loci from a japonica cultivar, Suyunuo, were respectively introgressed by molecular marker-assisted selection into a typical indica cultivar, Guichao2, through successive backcrossing, generating three sets of near-isogenic lines (NILs). Grain quality and starch property analysis showed that NIL-SSII-3j exhibited significant decreases in the following parameters: amylose content, average granule size, and setback viscosity and consistency; but increases in peak viscosity, hot paste viscosity, gelatinization temperature and relative crystallinity. Moreover, the proportion of short amylopectin chains and branching degree also increased when compared with those of NIL-SSII-3i (Guochao2). Similar effects were observed in NIL-SSII-1j, and certain alterations in the fine structure of starch (granule size) were revealed. However, NIL-SSII-2j did not exert significant effect on grain quality and starch properties. In brief, among the SSII gene family, the functional diversity occurred on SSII-1 and SSII-3, and not on SSII-2. Therefore, it appears that more attention should be directed to SSII-1 and SSII-3 loci for improving the eating and cooking quality of rice.
The B type star 53 Persei was discovered in 1977 by Smith (1977) as the prototype of a separate group of B-type variables showing light and line profile variability. The physical cause of the variability was thought to be nonradial pulsation (NRP) (see, e.g. Smith et al. 1984). However, the NRP model for this star has been questioned by Balona (1986) who suggested the rotational modulation (RM) model to explain the variability. In order to resolve the long lasting debate about 53 Persei, a campaign was initiated to organize coordinated optical photometry and spectroscopy from the ground, and Far-UV photometry from Voyager in 1991 January. This paper presents the results of period analysis on the groundbased UBV data. In another paper, Smith & Huang (1994) report the new identification of pulsation modes using Voyager Far-UV photometry combined with the results from optical observations. Some preliminary results from APT uvby observations taken at a single site are also cited for comparison.
Omicron Andromedae is a multiple system of at least four stars: a B ↔Be star (component A), a spectroscopic binary (components B1–B2) and a close companion (component a). According to several studies (see Hill et al. 1988, 1989):
-the distance between A and the B1–B2 system decreased from 0.39″ in 1975 to 0.25″ in 1987 (McAlister and Hartkopf 1988)
-the few previous speckle measurements of component a have shown the possibility of a 3.7 years orbit around A, according to the 1975 to 1984 observations (mean distance 0.05″). The calculations with this 3.7 yr orbit lead to the prediction of a maximum distance of 0.77″ at 1992.738, i.e. at the end of september 1992, with a North-South orientation.
ET And is a binary system with a B9 Si star as the main component (Porb = 48.308d, e=0.46). Controversial claims in the literature concerning pulsation with periods ranging from few minutes to few hours and with variable amplitudes indicated a challenging target and motivated us to organize several photometric and spectroscopic observing campaigns. The problem with pulsation of ET And is that Teff and log g put this star in the cool domain of Slowly Pulsating B-type (SPB) stars, but the pulsation periods would be too short by a factor of about four, relatively to the shortest hitherto known periods for SPB stars.
Some results of the photometry multi-site observations of two δ Scuti stars, V624 Tau and HD 23194, are presented. The observations were carried out in the framework of a STEPHI network in 1999. We collected 343 hours of useful data and detected seven frequencies in V624 Tau and two frequencies in HD 23194.
Multisite photoelectric b and V photometry of the δ Scuti variable θ2(78) Tau has been obtained on three continents. Five close pulsation frequencies (13.22965, 13.48073, 13.69360, 14.31764 and 14.61454 cycles per day) with visual amplitudes between 0.001 and 0.007 mag were found. The four frequencies found in the previous campaign were confirmed. All three main data sets (1982-1986) are in excellent agreement with each other and fit the solution to ±0.0028 mag per single measurement. Over the four years the amplitudes and frequencies of pulsation were constant.
Chinese psychiatrists have gradually started to focus on those who are deemed to be at ‘clinical high-risk (CHR)’ for psychosis; however, it is still unknown how often those individuals identified as CHR from a different country background than previously studied would transition to psychosis. The objectives of this study are to examine baseline characteristics and the timing of symptom onset, help-seeking, or transition to psychosis over a 2-year period in China.
The presence of CHR was determined with the Structured Interview for Prodromal Syndromes (SIPS) at the participants' first visit to the mental health services. A total of 86 (of 117) CHR participants completed the clinical follow-up of at least 2 years (73.5%). Conversion was determined using the criteria of presence of psychotic symptoms (in SIPS). Analyses examined baseline demographic and clinical predictors of psychosis and trajectory of symptoms over time. Survival analysis (Kaplan–Meier) methods along with Log-rank tests were performed to illustrate the relationship of baseline data to either conversion or non-conversion over time. Cox regression was performed to identify baseline predictors of conversion by the 2-year follow-up.
In total 25 (29.1%) of 86 completers transitioned to a psychotic disorder over the course of follow-up. Among the CHR sample, the mean time between attenuated symptom onset and professional help-seeking was about 4 months on average, and converters developed fully psychotic symptoms about 12 months after symptom onset. Compared with those CHR participants whose risk syndromes remitted over the course of the study, converters had significantly longer delays (p = 0.029) for their first visit to a professional in search of help. At baseline assessment, the conversion subgroup was younger, had poorer functioning, higher total SIPS positive symptom scores, longer duration of untreated prodromal symptoms, and were more often given psychosis-related diagnoses and subsequently prescribed antipsychotics in the clinic.
Chinese CHR identified primarily by a novel clinical screening approach had a 2-year transition rate comparable with those of specialised help-seeking samples world-wide. Early clinical intervention with this functionally deteriorating clinical population who are suffering from attenuated psychotic symptoms, is a next step in applying the CHR construct in China.
Nutrients are essential for the health and survival of human beings and animals. Also, they play a major role in enhancing reproductive efficiency. The aim of the current study was to investigate the effects of sodium butyrate (SB) on reproductive performance and colostrum composition in gilts. A total of 40 Large White×Landrace replacement gilts (at the age of 160 to 175 days) were fed either a standard diet (control group, n=20) or standard diet top dressed with encapsulated SB at the level of 500 mg/kg (SB group, n=20) from 1 month before mating to 7 days after farrowing. The rate of gilts regular return to estrus after insemination was lower in SB group than the control group. The total number of piglets born (P=0.179) and the litter weight at birth (P=0.063) did not differ between the two treatment groups. However, the mean BW at day 7 tended to be greater in SB group (P=0.051) and average daily gain of piglets was greater (P=0.011) compared with control group. Colostrum samples were collected at parturition and the concentrations of total protein (P=0.197), cholesterol (P=0.161) and lactose (P=0.923) were not influenced by SB supplementation. However, compared with control gilts, colostrum from SB-treated gilts contained lower triglyceride (P=0.050). Moreover, colostrum concentrations of prolactin (P=0.005) and leptin (P=0.006) were significantly lower in SB group. No significant differences were noted for the colostral concentrations of cortisol (P=0.899), thyroxine (P=0.891) or triiodothyronine (P=0.194). The concentration of lipopolysaccharide in colostrum was not influenced by SB supplementation (P=0.972). However, colostrum from SB-treated gilts had significantly lower tumor necrosis factor α (TNFα) (P=0.030) and higher immunoglobulin A (IgA) (P=0.042). Collectively, SB supplementation could reduce the rate of gilts return to estrus, alter the composition of colostrum and enhance the growth rate of piglets. Moreover, SB could alter the immune function of newborn piglets through decreased production of TNFα and increased IgA concentration in colostrum.
Most previous studies on the effects of lipopolysaccharide (LPS) in pigs focused on the body’s immune response, and few reports paid attention to body metabolism changes. To better understand the glucose metabolism changes in skeletal muscle following LPS challenge and to clarify the possible mechanism, 12 growing pigs were employed. Animals were treated with either 2 ml of saline or 15 µg/kg BW LPS, and samples were collected 6 h later. The glycolysis status and mitochondrial function in the longissimus dorsi (LD) muscle of pigs were analyzed. The results showed that serum lactate content and NADH content in LD muscle significantly increased compared with the control group. Most glycolysis-related genes expression, as well as hexokinase, pyruvate kinase and lactic dehydrogenase activity, in LD muscle was significantly higher compared with the control group. Mitochondrial complexes I and IV significantly increased, while mitochondrial ATP concentration markedly decreased. Significantly increased calcium content in the mitochondria was observed, and endoplasm reticulum (ER) stress has been demonstrated in the present study. The results showed that LPS treatment markedly changes glucose metabolism and mitochondrial function in the LD muscle of pigs, and increased calcium content induced by ER stress was possibly involved. The results provide new clues for clarifying metabolic diseases in muscle induced by LPS.
