Common bean (Phaseolus vulgaris L.) is an important crop of family Fabaceae used as a potential source of proteins, fibres and minerals. Thus, characterization of existing germplasm is useful for improvement and conservation. The Indian Himalayan Region harbours plentiful varieties of common bean, but it is nearly unexplored till date. In the present study, physical and genetic diversity of common bean was examined. Fifteen newly designed chloroplast microsatellite (cpSSR) markers were used to assess genetic diversity and population structure in 119 common bean individuals from 20 diverse accessions gathered from Uttarakhand, India. Significantly, positive (p< 0.05) relationship of seed weight was found with seed length (r = 0.813), seed width (r = 0.692) and seed length- width ratio (r = 0.694) using Pearson correlation analysis. A total of 20 alleles were identified using eight cpSSR markers. Mean number of alleles per locus (Na = 1.55), effective allele number (Ne = 1.370), expected heterozygosity (He = 0.213), average polymorphic loci (10.9) and Shannon information index (I = 0.313) were estimated based on cpSSR data. Maximum genetic diversity (He) was recorded in the AKJ/KK/DP/Jhalla/23 accession and minimum in the AKJ/YB/PS/Supi/43 accession. Bayesian-based STRUCTURE evaluation using cpSSR-based information partitioned 20 accessions into two distinct clusters which were also supported by neighbor-joining cluster analysis. These cpSSR markers also demonstrated transferability among other members like Vigna radiata, Macrotyloma uniflorum, Glycine max, Vigna mungo of Fabaceae family, therefore can be used to monitor their genetic heterogeneity. The findings from the study might be valuable to identify elite common bean accessions for production, conservation and future breeding programmes.

Sorghum (Sorghum bicolor (L.) Moench) is a dual nature crop, which is used for food as well as fodder, depending on plant ideotype. Sorghum forage is important for ruminants, but a major constraint is the anti-nutritional factor dhurrin, a hydrocyanic acid (HCN) producing glucoside. There are several additional effects of dhurrin, which reduce the nutritional value of sorghum fodder for livestock. This two-year study was aimed to evaluate the variation among diverse sorghum varieties, specifically for HCN content, forage yield and stem sugar content (brix value) under arid climate in Pakistan. Nine sorghum varieties were used for this experiment: JS-2002, Chakwal sorghum, Lines CS-17, Super late, PAK SS-2, Johar, JS-263, Sargodha-2011 and YSS-98. Results reveal that Sargodha-2011 had superior morphological traits for fresh forage and dry biomass yield, and stem brix value, compared to other varieties. Higher HCN contents were recorded in Super late compared to other varieties. Significant negative correlation of HCN with yield showed that improvement in yield will reduce the HCN content of sorghum. In conclusion, sorghum variety SGD-11 was shown best performing for higher biomass yield and brix value, and lower HCN content compared to other tested varieties under arid climate of Thal, Pakistan.

Pea landraces may hold genetic variation that can be exploitable in breeding/selecting new cultivars. In a small-seeded pea landrace, four phenotypes were distinguished according to testa (green, non-spotted and green, spotted) and hilum colour (white, black). The four phenotypes were tested for two growing seasons in the field for pod (seeds/pod) and seed traits (1000-seed weight, toughness, total phenols and tannins, testa colour, protein and carbon concentration, C/N ratio, bruchid infection). Significant differences were found for testa colour parameters, phenolic, tannin and carbon concentration and bruchid tolerance. The larger-seeded, spotted peas had darker testa and more phenols, while white-hilumed peas had lighter testa and more tannins. The spotted, black-hilumed phenotype, with the highest carbon concentration and C/N ratio was the most tolerant to bruchids. However, grouping the phenotypes, neither spotted nor black-hilumed ones showed to be more tolerant compared with their counterparts. Concluding, our results showed that phenotyping variation in seeds of a pea landrace revealed variation in seed traits, which could be exploitable. Since testa and hilum colour were associated with specific seed traits, they could, alone or in combination, be used as biomarkers of seed quality traits in pea. Testing of larger number of phenotypes is needed to solidify our findings.

The lack of excellent wheat germplasm resources on the Qinghai-Tibet Plateau has led to a gradual decrease in genetic diversity and an increasingly narrow genetic background in wheat grown in this region. Rational use of excellent genes from wheat relatives is important to increase genetic diversity, broaden the genetic base and achieve high yield and quality in common wheat. The objective of this study was to use principal component and cluster analyses of 13 important agronomic traits of 44 Polish wheat varieties over 3 years and comprehensively evaluate them to screen for excellent germplasm resources, thus providing the basic material for broadening the genetic base of Qinghai-Tibet Plateau wheat germplasm resources.

The evolutionary relationship between domesticated Oryza species and their wild relatives in North East India is not well understood. To improve the understanding of the evolutionary relationship, this study investigates the genetic diversity of 68 indigenous rice landraces from North East India, ten O. rufipogon genotypes, and nine O. nivara genotypes using chloroplast variable repeat markers which, due to non-recombination and uniparental inheritance, enable better phylogenetic inference. Reference genotypes IR64 (indica) and Nipponbare (japonica) were included to characterize various phylogenetic clusters. Using distance-based hierarchical clustering, model-based structuring and principal component analysis, selected landraces from the three North Eastern Indian states of Assam, Manipur and Arunachal Pradesh were grouped into two phylogenetically different clusters that represented the IR64 and Nipponbare groupings. Interestingly, despite the fact that a cluster analysis combining North East landraces and wild relatives likewise produced two separate clusters (cluster I: Nipponbare, cluster II: IR64), the majority of the wild relatives were only clustered in the IR64 cluster. This suggests that the two distinct evolutionary histories of the rice landraces in North East India are distinguished by their genetic affinity for wild relatives and their variation in the indica and japonica pools. These results highlight chloroplast divergence influencing the genetic diversity of North East landraces with wild relatives. Further, these findings will enable in-depth studies on the functional significance of chloroplast diversity on trait adaptation in rice landraces.

Ilex paraguariensis A. St.-Hil. (yerba mate) (Aquifoliaceae Bercht. & J. Presl) is a plant species with great economic and cultural importance because its leaves are processed and ground to make infusions like mate or tereré. The species is distributed in a continuous area that includes Southern Brazil and part of Paraguay and Argentina. Uruguay represents the Southern distribution limit of the species, where small populations can be found as part of ravine forests. Although there are previous reports of molecular markers for this and other species in the genus, the available markers were not informative enough to represent the intra- and interpopulation genetic diversity in marginal Uruguayan populations. In this study, we developed highly informative polymorphic microsatellite markers to be used in genetic studies in I. paraguariensis. Markers were identified in contigs from the genome sequence of two individuals and then tested for amplification and polymorphism content in a diverse panel. Markers which passed these filters were tested on populations from Uruguay. They detected higher diversity within populations (in terms of number of alleles and heterozygosity) than previously reported, and levels of heterozygosity similar to those reported for two Brazilian populations. This subset of seven markers were successfully multiplexed, substantially reducing the costs of the analysis. Combined with previously reported nuclear and plastid markers, they can be used to evaluate the genetic diversity of rear-edge populations, identify genotypes for paternity studies and provide relevant information for the conservation and management of germplasm.

In common with other grain legumes, the growth habit in horse gram is one of the domestication-driven adaptive traits. Horse gram exhibits determinate and indeterminate types of growth habits. Determinate genotypes have non-overlapping vegetative and reproductive phases combined with a compact growth habit which enables farmers to produce horse gram in intercropping and multiple cropping systems, a common practice for sustainable agriculture production. Also, synchronous flowering, and pod development and maturity of determinate genotypes enable mechanical harvesting. Empirical studies have indicated a greater grain yield potential of indeterminate compared to determinate genotypes. However, we hypothesize that indeterminate and determinate genotypes do not differ for their grain yield potential if they are in isogenic genetic backgrounds. To test this hypothesis, we compared eight pairs of near isogenic (NI) indeterminate and determinate genotypes for their grain yield potential during two rainy seasons in one location and one rainy season in another location. The eight pairs of NI determinate and indeterminate genotypes differed significantly in each of the three test environments. Indeterminate genotypes produced a greater pod (harvestable) and grain (marketable) yields than their determinate counterparts only in a few genetic backgrounds. These results thus support our hypothesis that determinate genotypes are comparable to indeterminate ones, if not better than the latter.

The potential of Momordica charantia var. muricata, a little-known wild form, as a source of favourable alleles in breeding and improvement of bitter melons (M. charantia var. charantia), is demonstrated. The M. charantia var. charantia cultivar Priyanka and M. charantia var. muricata accession IC634896 were crossed with each other and cross ability and performance of F1 plants were studied. These botanical varieties were fully cross-compatible on either direction. Surprisingly, cross of Priyanka × IC634896 produced 80.0 fruits compared to 17.0 in Priyanka and fruit yield per plant (2287.4 g) was more than double that in Priyanka (1042.3 g). This hybrid has to be back crossed with commercial cultivar to improve its fruit size, with no compromise on fruit number. This is the first study showing the promise of M. charantia var. muricata as a potential donor in bitter melon breeding.
