MeerTime is a five-year Large Survey Project to time pulsars with MeerKAT, the 64-dish South African precursor to the Square Kilometre Array. The science goals for the programme include timing millisecond pulsar (MSPs) to high precision ( ${<} 1 \unicode{x03BC} \mathrm{s}$ ) to study the Galactic MSP population and to contribute to global efforts to detect nanohertz gravitational waves with the International Pulsar Timing Array (IPTA). In order to plan for the remainder of the programme and to use the allocated time most efficiently, we have conducted an initial census with the MeerKAT ‘L-band’ receiver of 189 MSPs visible to MeerKAT and here present their dispersion measures, polarisation profiles, polarisation fractions, rotation measures, flux density measurements, spectral indices, and timing potential. As all of these observations are taken with the same instrument (which uses coherent dedispersion, interferometric polarisation calibration techniques, and a uniform flux scale), they present an excellent resource for population studies. We used wideband pulse portraits as timing standards for each MSP and demonstrated that the MeerTime Pulsar Timing Array (MPTA) can already contribute significantly to the IPTA as it currently achieves better than $1\,\unicode{x03BC}\mathrm{s}$ timing accuracy on 89 MSPs (observed with fortnightly cadence). By the conclusion of the initial five-year MeerTime programme in 2024 July, the MPTA will be extremely significant in global efforts to detect the gravitational wave background with a contribution to the detection statistic comparable to other long-standing timing programmes.

ABSTRACT IMPACT: Characterizing the cellular composition of human adipose tissue may contribute to the prevention and/or treatment of obesity-associated metabolic diseases. OBJECTIVES/GOALS: Our aims in this study were to use single-cell techniques 1. to characterize cell types within the stromavascular fraction of human adipose tissue, 2. to identify subsets of cells within each type (sub-clustering), 3. to identify gene sets and pathways that may provide information on the function and significance of each cell cluster. METHODS/STUDY POPULATION: Abdominal subcutaneous adipose tissue samples from n=6 healthy volunteers (1M, 5F, age 28-38 y, BMI 24.5-63.0 kg/m2) were collected by aspiration or during surgery. In 3 subjects, all females, paired femoral samples were also collected. After collagenase digestion approximately n=10,000 cells/sample were used for single-cell RNA sequencing using the 10X Genomics platform. After QC and downstream analysis, data were analyzed in Seurat v.3.1.5. We identified first different cell types and then subclusters in an unbiased fashion. Gene Set Enrichment Analysis (GSEA) was used for pathway analysis. RESULTS/ANTICIPATED RESULTS: Progenitor markers are related to extracellular matrix, eg DCN (logFC progenitors vs other cells types=3.17, expressed in 99.7% (pct.1) of progenitors, 26.1% (pct.2) of others). Endothelial and pericytes shared markers like RBP7 (logFC=1.67, pct.1 0.88, pct.2 0.39); pericytes also showed unique markers, eg RGS5 (logFC=2.29, pct.1 0.89, pct.2 0.17). Progenitors are further divided into 11 sub-clusters, one of which showed enrichment of CD36 (high proliferation potential), FABP4 (differentiation), and of the novel marker PALMD (logFC=7.13, pct.1 0.94, pct.2 0.48). All p<10E-5. GSEA analysis suggests that inflammatory pathways are downregulated in both adipose progenitors and endothelial/pericyte cells in the femoral compared to the abdominal depot. DISCUSSION/SIGNIFICANCE OF FINDINGS: Single cell RNA sequencing provides unique insights into the molecular profile of cell types and the identification of novel subsets of cells within the human adipose tissue. Such cellular heterogeneity may explain differences in adipose function between individuals and eventually in the risk of obesity-associated metabolic diseases.

We describe system verification tests and early science results from the pulsar processor (PTUSE) developed for the newly commissioned 64-dish SARAO MeerKAT radio telescope in South Africa. MeerKAT is a high-gain ( ${\sim}2.8\,\mbox{K Jy}^{-1}$ ) low-system temperature ( ${\sim}18\,\mbox{K at }20\,\mbox{cm}$ ) radio array that currently operates at 580–1 670 MHz and can produce tied-array beams suitable for pulsar observations. This paper presents results from the MeerTime Large Survey Project and commissioning tests with PTUSE. Highlights include observations of the double pulsar $\mbox{J}0737{-}3039\mbox{A}$ , pulse profiles from 34 millisecond pulsars (MSPs) from a single 2.5-h observation of the Globular cluster Terzan 5, the rotation measure of Ter5O, a 420-sigma giant pulse from the Large Magellanic Cloud pulsar PSR $\mbox{J}0540{-}6919$ , and nulling identified in the slow pulsar PSR J0633–2015. One of the key design specifications for MeerKAT was absolute timing errors of less than 5 ns using their novel precise time system. Our timing of two bright MSPs confirm that MeerKAT delivers exceptional timing. PSR $\mbox{J}2241{-}5236$ exhibits a jitter limit of $<4\,\mbox{ns h}^{-1}$ whilst timing of PSR $\mbox{J}1909{-}3744$ over almost 11 months yields an rms residual of 66 ns with only 4 min integrations. Our results confirm that the MeerKAT is an exceptional pulsar telescope. The array can be split into four separate sub-arrays to time over 1 000 pulsars per day and the future deployment of S-band (1 750–3 500 MHz) receivers will further enhance its capabilities.

We describe the design and deployment of GREENBURST, a commensal Fast Radio Burst (FRB) search system at the Green Bank Telescope. GREENBURST uses the dedicated L-band receiver tap to search over the 960–1 920 MHz frequency range for pulses with dispersion measures out to $10^4\ \rm{pc\,cm}^{-3}$ . Due to its unique design, GREENBURST is capable of conducting searches for FRBs when the L-band receiver is not being used for scheduled observing. This makes it a sensitive single pixel detector capable of reaching deeper in the radio sky. While single pulses from Galactic pulsars and rotating radio transients will be detectable in our observations, and will form part of the database we archive, the primary goal is to detect and study FRBs. Based on recent determinations of the all-sky rate, we predict that the system will detect approximately one FRB for every 2–3 months of continuous operation. The high sensitivity of GREENBURST means that it will also be able to probe the slope of the FRB fluence distribution, which is currently uncertain in this observing band.

Over the last decade or so, it has become clear that pulsars exhibit sudden and significant changes in their spin properties. At the same time, a better understanding of the geometry of young and older pulsars, is providing clues about the long-term evolution of the magnetic inclination angle. In this talk, we present a simple simulation of the pulsar population that takes into account current observational facts. We show how, with very few assumptions, the observed P-Ṗ diagram can be reproduced for a synthesized population. The implications are interesting and testable.

The Low Frequency Array (LOFAR) is ideally suited to pulsar scattering studies, providing broad bands at low frequencies where the imprints of the ionized Interstellar Medium (IISM) are exaggerated. We analyse a set of sources at 110–190 MHz, and find unexpectedly shallow dependencies of pulse scatter broadening on frequency. These anomalous scattering values are discussed by considering evidence for anisotropic scattering and small scattering clouds.

The next generation of radio telescopes will have unprecedented sensitivity and time-resolution offering exciting new capabilities in time-domain science. However, this will result in very large numbers of pulsar and transient event candidates and the associated data rates will be technically challenging in terms of data storage and signal processing. Automated detection and classification techniques are therefore required and must be optimized to allow high-throughput data processing in real time. In this paper we provide a summary of the emerging machine learning techniques being applied to this problem.

The recent revelation that there are correlated period derivative and pulse shape changes in pulsars has dramatically changed our understanding of timing noise as well as the relationship between the radio emission and the properties of the magnetosphere as a whole. Using Gaussian processes we are able to model timing and emission variability using a regression technique that imposes no functional form on the data. We revisit the pulsars first studied by Lyne et al. (2010). We not only confirm the emission and rotational transitions revealed therein, but reveal further transitions and periodicities in 8 years of extended monitoring. We also show that in many of these objects the pulse profile transitions between two well-defined shapes, coincident with changes to the period derivative. With a view to the SKA and other telescopes capable of higher cadence we also study the detection limitations of period derivative changes.

Here, we present initial results from the ALFABURST radio transient survey, which is currently running in a commensal mode with the ALFA receiver at the Arecibo telescope. We observed for a total of 1400 hours and have detected single pulses from known pulsars but did not detect any FRBs. The non-detection of FRBs is consistent with the current FRB sky rates.

The Square Kilometre Array will be an amazing instrument for pulsar astronomy. While the full SKA will be sensitive enough to detect all pulsars in the Galaxy visible from Earth, already with SKA1, pulsar searches will discover enough pulsars to increase the currently known population by a factor of four, no doubt including a range of amazing unknown sources. Real time processing is needed to deal with the 60 PB of pulsar search data collected per day, using a signal processing pipeline required to perform more than 10 POps. Here we present the suggested design of the pulsar search engine for the SKA and discuss challenges and solutions to the pulsar search venture.

An evolution of the low-frequency pulse profile of PSR B2217+47 is observed during a six-year observing campaign with the LOFAR telescope at 150 MHz. The evolution is manifested as a new component in the profile trailing the main peak. The leading part of the profile, including a newly-observed weak component, is steady during the campaign. The transient component is not visible in simultaneous observations at 1500 MHz using the Lovell telescope, implying a chromatic effect. A variation in the dispersion measure of the source is detected in the same timespan. Precession of the pulsar and changes in the magnetosphere are investigated to explain the profile evolution. However, the listed properties favour a model based on turbulence in the interstellar medium (ISM). This interpretation is confirmed by a strong correlation between the intensity of the transient component and main peak in single pulses. Since PSR B2217+47 is the fourth brightest pulsar visible to LOFAR, we speculate that ISM-induced pulse profile evolution might be relatively common but subtle and that SKA-Low will detect many similar examples. In this scenario, similar studies of pulse profile evolution could be used in parallel with scintillation arcs to characterize the properties of the ISM.

Fifty years of pulsar data has led to the discovery of emission and rotation variability on timescales of months and years; we have developed techniques to identify this long timescale variability. Individual observations may be too noisy to identify subtle changes in a pulse profile; we use Gaussian process regression to model noisy observations and produce a continuous map of pulse profile variability. Generally, multiple observing epochs are required to obtain the pulsar spin frequency derivative. Gaussian process regression is, therefore, also used to monitor this rate of spindown. We have applied variability detection techniques to both millisecond and long period pulsar datasets. I will discuss the techniques used and present the most interesting results from the pulsars analysed.

The aim of our project is to search for ways to best extract information on pulsar profiles and the interstellar medium (ISM), using the wide frequency bands that are typical of radio telescopes today. Pulsar profiles typically show a strong dependence on frequency. This depends both on the intrinsic radio emission mechanism, and the interaction of the radio waves with the ISM that lies between the pulsars and our detectors on Earth, due mostly to the effects of dispersion and scattering. In this work, we make use of radio pulsar beam models from the existing literature, to generate simulated pulse profiles, observed across various bands (centre frequencies and bandwidths), for each beam model. For all the chosen geometric parameters of the pulsar beam, observed in any frequency band, the simulated profiles manifest a relative shift in phase in their observed components, as a result of the intrinsic profile evolution. This relative shift in phase could be interpreted as an additional component to the ISM induced dispersion measure (DM). This additional DM component due to profile evolution is frequency dependent. We discuss the systematics introduced to pulsar data due to this effect.

The LOw Frequency ARray – LOFAR – is a new radio interferometer designed with emphasis on flexible digital hardware instead of mechanical solutions. The array elements, so-called stations, are located in the Netherlands and in neighbouring countries. The design of LOFAR allows independent use of its international stations, which, coupled with a dedicated backend, makes them very powerful telescopes in their own right. This backend is called the Advanced Radio Transient Event Monitor and Identification System (ARTEMIS). It is a combined software/hardware solution for both targeted observations and real-time searches for millisecond radio transients which uses Graphical Processing Unit (GPU) technology to remove interstellar dispersion and detect millisecond radio bursts from astronomical sources in real-time.

Recently, PSR J0738-4042 has grown a bright new emission component in its average pulse profile. Using data from Parkes and HartRAO, spanning back to the early 1980s, and applying statistical techniques to model the pulse profile shape with time, we have uncovered unexpected long-term variability, which is very well correlated with changes in the spin-down rate. We present these findings in the context of a growing population of radio-variable pulsars with correlated timing irregularities, including the intermittent pulsars, state-changing pulsars and other individual examples.

We present high quality polarimetric observations with the Effelsberg radio telescope of the Crab pulsar at 8.35 GHz using a new sensitive receiver with a very large bandwidth. We present our observations and discuss them in the context of the conclusions reached by Moffett & Hankins (1999) about the geometry and the origin of the various pulse components.

We have obtained simultaneous observations of single pulses from the bright pulsars B0329+54 and B1133+16 at two frequencies in full polarization and a total of four frequencies in total power only. Here is a summary of our fantastic results!
